Summary Tolbutamide activates the oxidation of hexobarbital catalyzed by microsomal enzymes in the liver of rats, rabbits, and dogs. Consequently the duration of hexobarbital-action falls down to one fifth of the normal value. Sulfaethidole and sulfanilamide did not produce this enzymestimulation tested on rats, whereas carbutamide acts like tolbutamide; but the effect is weaker. Tolbutamide activates its own breakdown after a single dose as well as repeated administrations only in dogs. It seems to be metabolized by N-desalkylation with microsomal enzymes in the dog-liver. Other species, such as rats and rabbits, use different enzymes for the degradation of tolbutamide, which cannot be activated. After the stimulation of microsomal enzymes by tolbutamid morphological changes appear in the liver-cells, which are very similar to those observed after induction of microsomal enzymes after treatment with phenobarbital. Like phenobarbital tolbutamide seems to induce those enzymes.

Zusammenfassung Tolbutamid stimuliert die Oxydation von Hexobarbital mit Hilfe mikrosomaler Leberenzyme bei Ratte, Kaninchen und Hund, wodurch die Wirkungsdauer bis auf ein Fünftel der Norm absinkt. Sulfaethidol und Sulfanilamid besitzen diese nur an der Ratte getestete Wirkung nicht, während Carbutamid einen schwächeren Effekt als Tolbutamid ausübt. Nur beim Hunde, nicht aber beim Kaninchen, vermag es auch seine eigene Umwandlung sowohl bei wiederholter Gabe als auch nach einmaliger Applikation zu beschleunigen, was für eine mikrosomale N-Desalkylierung des Tolbutamids in der Hundeleber spricht. Andere Tierarten, wie Ratte und Kaninchen, benutzen einen anderen Abbauweg, der nicht auf mikrosomale Enzyme angewiesen ist und deshalb auch nicht stimuliert werden kann. Bei der Aktivierung mikrosomaler Enzyme durch Tolbutamid treten in der Leber nur elektronenoptisch sichtbare Veränderungen auf, die denjenigen entsprechen, die bei der Induktion mikrosomaler Enzyme durch Behandlung mit Phenobarbital beobachtet werden.
