    Abstract: Objectives. To explore the effective and safe medicines for treating diabetes. Methods. Hydroalcoholic extracts of 130 medicinal plants belonging to 66 families were evaluated using porcine pancreatic lipase (PPL) inhibition and glucose uptake methods together with a literature review. Results. The extracts of 22 species showed the PPL inhibition activity; 18 extracts of 15 species stimulated glucose uptake in 3T3-L1 adipocytes. Among them, Mansonia gagei J.R. Drumm., Mesua ferrea L., and Centella asiatica (L.) Urb. exhibited both activities. The extracts of Caladium lindenii (André) Madison rhizomes and Azadirachta indica A. Juss. leaves presented the utmost lipase inhibitory activity with IC50 of 6.86 ± 0.25 and 11.46 ± 0.06 μg/mL, respectively. The extracts of Coptis teeta Wall. rhizomes and Croton tiglium L. seeds stimulated the maximum glucose uptake. Ten species are reported to have antidiabetic activity for the first time. Flavonoids and triterpenoids are the dominant antidiabetic compounds in selected medicinal plants from Myanmar. Conclusions. P. zeylanica, L. cubeba, H. crenulate, M. gagei, C. teeta, and M. ferrea are worthy to advance further study according to their strong antidiabetic activities and limited research on effects in in vivo animal studies, unclear chemical constitutes, and safety.

    Abstract: A novel two-phase method for two-dimensional (2D) direction-of-arrival (DOA) estimation with L-shaped array based on decoupled atomic norm minimization (DANM) is proposed in this paper. In the first phase, given the sample crosscorrelation matrix, the gridless DANM technique considering the noise and finite snapshots effects is employed to exploit the structure and sparse properties of the crosscorrelation matrix. The resulting DANM-based algorithm not only enables the crosscorrelation matrix reconstruction (CCMR) but also reconstructs the covariance matrix of the L-shaped array. Hence, sequentially, in the second phase, the conventional 2D DOA estimators for the L-shaped array can be adopted for the angle estimation. With appropriate 2D DOA estimators, the resulting proposed algorithms can not only achieve better performance but also detect more source number, compared with conventional crosscorrelation-based DOA estimators. Moreover, the proposed method, termed CCMR-DANM, not only has blind characteristic that it does not require the prior information of source numbers but also is more efficient than the existing CCMR-based counterparts. Numerical simulations demonstrate the effectiveness and outperformance of the proposed method.

    Abstract: Background. In the past two decades, the surge of research on bacterial persisters has been inspired as increasingly concerning about the frequent failure of antibiotics treatment. This study was aimed at presenting a bibliometric and visualized analysis of relative publications on bacterial persisters, which offered insights into the development and research trends of this field. Methods. The Web of Science Core Collection and Ovid MEDLINE databases were utilized to retrieve relevant publications on bacterial persisters from 2001 to 2021. After manual selection, data including titles, authors, journals, author keywords, addresses, the number of citations, and publication years were subsequently extracted. The data analysis and visual mapping were conducted with Excel, SPSS, R studio, and VOSviewer. Results. In this study, 1,903 relevant publications on bacterial persisters were included. During 2001-2021, there was an exponential growth in the quantity of publications. It was found that these studies were conducted by 7,182 authors from 74 different countries. The USA led the scientific production with the highest total number of publications (859) and citation frequency (52,022). The Antimicrobial Agents and Chemotherapy was the most influential journal with 113 relevant publications. The cooccurrence analysis revealed that studies on bacterial persisters focused on four aspects: “the role of persisters in biofilms,” “clinical persistent infection,” “anti-persister treatment,” and “mechanism of persister formation.” Conclusion. In the past two decades, the global field of bacterial persisters has significantly increased. The USA was the leading country in this field. Mechanistic studies continued to be the future hotspots, which may be helpful to adopt new strategies against persisters and solve the problem of chronic infection in the clinic.

    Abstract: Objectives. The clinical and imaging features of asymptomatic carriers of severe acute respiratory syndrome coronavirus 2 and symptomatic COVID-19 patients. Methods. The clinical and chest computed tomography imaging data of 47 asymptomatic carriers and 36 symptomatic COVID-19 patients were derived. All patients underwent 4–6 CT scans over a period of 2–5 days. Results. The bulk of asymptomatic carriers who developed symptoms and most of the COVID-19 patients were older than 18 years of age with a decreased lymphocyte count, abnormal hepatic and renal function, and increased D-dimer and C-reactive protein. In the early stage, the pulmonary lesion involved mostly 1–2 lobes at the peripheral area in asymptomatic carriers but more than three lobes at both the central and peripheral areas in COVID-19 patients. In the progression stage, the lesion of asymptomatic carriers extended from the peripheral to the central area, and no significant difference was found in the lesion range compared with the symptomatic control group. In early improvement stage, the lesion was rapidly absorbed, and lesions were located primarily at the peripheral area in asymptomatic carriers; contrastingly, lesions were primarily located at both the central and peripheral areas in symptomatic patients. Asymptomatic carriers reflected a significantly shorter duration from disease onset to peak progression stage compared with the symptomatic. Conclusions. Asymptomatic carriers are a potential source of transmission and may become symptomatic COVID-19 patients despite indicating less severe pulmonary damage, earlier improvement, and better prognosis. Early isolation and intervention can eliminate such carriers as potential sources of transmission and improve their prognosis.

    Abstract: Sepsis-associated encephalopathy (SAE) is linked to increased morbidity and mortality rates in patients with sepsis. Increased cytokine production and neuronal apoptosis are implicated in the pathogenesis of the SAE. Neuroinflammation plays a major role in sepsis-induced brain injury. Thioredoxin-interacting protein (TXNIP), an inhibitor of thioredoxin, is associated with oxidative stress and inflammation. However, whether the TXNIP is involved in the sepsis-induced brain injury and the underlying mechanism is yet to be elucidated. Therefore, the present study was aimed at elucidating the effects of TXNIP knockdown on sepsis-induced brain injury and cognitive decline in mice. Lipopolysaccharide (LPS) was injected intraperitoneally to induce sepsis brain injury in mice. The virus-carrying control or TXNIP shRNA was injected into the lateral ventricle of the brain 4 weeks before the LPS treatment. The histological changes in the hippocampal tissues, encephaledema, and cognitive function were detected, respectively. Also, the 7-day survival rate was recorded. Furthermore, the alterations in microglial activity, oxidative response, proinflammatory factors, apoptosis, protein levels (TXNIP and NLRP3 inflammasome), and apoptosis were examined in the hippocampal tissues. The results demonstrated that the TXNIP and NLRP3 inflammasome expression levels were increased at 6, 12, and 24 h post-LPS injection. TXNIP knockdown dramatically ameliorated the 7-day survival rate, cognitive decline, brain damage, neuronal apoptosis, and the brain water content, inhibited the activation of microglia, downregulated the NLRP3/caspase-1 signaling pathway, and reduced the oxidative stress and the neuroinflammatory cytokine levels at 24 h post-LPS injection. These results suggested a crucial effect of TXNIP knockdown on the mechanism of brain injury and cognitive decline in sepsis mice via suppressing oxidative stress and neuroinflammation. Thus, TXNIP might be a potential therapeutic target for SAE patients.

    Abstract: Background. Overexpression of miR-195 can make gastric cancer cells stay in G1/G2 phase. miR-195 has been shown to inhibit gastric cancer cell replication and accelerate cell death by targeting JAK2. However, the relationship between miR-195, JAK2, and gastric cancer is not clear. Objective. To observe the effect of mir-195 regulated by JAK2 on the growth, invasion, and death of gastric cancer cells. Methods. MGC803 and NCI gastric N87 cells were introduced into the negative control sequences of miR-195 and RNA, respectively. To detect the expression of miR-195 in cells, to detect the effect of miR-195 on mitosis and proliferation of tumor cells, to analyze the effect of miR-195 on cell invasion and metastasis, and to detect the regulation of miR-195 on JAK2 expression. Results. The level of miR-195 in miR-195-MIMICS group was significantly higher than that in miR-NC group. The cell survival rate of miR-195 mimic group was lower than that of miR-NC group ( P 〈 0.05 ). Compared with miR-NC group, the number of cells in G1 phase increased, the cells in G2 phase and S phase decreased, and the proportion of cells in G2 and S phase decreased in miR-195 mimic group. The scratch distance of miR-195 simulator group was larger than that of control group. The number of invasive cells in the miR-195 mimic group was significantly lower than that in the control group. The expression of JAK2 protein in miR-195 mimic group was lower than that in miR-NC group. There was a significant negative correlation between the expression level of miR-195 and JAK2 (rhabdomile 0.326 and record 0.00). There are continuous interaction fragments between JAK2 and miR-195. The luciferase activity of miR-195 mimic and wild type JAK2 sequence expression vector was significantly lower than that of wild type JAK2 sequence expression vector. Conclusion. miR-195 may inhibit the occurrence, metastasis, and invasion of gastric tumor by downregulating the expression of JAK2. miR-195/JAK2 may be a new molecular target for the treatment of gastrointestinal tumors.

    Abstract: As an accurate, safe, and effective noninvasive examination method, imaging examination has been widely used in the diagnosis and differential diagnosis of focal liver lesions. Enhanced ultrasonography (CEUS), enhanced CT (CECT), and enhanced magnetic resonance imaging (CEMRI) are the most commonly used enhanced imaging methods in clinical practice, all of which can accurately determine the nature of liver lesions. The purpose of this paper is to study the application of contrast-enhanced ultrasound and magnetic resonance enhancement in cancer diagnosis based on the Internet of Things medical system. The basic clinical data, CEUS, and enhanced CT/MRI findings of 120 CHC patients were retrospectively analyzed. The clinicopathological features of CHC patients were investigated by contrast-enhanced ultrasonography and CT/MRI enhanced mode. The diagnostic value of contrast-enhanced ultrasound and enhanced CT/MRI combined with tumor markers in CHC was analyzed. The experimental results showed that the sensitivities of CEUS, enhanced MRI, and their combination in diagnosing CHC were 72.44%, 81.56%, and 93.78%, respectively. This experiment has an important value in the diagnosis of primary liver cancer.

    Abstract: A special design is needed for an unmanned tracked vehicle (UTV) to meet the requirements of off-road environments and complex tasks. A loose surface is the main terrain for tracked vehicles in off-road driving. Slope steering is inevitable while driving in such conditions; hence, its performance is a major concern for tracked vehicles on loose terrain. This study investigates the slope steering performance of a tracked vehicle. An improved dynamic steering model is proposed when considering the shear stress-shear displacement relation of soil at the track-ground interface. The influence of ground characteristics on the slope steering performance of a tracked vehicle is illustrated. The track slip rate is adopted as an index to evaluate the influence of typical vehicle structure parameters on the slope steering performance of a tracked vehicle. This study provides technical support for the design and optimization of UTV.

    Abstract: In view of the geological environment, in which the upper part is the “soft” layer of completely weathered siliceous rock and the lower part is the “hard” layer of weakly weathered limestone, the rail surface line is designed to pass through the interface between the upper “soft” layer and lower “hard” layer. First, the MIDAS/GTS software was used to comprehensively simulate the stress and deformation law of surrounded rock under three excavation methods, which are the positive benching and retaining core soil method, CRD method, and double side drift method, for selecting the appropriate excavation method. After the excavation method was determined, the rule of change in the stress and deformation of the surrounded rock with time in the actual excavation process was analyzed by field monitoring means. Finally, on the basis of the findings in the numerical analysis as well as the field monitoring, the engineering characteristics, such as the stress and deformation of the surrounded rock during the excavation of deep buried tunnels in the upper-soft lower-hard ground, were analyzed. The results revealed the following: (1) Under all three excavation methods, the tunnel deformation was small and met the requirements for tunnel deformation control. (2) Due to the different excavation support sequence, the distribution of lining bending moment of the three excavation methods was widely different. The bending moment of the lining produced by the positive benching method was far less than that of the other two methods. (3) Due to the difference in lithology, the stress of the tunnel lining in the thick upper “soft” and “hard” strata was mainly concentrated on the upper soft rock area, while the stress in the lower hard rock area was relatively small, and the lining stress value generated using the three excavation methods was relatively large. In general, the deformation and stress in the positive benching method construction were less than those in the other two methods. In addition, the positive benching method was convenient for mechanized operation, the construction progress was fast, and the cost was relatively low. Therefore, the positive benching and retaining core soil method is adoptable for this kind of tunnel. (4) The measured stress and deformation at the rail surface line exhibited a change law of first increase, and subsequently, this tended to be stable. The change in stress-time can be expressed by the Boltzmann function, and the change in deformation with time can be well expressed by exponential functions. (5) The stress value and deformation value detected in the actual excavation were greater than the results of the theoretical numerical analysis. However, the findings in the theoretical numerical analysis still have certain guiding significance for actual excavation.
