    Abstract: Gallbladder cancer (GBC), the sixth most common gastrointestinal tract cancer, poses a significant disease burden in China. However, no national representative data are available on the clinical characteristics, treatment and prognosis of GBC in the Chinese population. Methods and analysis The Chinese Research Group of Gallbladder Cancer (CRGGC) study is a multicentre retrospective registry cohort study. Clinically diagnosed patient with GBC will be identified from 1 January 2008 to December, 2019, by reviewing the electronic medical records from 76 tertiary and secondary hospitals across 28 provinces in China. Patients with pathological and radiological diagnoses of malignancy, including cancer in situ, from the gallbladder and cystic duct are eligible, according to the National Comprehensive Cancer Network 2019 guidelines. Patients will be excluded if GBC is the secondary diagnosis in the discharge summary. The demographic characteristics, medical history, physical examination results, surgery information, pathological data, laboratory examination results and radiology reports will be collected in a standardised case report form. By May 2021, approximately 6000 patient with GBC will be included. The clinical follow-up data will be updated until 5 years after the last admission for GBC of each patient. The study aimed (1) to depict the clinical characteristics, including demographics, pathology, treatment and prognosis of patient with GBC in China; (2) to evaluate the adherence to clinical guidelines of GBC and (3) to improve clinical practice for diagnosing and treating GBC and provide references for policy-makers. Ethics and dissemination The protocol of the CRGGC has been approved by the Committee for Ethics of Xinhua Hospital, Shanghai Jiao Tong University School of Medicine (SHEC-C-2019–085). All results of this study will be published in peer-reviewed journals and presented at relevant conferences. Trial registration number NCT04140552 , Pre-results.

    Abstract: High plasma 3‐carboxy‐4‐methyl‐5‐propyl‐2‐furanpropanoic acid ( CMPF ) levels are significantly associated with type 2 diabetes mellitus, which is usually accompanied by metabolic syndrome and non‐alcoholic fatty liver disease ( NAFLD ) with increased triglyceride levels. Thus, we hypothesized that elevated CMPF levels might be related to lipid metabolism and NAFLD risk. Materials and Methods Serum CMPF levels were determined using an enzyme‐linked immunosorbent assay in a total of 466 individuals, including 116 controls with no NAFLD or type 2 diabetes mellitus, 53 individuals with NAFLD but no type 2 diabetes mellitus, 151 individuals with type 2 diabetes mellitus but no NAFLD, and 146 individuals with both NAFLD and type 2 diabetes mellitus. The associations with age, blood pressure, lipid profiles, body mass index and liver injury marker levels were examined, and a meta‐analysis of non‐diabetic and diabetic groups was carried out to detect the combined effects. Results The CMPF concentration in NAFLD patients was significantly lower than individuals without NAFLD in both the non‐diabetic group ( P  〈  0.05) and diabetic group ( P  〈  0.01), and correlated negatively with several parameters of liver function and the adiposity index. Meta‐analysis showed that serum CMPF levels was associated with decreased risk of NAFLD after combining the results (odds ratio 0.677, 95% confidence interval 0.552–0.831, P  〈  0.001). Additionally, the CMPF concentration was independently negatively associated with triglycerides and high‐density lipoprotein cholesterol in the meta‐analysis. Multiple stepwise regression analysis showed that body mass index , high‐density lipoprotein cholesterol, triglyceride level, age, sex and fasting plasma glucose were independently associated with CMPF (all P   〈  0.05). Conclusions The results suggest that serum CMPF levels are negatively related to lipid metabolism and could be used to predict NAFLD development.

    Abstract: Striking networks of livestock tracks, or terracettes, molded to the contours are a common feature on hilly rangelands of the semiarid Loess Plateau, one of the regions with the most severe soil erosion in the world. The formation of livestock tracks results in modified micro‐topography and ecological processes and leads to strong patterns in spatial distribution of vegetation, water, and nutrients. We investigated the spatial pattern of the topo‐edaphic, hydrological, biogeochemical, and biological attributes of these terracette landscapes and explored their potential implications to ecosystem functions. The results showed strong spatial heterogeneity in topo‐edaphic and biological structure of these landscapes, organized with three segments of a basic landscape unit—track, shoulder, and interslope. The above‐ and below‐ground biomass and soil nutrients concentrated in the shoulder segment forming a “band of fertility”. The network of livestock tracks likely reduced surface runoff and erosion by intercepting runoff and facilitate infiltration, while posing little risk of increasing shallow slope failures given the structure of loess soil. There appears a positive feedback loop for maintaining the structure of terracette landscapes. Trampling by goats maintains the tracks, which leads to spatial heterogeneity in biophysical structure and processes. The network of tracks enabled the goats to travel and graze in ways that reduce energy expenditure and increase foraging efficiency, which leads to strongly preferential use of the tracks by the goats. There are also evidence of a possible process resemble a slope parallel retreat at a micro‐scale, which may result in upslope movement of the terracettes. There may be important functional differences between landscapes with and without networks of livestock tracks, in regulating water runoff and soil erosion, forage production, and soil carbon storage. These functional differences may have significant implications to land use policies and practices aiming at soil and water conservation and socioecological sustainability of the Loess Plateau.

    Abstract: Several recent clinical studies have suggested that the levels of circulating 3-carboxy-4-methyl-5-propyl-2-furanpropanoic acid (CMPF) are significantly higher in patients with gestational diabetes mellitus (GDM), impaired glucose tolerance (IGT), and type 2 diabetes mellitus (T2DM). This study recruited a total of 516 participants. The following patient populations were enrolled: 99 newly diagnosed cases with T2DM, 219 cases with prediabetes [82 with isolated impaired glucose tolerance (I − IGT), 66 with isolated impaired fasting glucose (I − IFG) and 71 with impaired glucose tolerance and impaired fasting glucose (IGT + IFG)], and 198 cases with normal glucose tolerance [NGT, including 99 first-degree relatives of type 2 diabetes patients (FDRs) and 99 non-FDRs] . We investigated the circulating CMPF levels in subjects with different glucose metabolism statuses and examined the potential link between CMPF and β cell function. Our results indicate that the serum CMPF levels were elevated in the prediabetes, T2DM, and FDRs groups compared to the NGT group. Additionally, the serum CMPF concentrations were independently and negatively associated with the triglyceride levels and Stumvoll first-phase insulin secretion index. Cumulatively, our findings suggest that the circulating CMPF levels can predict glycolipid metabolism disorders. Furthermore, elevated serum CMPF concentrations may determine hyperglycemia and β cell dysfunction.

    Abstract: Quantitatively estimating the grazing intensity (GI) effects on vegetation in semiarid hilly grassland of the Loess Plateau can help to develop safe utilization levels for natural grasslands, which is a necessity of maintaining livestock production and sustainable development of grasslands. Normalized difference vegetation index (NDVI), field vegetation data, and 181 days (one goat per day) of GPS tracking were combined to quantify the spatial pattern of GI, and its effects on the vegetation community structure. The spatial distribution of GI was uneven, with a mean value of 0.50 goats/ha, and 95% of the study area had less than 1.30 goats/ha. The areas with utilization rates of rangeland (July) lower than 45% and 20% made up about 95% and 60% of the study area, respectively. Grazing significantly reduced monthly aboveground biomass, but the grazing effects on plant growth rate were complex across the different plant growth stages. Grazing impaired plant growth in general, but the intermediate GI appeared to facilitate plant growth rate at the end of the growing seasons. Grazing had minimal relationship with vegetation community structure characteristics, though Importance Value of forbs increased with increasing GI. Flexibility in the number of goats and conservatively defining utilization rate, according to the inter-annual variation of utilization biomass, would be beneficial to achieve ecologically healthy and economically sustainable GI.

    Abstract: Currently, many researches have developed several strategies to design the surface structures of hydroxyapatite (HA), and have proved that the surface structures are pivotal in guiding the adhesion of bone marrow mesenchymal stem cells (BMSCs) as well as subsequent cellular behaviours. Most of these strategies, such as altering roughness and constructing surface patterning of HA, involve the construction of geometric topographies at the micro/nanoscale. However, besides geometric topographies, crystal defects are also important characteristics of surface structures and would alter many local physicochemical properties, which is critical for contact between cells and bioceramic surfaces. For the practical applications of crystal defects, a major hindrance is that crystal defects are usually unstable and easily eliminated during crystallization, which limits the large-scale fabrication of materials with crystal defects. In this work, given that stepped structures contain massive stable crystal defects on their step edges and kinks, we proposed a feasible and efficient method to fabricate HA dishes with stepped structures on their surfaces. First, plate-like HA mesocrystals were prepared from CaHPO 4 via topotactic transformation, and were shaped into HA dishes by vacuum-filtration. Then, a sintering process was applied to facilitate the formation of stepped structures on the surfaces. We demonstrated that the generation of stepped structures could restrict the adhesion of BMSCs and showed the restriction effect is highly correlated with the density of exposed stepped structures. This phenomenon is interesting and the construction of a cell adhesion model is robust and easy, the underlying mechanisms of which deserve further exploration. Furthermore, constructing stepped structures on surfaces may be a new useful strategy to regulate cell adhesion and could also cooperate with other methods that do not need change in the surface crystal structure.
