    Abstract: Nanozymes own striking merits, including high enzyme-mimicking activity, good stability, and low cost. Due to the powerful and distinguished functions, nanozymes exhibit widespread applications in the field of biosensing and immunoassay, attracting researchers in various fields to design and engineer nanozymes. Recently, nanozymes have been innovatively used to bridge nanotechnology with analytical techniques to achieve the high sensitivity, specificity, and reproducibility. However, the applications of nanozymes in food applications are seldom reviewed. In this review, we summarize several typical nanozymes and provide a comprehensive description of the history, principles, designs, and applications of nanozyme-based analytical techniques in food contaminants detection. Based on engineering and modification of nanozymes, the food contaminants are classified and then discussed in detail via discriminating the roles of nanozymes in various analytical methods, including fluorescence, colorimetric and electrochemical assay, surface-enhanced Raman scattering, magnetic relaxing sensing, and electrochemiluminescence. Further, representative examples of nanozymes-based methods are highlighted for contaminants analysis and inhibition. Finally, the current challenges and prospects of nanozymes are discussed.

    Abstract: Sepsis-associated thrombocytopenia (SAT) is a common complication in the intensive care unit (ICU), which significantly increases the mortality rate and leads to poor prognosis of diseases. Machine learning (ML) is widely used in disease prediction in critically ill patients. Here, we aimed to establish prediction models for platelet decrease and severe platelet decrease in ICU patients with sepsis based on four common ML algorithms and identify the best prediction model. The research subjects were 1,455 ICU sepsis patients admitted to Dongyang People's Hospital affiliated with Wenzhou Medical University from January 1, 2015, to October 31, 2019. Basic clinical demographic information, biochemical indicators, and clinical outcomes were recorded. The prediction models were based on four ML algorithms: random forest, neural network, gradient boosting machine, and Bayesian algorithms. Thrombocytopenia was found to occur in 732 patients (49.7%). The mechanical ventilation time and length of ICU stay were longer, and the mortality rate was higher for the thrombocytopenia group than for the non-thrombocytopenia group. The models were validated on an online international database (Medical Information Mart for Intensive Care III). The areas under the receiver operating characteristic curves (AUCs) of the four models for the prediction of thrombocytopenia were between 0.54 and 0.72. The AUCs of the models for the prediction of severe thrombocytopenia were between 0.70 and 0.77. The neural network and gradient boosting machine models effectively predicted the occurrence of SAT, and the Bayesian models had the best performance in predicting severe thrombocytopenia. Therefore, these models can be used to identify such high-risk patients at an early stage and guide individualized clinical treatment, to improve the prognosis of diseases.

    Abstract: As one of the important submesoscale instabilities, symmetric instability (SI) widely exists in the ocean surface mixed layer (SML), which enhances the vertical material transport in the SML and also the exchanges between the SML and the ocean interior. Due to the small spatial scales of SI, O (10 m–1 km), which are not resolved by most current ocean models, the application of SI parameterization is an alternative choice in the coming decades to include the SI effects in ocean models and improve the model performance. In this study, we evaluate the impacts of SI in a realistic configuration with the SI parameterization scheme applied in the Xisha-Zhongsha waters, South China Sea in winter by using the Coastal and Regional Ocean Community Model (CROCO) version of the Regional Ocean Modeling System. Compared to the SI-lacking case, the SI energy source, the geostrophic shear production, is increased and elimination of anticyclonic potential vorticity is revealed in the SI-parameterized case. According to the energy analysis, multi-scale interactions are also influenced by the SI. The effective wind energy input is reduced, and the potential energy release in the SML is suppressed. Moreover, the SI scheme makes the SML depth shallower and closer to the reanalysis one. This work demonstrates a good performance of the SI scheme applied in regional models in representing SI effects.

    Abstract: Introduction: According to traditional Chinese veterinary medicine, endometritis is caused by a combination of Qi deficiency, blood stasis, and external evil invasion. Salvia miltiorrhiza is a traditional Chinese medicine that counteracts blood stasis and has additional demonstrated effects in boosting energy and restraining inflammation. Salvia miltiorrhiza has been employed in many traditional Chinese prescriptions that have proven effective in healing clinical dairy cow endometritis. Methods: the in vivo effect of Salvia miltiorrhiza in treating endometritis was evaluated in dairy cows. In addition, bovine endometrial epithelium cell inflammation and rat blood stasis models were employed to demonstrate the crosstalk between energy, blood circulation and inflammation. Network analysis, western blotting, qRT-PCR and ELISA were performed to investigate the molecular mechanism of Salvia miltiorrhiza in endometritis treatment. Results: The results demonstrate that treatment with Salvia miltiorrhiza relieves uterine inflammation, increases blood ATP concentrations, and prolongs blood clotting times. Four of the six Salvia miltiorrhiza main components (SMMCs) (tanshinone IIA, cryptotanshinone, salvianolic acid A and salvianolic acid B) were effective in reversing decreased ATP and increased IL-1β, IL-6, and IL-8 levels in an in vitro endometritis model, indicating their abilities to ameliorate the negative energy balance and external evil invasion effects of endometritis. Furthermore, in a blood stasis rat model, inflammatory responses were induced in the absence of external infection; and all six SMMCs inhibited thrombin-induced platelet aggregation. Network analysis of SMMC targets predicted that Salvia miltiorrhiza may mediate anti-inflammation via the Toll-like receptor signaling pathway; anti-aggregation via the Platelet activation pathway; and energy balance via the Thermogenesis and AMPK signaling pathways. Multiple molecular targets within these pathways were verified to be inhibited by SMMCs, including P38/ERK-AP1, a key molecular signal that may mediate the crosstalk between inflammation, energy deficiency and blood stasis. Conclusion: These results provide mechanistic understanding of the therapeutic effect of Salvia miltiorrhiza for endometritis achieved through Qi deficiency, blood stasis, and external evil invasion.

    Abstract: Six new phthalan derivatives cytorhizophins D-I (1-6) as well as three known derivatives cytorhizophin C, pestacin and rhizophol B were isolated from Cytospora rhizophorae. Among them, cytorhizophins D-E (1-2) and F-G (3-4) were two pairs of diastereoisomers, all of them featuring a 1-phenyl-1,3-dihydroisobenzofuran scaffold with a highly oxygenated O -linked isopentenyl unit. Besides, cytorhizophins H-I (5-6) represent the first examples of phthalide family with fascinating 6/6/6/5 tetracyclic ring system fusing as unprecedented furo [4,3,2- kl ]xanthen-2 (10b H )-one skeleton. The structures of the new phthalan derivatives were extensively confirmed by detail spectroscopic analysis. The partial absolute configurations of compounds 1-6 were established through electronic circular dichroism (ECD) calculations. Moreover, compounds 1-4 showed remarkable antioxidant activities with EC 50 values ranging from 5.86 to 26.80 μM, which were better than or comparable to that of ascorbic acid (positive control).

    Abstract: Satellite observations play important roles in ocean operational forecasting systems, however, the direct assimilation of satellite observations cannot provide sufficient constraints on the model underwater structure. This study adopted the indirect assimilation method. First, we created a 3D temperature and salinity reconstruction model that took into account the advantage of the nonlinear regression of the generalized regression neural network with the fruit fly optimization (abbreviated as FOAGRNN). Compared with the reanalysis product and the WOA13 climatology data, the synthetic T/S (temperature and salinity) profiles had sufficient accuracy and could better describe the characteristics of mesoscale eddies. Then, the synthetic T/S profiles were assimilated into the Regional Ocean Model System (ROMS) using the Incremental Strong constraint 4D Variational (I4D-Var) data assimilation algorithm. The quantitative and qualitative analysis results indicated that compared with the direct assimilation of satellite observations, the root mean square errors (RMSEs) of temperature and salinity were reduced by 26.0% and 23.1% respectively by assimilating the synthetic T/S profiles. Furthermore, this method significantly improved the simulation effect of the model underwater structure, especially in the 300 m to 500 m water layer. Compared with the National Marine Data Center’s real-time analysis data, the machine learning-based assimilation system demonstrated a significant advantage in the simulation of underwater salinity structure, while showing a similar performance in the simulation of underwater temperature structure.

    Abstract: The genomic context of the mcr-1 gene in Escherichia coli from animal feces has been widely reported. However, less is known about the mcr-1 -carrying plasmid characteristics and other functional regions of Escherichia coli isolates from animal organs with lesions. The present study investigated the antimicrobial resistance, population structure, and genetic features of mcr-1 -positive Escherichia coli strains isolated from animal organs with lesions. The antimicrobial susceptibility testing indicated that 24 mcr-1 -positive Escherichia coli isolates were resistant to at least three or all antimicrobial categories. MLST analysis suggested that the dominant clone complexes (CC) were mainly CC156, CC448, and CC10. In addition, ST10596, a newly discovered sequence type in swine, failed to be classified. Meanwhile, the mcr-1 gene located on the different plasmids was successfully transferred to the recipients, and whole-genome sequencing indicated the mcr-1 gene was embedded in mcr-1-pap2 cassette but not flanked by IS Apl1 . The mcr-1 gene is located on the chromosome and embedded in Tn 6330 . Furthermore, NDM-5 was found on the IncX3-type plasmid of J-8. The virB6 and traI gene of type IV secretion system (T4SS) were truncated by IS 2 and IS 100 and located on the IncX4- and the IncHI2/HI2A/N-type plasmids, respectively. The multidrug-resistant (MDR) region of IncHI2/HI2A/N-type plasmids contained two class 1 integrons (In 0 , In 640 ) and four composite transposons (Tn 4352 , Tn 6010 , cn_4692_IS 26 , cn_6354_IS 26 ). Overall, 24 mcr-1 -positive Escherichia coli isolates in our study showed MDR, or even extensively drug resistant (XDR), and exhibited population diversity. The T4SS gene truncation by the insertion sequence may affect the efficiency of plasmid conjugative transfer. Furthermore, the class 1 integrons and composite transposons in the MDR region of IncHI2/HI2A/n-type plasmid contributed to the multireplicon plasmid formation, the acquisition, and transfer of antimicrobial resistance genes (ARGs).

    Abstract: Responses of the South China Sea (SCS) to a typhoon are complex due to the susceptible upper layer and active multiscale motions and thus need to be urgently resolved and validated in numerical simulations. A coupled atmosphere–ocean–wave model and various in-situ observations were applied to understand the strong interactions between Super Typhoon Megi (2010) and the SCS, especially the wave effects on typhoon simulation. Five sensitive experiments using different combinations of models were firstly conducted and compared to validate the effectiveness of the ocean coupling. Compared with WRF-only and ROMS-only outputs, the coupled experiments evidently improved the accuracy of typhoon intensity, the typhoon-induced cold wake, and significant wave height, along with the thermodynamical responses in the upper 400 m layer, including the near-inertial currents, the variation in ocean heat content, and mixed layer depth. However, the differences between WRF-ROMS and COAWST were slight, though the significant wave height was more than 9 m high in COAWST. Further analysis showed that the modification of heat flux, which could cancel out the effect due to the wave-induced surface roughness, is consistent with that of momentum flux in the wave-coupled experiment. This resulted in similar overall results. To further figure out the wave effects on typhoon and eliminate the contingency brought by the surface physical parameterization scheme, six experiments using three surface physical parameterization schemes were designed with and without wave coupling, separately. The sensible heat flux showed significant differences between three schemes, followed by the latent heat flux and the correspondingly changing momentum loss. Results support the above-mentioned conclusion that the typhoon intensity was determined by the net surface flux. Our findings highlight the necessity in using a high-resolution coupled atmosphere–ocean–wave model and proper surface physical parameterization, especially when coupling waves to make accurate regional numerical environment predictions.
