    Abstract: Objective: Lung auscultation plays an important role in the diagnosis of pulmonary diseases in children. The objective of this study was to evaluate the use of an artificial intelligence (AI) algorithm for the detection of breath sounds in a real clinical environment among children with pulmonary diseases. Method: The auscultations of breath sounds were collected in the respiratory department of Shanghai Children's Medical Center (SCMC) by using an electronic stethoscope. The discrimination results for all chest locations with respect to a gold standard (GS) established by 2 experienced pediatric pulmonologists from SCMC and 6 general pediatricians were recorded. The accuracy, sensitivity, specificity, precision, and F1-score of the AI algorithm and general pediatricians with respect to the GS were evaluated. Meanwhile, the performance of the AI algorithm for different patient ages and recording locations was evaluated. Result: A total of 112 hospitalized children with pulmonary diseases were recruited for the study from May to December 2019. A total of 672 breath sounds were collected, and 627 (93.3%) breath sounds, including 159 crackles (23.1%), 264 wheeze (38.4%), and 264 normal breath sounds (38.4%), were fully analyzed by the AI algorithm. The accuracy of the detection of adventitious breath sounds by the AI algorithm and general pediatricians with respect to the GS were 77.7% and 59.9% ( p & lt; 0.001), respectively. The sensitivity, specificity, and F1-score in the detection of crackles and wheeze from the AI algorithm were higher than those from the general pediatricians (crackles 81.1 vs. 47.8%, 94.1 vs. 77.1%, and 80.9 vs. 42.74%, respectively; wheeze 86.4 vs. 82.2%, 83.0 vs. 72.1%, and 80.9 vs. 72.5%, respectively; p & lt; 0.001). Performance varied according to the age of the patient, with patients younger than 12 months yielding the highest accuracy (81.3%, p & lt; 0.001) among the age groups. Conclusion: In a real clinical environment, children's breath sounds were collected and transmitted remotely by an electronic stethoscope; these breath sounds could be recognized by both pediatricians and an AI algorithm. The ability of the AI algorithm to analyze adventitious breath sounds was better than that of the general pediatricians.

    Abstract: 〈 i 〉 Background: 〈 /i 〉 Bone marrow-derived cells may play a role in tissue injury and repair. Growth factors facilitate the mobilization of bone marrow-derived cells to the site of injury. 〈 i 〉 Objectives: 〈 /i 〉 The aim of this study was to determine the effect of the mobilization of autologous bone marrow-derived cells by granulocyte colony-stimulating factor (CSF3) on bleomycin-induced lung injury in mice. 〈 i 〉 Methods: 〈 /i 〉 The bone marrow from male green fluorescent protein transgenic (C57Bl/6J) mice was transplanted into irradiated female C57Bl/6J mice. Bleomycin lung injury was induced in these bone marrow-reconstituted mice and unreconstituted C57Bl/6J mice, and some mice were treated with recombinant CSF3. Lung histology, survival, cytokine expression and matrix metalloproteinase (MMP) expression were evaluated to determine the effect of CSF3 after bleomycin-induced lung injury. 〈 i 〉 Results: 〈 /i 〉 Histology and flow cytometry analysis showed successful mobilization of bone marrow-derived cells by CSF3 treatment in the recipient lungs. Importantly, CSF3 attenuated bleomycin-induced lung injury and improved survival. Furthermore, CSF3 administration regulated transforming growth factor-β, interferon-γ, MMP9 and tissue inhibitors of MMP1 expression during bleomycin injury. 〈 i 〉 Conclusions: 〈 /i 〉 These data demonstrated that the mobilization of bone marrow-derived cells by CSF3 has a protective effect against bleomycin-induced lung injury and fibrosis.

    Abstract: The carotenoid isomerase gene ( BoaCRTISO ) of Chinese kale was targeted and edited using the CRISPR/Cas9 system in the present study. The results showed a high mutation rate (81.25%), and 13 crtiso mutants were obtained. Only two types of mutations, insertions and replacements, were found. Both the total and individual carotenoid and chlorophyll concentrations of the biallelic and homozygous mutants were reduced, and the total levels declined by 11.89–36.33%. The color of the biallelic and homozygous mutants changed from green to yellow, likely reflecting a reduction in the color-masking effect of chlorophyll on carotenoids. The expression levels of most carotenoid and chlorophyll biosynthesis-related genes, including CRTISO , were notably lower in the mutants than in the WT plants. In addition, the functional differences between members of this gene family were discussed. In summary, these findings indicate that CRISPR/Cas9 is a promising technique for the quality improvement of Chinese kale and other Brassica vegetables.

    Abstract: Two 2D 4d‐4f heterometallic coordination polymers, [ Ln Ag(Py26DC) 2 (H 2 O) 3 ] · 3H 2 O [ Ln = Nd ( 1 ), La ( 2 ); H 2 Py26DC = pyridine‐2,6‐dicarboxylic acid], and one 2D lanthanide homometallic coordination polymer, [ Ln (Py25DC)(ox) 0.5 (H 2 O) 2 ] [ Ln = Tm ( 3 ); H 2 Py25DC = pyridine‐2,5‐dicarboxylic acid; ox = oxalate], were synthesized and characterized by elemental analysis, IR spectroscopy, thermogravimetric analysis, and single‐crystal X‐ray diffraction analysis. Both complexes 1 and 2 are isostructural and exhibit 3‐connected 2D heterometallic layer structures with the Schläfli symbol of (8 2 · 10), whereas complex 3 represents an extended 2D homometallic network structure with (4,4) topology.

    Abstract: Objective: The test for respiratory and asthma control in kids (TRACK) is currently the only standard follow-up tool for children under 5 years of age with asthma. The purpose of this study was to investigate the relationship between the TRACK initial score (Ti) and their prognosis after 6 months of follow-up in pre-schoolers with asthma. Design: A prospective cohort study. Methods: The study included pre-schoolers diagnosed with asthma at the Shanghai Children's Medical Center between January 2019 and June 2020, and follow-up for 6 months. TRACK scores, frequency of wheezing and respiratory infections, number of Emergency Department (ED) visits and treatment regimen were collected. According to the TRACK initial score, the children were divided into “Ti & lt; 60 group” and “Ti ≥ 60 group,” and the two groups were compared in terms of TRACK score related indicators, clinical manifestations and treatment. Results: There are 102 pre-schoolers included in the analysis [78 boys (76.5%) and 24 girls (23.5%); mean (SD) age, 28.05 (11.63) months]. After 6 months of follow-up, the TRACK score was improved in both groups, and the “Ti ≥ 60 group” had a higher score, lower rate of uncontrolled asthma and fewer reassessments were required. There was no difference in the number of wheezing attacks between the two groups in terms of clinical presentation, but the “Ti & lt; 60 group” had more respiratory infections and ED visits. Regarding the use of ICSs, in the “Ti & lt; 60 groups,” the dose of ICSs was higher and reduced slowly, and the dose difference between the two groups began to appear after 5 months of follow-up. Conclusion: TRACK is essential for pre-schoolers with asthma at the time they are diagnosed. In addition, if the TRACK initial score is & lt; 60, the probability of poor prognosis is higher.

    Abstract: The AOP2.3 gene is an important gene affecting the side chain modification of aliphatic glucosinolate. Here, the Brassica oleracea var. capitata AOP2.3 (BocAOP2.3) gene sequence was obtained from the Brassica database (BRAD) and subjected to sequence analysis. It is indicated that the BocAOP2.3 gene contains a 936 bp open reading frame (ORF) encoding a protein of 311 amino acids with a calculated molecular weight of 33.62 kD, the isoelectric point (pI) is 4.46. The highest number of amino acids is Valine (Val), whereas the lowest number is Tryptophan (Trp). Its prediction formula is C 1455 H 2295 N 397 O 505 S 6 . Its total hydrophobicity index is -0.5, the solution instability index is 29.25, and the fat solubility index is 77.46. Sequence alignment indicated that the BocAOP2.3 protein is more closely related to Brassica napus and Brassica Rapa , those all belonged to the Cruciferous genus. The results of this study provide a molecular basis for elucidating the function of BocAOP2.3 gene in cabbage.

    Abstract: The experiment used the ‘Sijicutiao’ mustard sterile seedlings as the material, firstly constructed the BoaZDS subcellular localization vector with the cloned BoaZDS gene, and extracted the plasmid, then separated the Chinese kale leaf protoplast by enzymatic hydrolysis. After purification, the BoaZDS gene was transiently transformed, and then the position of the BoaZDS gene was observed under a fluorescence microscope. The results showed that the BoaZDS gene was localized in the chloroplast, which was consistent with the predicted results of bioinformatics software. The results provide a basis for studying the function of the ZDS gene in Chinese kale.

    Abstract: Carotenoids are organic pigments that play an important role in both plant coloration and human health; they are a critical subject in molecular breeding due to growing demand for natural molecules in both food and medicine. In this study, we focus upon characterizing BoaCRTISO , the carotenoid isomerase gene before the branch of the carotenoid biosynthetic pathway, which is expressed in all organs and developmental stages of Chinese kale, and BoaCRTISO, which is located in the chloroplast. The expression of BoaCRTISO is induced by strong light, red and blue combined light, and gibberellic acid treatment, but it is suppressed by darkness and abscisic acid treatment. We obtained BoaCRTISO -silenced plants via virus-induced gene silencing technology, and the silence efficiencies ranged from 52 to 77%. The expressions of most carotenoid and chlorophyll biosynthetic genes in BoaCRTISO -silenced plants were downregulated, and the contents of carotenoids and chlorophyll were reduced. Meanwhile, BoaCRTISO -silenced plants exhibited phenotypes of yellowing leaves and inhibited growth. This functional characterization of BoaCRTISO provides insight for the biosynthesis and regulation of carotenoid in Chinese kale.
