    Abstract: Addition of deuterium oxide (D 〈 sub 〉 2 〈 /sub 〉 O), 6–36%, resulted in a dose-dependent increase in allergen- or anti-IgE-induced leukotriene C 〈 sub 〉 4 〈 /sub 〉 (LTC 〈 sub 〉 4 〈 /sub 〉 ) generation from human basophils. In the presence of 36% D 〈 sub 〉 2 〈 /sub 〉 O, the enhancement was 260 ± 135% for allergen stimulation and 480 ± 152% for anti-IgE stimulation as compared with the control incubated in normal buffer. The increasing effect of D 〈 sub 〉 2 〈 /sub 〉 O on LTC 〈 sub 〉 4 〈 /sub 〉 generation from basophils was completely reversed by washing the cells before incubation with allergen. Vinblastine as well as colchicine, at a concentration of 100 μ 〈 i 〉 M 〈 /i 〉 , counteracted the effect of D 〈 sub 〉 2 〈 /sub 〉 O. The enhanced release of histamine and LTC 〈 sub 〉 4 〈 /sub 〉 from basophils challenged with allergen was suppressed by Dimaprit, a histamine H 〈 sub 〉 2 〈 /sub 〉 receptor agonist, at a concentration required to inhibit the release by 50% of 5 × 10 〈 sup 〉 ––5 〈 /sup 〉 〈 i 〉 M 〈 /i 〉 for histamine and 10 〈 sup 〉 ––5 〈 /sup 〉 〈 i 〉 M 〈 /i 〉 for LTC 〈 sub 〉 4 〈 /sub 〉 . These observations suggest that microtubules may be involved in LTC 〈 sub 〉 4 〈 /sub 〉 generation from immunologically stimulated basophils.

    Abstract: Two major allergens, DPI 〈 i 〉 (Der p 〈 /i 〉 I) and DP2 〈 i 〉 (Der p 〈 /i 〉 II), were isolated from the whole culture extract of 〈 i 〉 Dermatophagoides pteronyssinus, 〈 /i 〉 and the physicochemical and immunochemical properties of these allergens were compared with those of the corresponding allergens from 〈 i 〉 Dermatophagoides farinae, 〈 /i 〉 DF1 ( 〈 i 〉 Der f 〈 /i 〉 I)and DF2 ( 〈 i 〉 Der f 〈 /i 〉 II).On sodium dodecyl sulfate-polyacrylamide gel electrophoresis, polyacrylamide gel isoelectric focusing and amino acid analysis, both DP1 and DP2 were demonstrated to have close physicochemical similarity with DF1 and DF2, respectively. On immunodiffusion with the use of rabbit antisera, the two 〈 i 〉 Der 〈 /i 〉 I allergens showed the reaction of typical partial identity, while the two 〈 i 〉 Der 〈 /i 〉 II allergens showed the reaction of almost complete identity. Radioallergosorbent test (RAST) and RAST absorption experiments with the use of sera from mite-allergic patients showed that human IgE antibody response to the 〈 i 〉 Der 〈 /i 〉 I allergens was directed against both cross-reactive and species-specific determinants. In contrast, IgE antibodies to the 〈 i 〉 Der 〈 /i 〉 II allergens were demonstrated to react almost completely to cross-reactive determinants.

    Abstract: After intraperitoneal injection of 〈 i 〉 Bordetella pertussis 〈 /i 〉 vaccine to guinea pigs, the 〈 i 〉 α 〈 /i 〉 〈 sub 〉 1 〈 /sub 〉 〈 i 〉 - 〈 /i 〉 and 〈 i 〉 β 〈 /i 〉 -adrenergic and cholinergic muscarinic receptors of the whole lung were measured by binding assays with the radioisotope-labeled antagonists, 〈 sup 〉 3 〈 /sup 〉 H-prazosin, 1- 〈 sup 〉 3 〈 /sup 〉 H-dihydroalprenolol and 1- 〈 sup 〉 3 〈 /sup 〉 H-quinuclidinyl benzilate, respectively. Injection of guinea pigs with pertussis vaccine resulted in an increase in the maximum binding (B 〈 sub 〉 max 〈 /sub 〉 ) of 〈 sup 〉 3 〈 /sup 〉 H-prazosin, while there was about a 30% reduction in 1- 〈 sup 〉 3 〈 /sup 〉 H-dihydroalprenolol binding to 〈 i 〉 β 〈 /i 〉 -adrenergic receptors. No difference was observed in the dissociation constant (K 〈 sub 〉 D 〈 /sub 〉 ) for binding of each ligand to 〈 i 〉 α 〈 /i 〉 〈 sub 〉 1 〈 /sub 〉 〈 i 〉 - 〈 /i 〉 and 〈 i 〉 β 〈 /i 〉 -adrenergic receptors. 1- 〈 sup 〉 3 〈 /sup 〉 H-quinuclidinyl benzilate binding indicated that the B 〈 sub 〉 max 〈 /sub 〉 and K 〈 sub 〉 D 〈 /sub 〉 for cholinergic muscarinic receptor in pertussis-sensitized guinea pigs were not significantly different from those normal control animals.

    Abstract: The effect of immunization of guinea pigs with ovalbumin on the number and affinity of alpha 〈 sub 〉 1 〈 /sub 〉 - and beta-adrenergic and cholinergic muscarinic receptors was studied in lung membranes by direct binding techniques using 〈 sup 〉 3 〈 /sup 〉 H-prazosin, 〈 i 〉 l 〈 /i 〉 - 〈 sup 〉 3 〈 /sup 〉 H-dihydroalprenolol and 〈 i 〉 l 〈 /i 〉 - 〈 sup 〉 3 〈 /sup 〉 H-quinuclidinyl benzilate. After immunization by intraperitoneal injection of ovalbumin to guinea pigs, the number and affinity of each receptor in sensitized animals were not significantly different from those of control animals. Sensitization of guinea pigs by an aerosol exposure with the antigen resulted in a decreased number of beta-adrenergic receptor (458 ± 29 vs. 687 ± 56 fmol/mg protein; p 〈 0.01), and an increased number of alpha 〈 sub 〉 1 〈 /sub 〉 -adrenergic receptor (36 ± 2 vs. 25 ± 2 fmol/mg protein; p 〈 0.01), but no change was observed in the number of muscarinic receptors, as compared with control animals. On the other hand, following a prolonged sensitization of guinea pigs with a low dose of the aerosolized antigen, the number of muscarinic receptors was significantly increased in the lung of sensitized animals (50 ± 2 vs. 42 ± 2 fmol/mg protein; p 〈 0.01); however, we found no significant differences between sensitized and normal animals in the number of alpha 〈 sub 〉 1 〈 /sub 〉 - and beta-adrenergic receptors. There were no different changes in the affinity of these receptors in all experiments.

    Abstract: Two purified allergens, designated as DF1 and DF2, were isolated from the extract of the whole culture of 〈 i 〉 Dermatophagoides farinae 〈 /i 〉 by a combination of ammonium sulfate precipitation and ion exchange, hydrophobic, chelate and gel chromatography. DF1 was isolated as a heat-sensitive acidic protein with an apparent molecular weight of 25,000 and an isoelectric point of 4.6–7.2. DF2 was isolated as a heat-stable basic protein with an apparent molecular weight of 15,000 and an isoelectric point of 7.8–8.3. No allergenic cross-reactivity was seen between DF1 and DF2. Both DF1 and DF2 were shown to be the major allergens of 〈 i 〉 D. farinae 〈 /i 〉 by the results of radioallergosorbent test and histamine release assay.

    Abstract: Sensitive radioimmunoassays were developed to measure the two groups of major allergens ( 〈 i 〉 Der 〈 /i 〉 I and 〈 i 〉 Der 〈 /i 〉 II) of 〈 i 〉 Dermatophagoides pteronyssinus 〈 /i 〉 and 〈 i 〉 Dermatophagoides farinae 〈 /i 〉 , in which radiolabeled protein A served as a general tracer. Glass rods covalently coupled with F(ab′) 〈 sub 〉 2 〈 /sub 〉 fragments of rabbit antiserum IgG were incubated first with mite or house dust extracts, and then with affinity-purified rabbit antibodies. The bound allergen-antibody complex was detected with a 〈 sup 〉 125 〈 /sup 〉 I-labeled protein A. The two 〈 i 〉 Der 〈 /i 〉 I allergens, 〈 i 〉 Der p 〈 /i 〉 I and 〈 i 〉 Der f 〈 /i 〉 I, were measured separately with the use of immunoabsorbed rabbit antibodies directed against species-specific determinants, while the two 〈 i 〉 Der 〈 /i 〉 II allergens, 〈 i 〉 Der p 〈 /i 〉 II and 〈 i 〉 Der f 〈 /i 〉 II, were measured as ‘ 〈 i 〉 Der 〈 /i 〉 II’ with the use of antibodies directed against common determinants on both allergens. Each assay demonstrated consistently parallel dilution curves with mite and house dust extracts. The mean intraassay and interassay coefficient of variation for each assay ranged from 4.3 to 7.7% and from 3.6 to 9.0%, respectively. The 〈 i 〉 Der p 〈 /i 〉 I and 〈 i 〉 Der f 〈 /i 〉 I assays were shown to be highly species-specific so that the ratio of 〈 i 〉 Der p 〈 /i 〉 l: 〈 i 〉 Der f 〈 /i 〉 I would provide a good index of the distribution of the two mite species in a dust sample. The concentrations of the 〈 i 〉 Der 〈 /i 〉 I and 〈 i 〉 Der 〈 /i 〉 II allergens in different types of mite extract and dust samples from houses were compared using these assays. The results demonstrated that the ratio of 〈 i 〉 Der 〈 /i 〉 I: 〈 i 〉 Der 〈 /i 〉 II in the mite body extracts varied markedly from those in the whole culture extracts, while the ratio in individual dust samples was nearly constant and was close to that in the mite body extracts. These assays will be useful in standardizing mite and house dust extracts and for assessments of mite allergen exposure. Furthermore, the high assay sensitivity will make it possible to use these assays to measure mite allergens associated with airborne particles.
