    Abstract: Treatment for advanced gastric cancer is challenging. Epidermal growth factor receptor (EGFR) contributes to the proliferation and development of gastric cancer (GC), and its overexpression is associated with unfavorable prognosis in GC. Cetuximab, a monoclonal antibody targeting EGFR, failed to improve the overall survival of gastric cancer patients indicated in phase III randomized trials. Glutamine is a vital nutrient for tumor growth and its metabolism contributes to therapeutic resistance, making glutamine uptake an attractive target for cancer treatment. The aim of the present study was to investigate whether intervention of glutamine uptake could improve the effect of cetuximab on GC. The results of MTT assay showed that by glutamine deprivation or inhibition of glutamine uptake, the viability of gastric carcinoma cells was inhibited more severely than that of human immortal gastric mucosa epithelial cells (GES-1). The expression of the key glutamine transporter alanine-serine-cysteine (ASC) transporter 2 (ASCT2; SLC1A5) was significantly higher in gastric carcinoma tissues and various gastric carcinoma cell lines than in normal gastric tissues and cells, as shown by immunohistochemistry and western blotting, while silencing ASCT2 significantly inhibited the viability and proliferation of gastric carcinoma cells. Consistent with previous studies, it was shown herein by MTT and EdU assays that cetuximab had a weak inhibitory effect on the cell viability of gastric carcinoma cells. However, inhibiting glutamine uptake by blockade of ASCT2 with l-γ-glutamyl- p-nitroanilide (GPNA) significantly enhanced the inhibitory effect of cetuximab on suppressing the proliferation of gastric cancer both in vitro and in vivo. Moreover, combining cetuximab and GPNA induced cell apoptosis considerably in gastric carcinoma cells, as shown by flow cytometry, and had a higher depressing effect on gastric cancer proliferation both in vitro and in vivo, as compared to either treatment alone. The present study suggested that inhibition of glutamine uptake may be a promising strategy for improving the inhibitory efficacy of cetuximab on advanced gastric cancer.

    Abstract: This study investigated the association between soluble scavenger receptor differentiation antigen 163 (sCD163) and the severity and prognosis of renal injury in lupus nephritis (LN). Methods Serum sCD163 levels in 121 Eastern Chinese patients with LN who underwent renal biopsy were determined by enzyme-linked immunosorbent assays. Clinical data were collected, and the glomerular filtration rate and disease activity score of lupus were calculated. Pathological classification was performed, and renal pathological scores were assessed by the activity index (AI) and chronic index (CI). Kaplan–Meier survival curves were drawn to evaluate prognosis. Results The pathological classification, AI and CI scores in the high sCD163 group were increased. The sCD163 levels were positively correlated with serum creatinine, blood urea nitrogen, AI scores and CI scores and negatively correlated with the estimated glomerular filtration rate. Kaplan–Meier survival analysis showed that the incidence of renal endpoint events was increased in the high sCD163 group compared with the normal sCD163 group. Conclusion The serum sCD163 level correlates with the severity of LN and is an important indicator of poor renal prognosis in patients with LN.

    Abstract: The suction pressure pulsation plays a significant role in rotary compressor performance, so employing gas dynamic model for simulation is critical for compressor performance prediction and structure design. Comparing with the conventional Lax-wendroff two-step (LW2) scheme which has been usually utilized with good behaviors of speed and stability, this paper adopted the total variation diminishing scheme with higher convergence speed and less dispersion. And the common zero-dimensional model for the accumulator is updated to a one-dimensional model achieving more accurate pressure results at the suction pipe inlet. The simulation results of instantaneous pressure are also compared with experimental measured data under different conditions. The Pearson Correlation Coefficient of simulated pressure curves are all higher than 0.9 and the relative errors of predicted performance are almost all less than 5%, which indicates better agreement on pressure pulsation simulation and performance prediction.

    Abstract: Corydalis yanhusuo W. T. Wang (Papaveraceae) is a traditional Chinese herbal medicine that has long been used to treat several conditions and is widely distributed in Asian countries. This review focuses on the traditional uses, botany, phytochemistry, pharmacology, pharmacokinetics, and toxicology of C. yanhusuo. The literature on C. yanhusuo was reviewed using several resources, including classic books on Chinese herbal medicine and scientific databases, namely, PubMed, Springer, Web of Science, Science Direct, and China National Knowledge Infrastructure. Based on information from these databases regarding the chemical components of C. yanhusuo, we evaluated the underlying interaction network between chemical components, biological targets, and associated diseases using Cytoscape software. To date, more than 160 compounds have been isolated and identified from C. yanhusuo, including alkaloids, organic acids, volatile oils, amino acids, nucleosides, alcohols, and sugars. The crude extracts and purified compounds of this plant have analgesic, antiarrhythmic, and antipeptic ulcer properties, along with hypnotic effects. However, studies on the pharmacokinetics of C. yanhusuo extracts remain limited. C. yanhusuo has therapeutic potential in diseases such as cancer and depression, probably due to glaucine and corydaline. Our network pharmacology analysis revealed interactions between 20 compounds, 54 corresponding targets, and 4 health conditions. We found that leonticine, tetrahydroberberine, and corydalmine may regulate the expression of PTGS2, PTGS1, KCNH2, SCN5A, RXRA, CAMKK2, NCOA2, and ESR1, representing a potential treatment strategy against pain, gastric ulcers, inflammation, and cardiac arrhythmias. Additionally, this article discusses the future directions of research on C. yanhusuo.

    Abstract: Pulmonary hypertension (PAH) is one of the more serious complications of Chronic kidney disease (CKD), and its exact pathogenesis has not been clarified. As an upstream proinflammatory factor, macrophage migration inhibitor (MIF) is involved in the occurrence and development of many diseases. This study aimed to detect the relationship between serum MIF and PAH in non-dialysis CKD patients. A total of 382 non-dialysis CKD patients were enrolled in this study. Bio-Plex cytokine assay was used to detect MIF. CKD patients were divided into the PAH group and non-PAH group according to echocardiographic results. Relative risk was determined by logistic regression analysis. The pulmonary artery pressure in the CKD group was higher than that in the control group ( p  〈  0.01). Pulmonary arterial pressure was higher in stage 4 to 5 CKD patients than in Stage 1 to 3 CKD patients ( p  〈  0. 01), and the incidence of PAH was also increased ( p  〈  0. 01). MIF in the CKD group were higher than in the control group ( p  〈  0.05). MIF in CKD patients with PAH were higher than those without PAH ( p  〈  0.05). Multivariate logistic regression analysis showed that MIF is correlated with PAH (OR = 10.745; 95% CI 2.288–89.447, p  〈  0.05). PAH is common in non-dialysis CKD patients, and with the deterioration of kidney disease, the incidence of PAH is gradually increased, indicating that MIF plays an important role in the development of PAH in CKD patients.

    Abstract: Pin coupling anti-rotation mechanism is adopted in mechanically driven automotive scroll compressor due to its simple structure and high reliability. However, the sidewall of anti-rotation hole in pin coupling anti-rotation mechanism is easy to be worn out, deteriorating the performance of scroll compressor. To figure out the cause and take measures, a dynamic model for pin coupling anti-rotation mechanism is developed in this paper. The dynamic model takes two important factors into consideration, offsetting scroll wraps and radial compliance mechanism. Using the dynamic model, the contact force between the anti-rotation pin and anti-rotation hole can be determined, thus the wear zone can be predicted during one rotation cycle. A simulation program adopting the above dynamic model is developed to predict the performance of automotive scroll compressor. The error between simulated shaft power of compressor and experimental result is no more than 4% for all test conditions. The wear zone predicted by the simulation program agrees well with the actual scroll compressor which survived a durability test.

    Abstract: Bonded retainers are widely used to maintain the positions of anterior teeth after orthodontic treatment. Various types of bonded retainer exist however, there is a lack of evidence to indicate which type is superior. Aim: To compare upper and lower CAD/CAM nitinol bonded retainers (Memotain ® ) with upper and lower chairside rectangular-chain bonded retainers (Ortho-FlexTech™), in terms of stability, retainer failures and patient satisfaction. Trial design: Multi-centre, two-arm, parallel-group, randomised controlled clinical trial with 1:1 allocation. Setting: Three trial centres: University Teaching Hospital; District General Hospital; and Specialist Orthodontic Practice. All treatment was provided free as part of a state-funded healthcare system. Materials and methods: A total of 68 patients were randomly allocated to receive either upper and lower Memotain ® bonded retainers or upper and lower Ortho-FlexTech™ bonded retainers. Ten trained operators placed and reviewed the bonded retainers. Measurements were carried out on study models taken at debond and after six months. Patient satisfaction questionnaires were completed at six months following debond. Results: The trial was terminated due to the high number of failures (50%) of the upper Memotain® retainers within six months. Memotain ® retainers were three times more likely to fail (unadjusted hazard ratio = 2.82, 95% confidence interval = 1.00-7.99) than Ortho-FlexTech™ retainers at six months in the upper arch. Patients were satisfied with both types of retainer. Limitations: Early termination of the trial means that the a priori sample size was not reached, so outcomes should be interpreted with caution. Conclusion: The trial was terminated early due to the high failure rate of upper Memotain ® bonded retainers. They had a higher risk of failure in the maxillary arch when compared to upper Ortho-FlexTech™ bonded retainers after six months.

    Abstract: Elevated level of interleukin-8 (IL-8) promotes hepatocellular carcinoma (HCC) development and contributes to poor prognosis. Previously, we have proved that Dicer inhibits HCC progression. In this study, we evaluated the potential interaction between IL-8 and Dicer as well as their influence on HCC. Methods: Hepatocellular carcinoma cells of SMMC-7721 were divided into 2 groups for subsequent analysis: pCMV-Dicer group for Dicer-overexpressing lentivirus transfected cells (pCMV-Dicer cells) and pCMV-NC group for empty lentivirus transfected cells (pCMV-NC cells). Cell Counting kit-8 (CCK8), wound healing, and transwell were used to evaluate the inhibitory effect of Dicer overexpression on proliferation, migration, and invasion of HCC cells. The level of IL-8 was measured by flow cytometry bead-based immunoassays. Male nude BALB/c mice injected with pCMV-Dicer or pCMV-NC cell suspensions was used for transplant of HCC tumor. Results: We found that the secretion of IL-8 was reduced in the medium of pCMV-Dicer cells ( P = .027). Recombinant human IL-8 (rhIL-8) reversed the inhibitory effect of Dicer on proliferation ( P 〈 .01), migration ( P = .003), and invasion ( P = .001), whereas IL-8 inhibitor of reparixin enhanced inhibitory effect of Dicer on proliferation ( P 〈 .05), migration ( P = .008), and invasion ( P = .000). Lenvatinib downregulated the IL-8 level of HCC cells ( P = .000) as well as promote Dicer-induced inhibition for HCC cells referring to proliferation ( P 〈 .05), migration ( P = .000), and invasion ( P = .000). Animal experiments also demonstrated that Dicer cooperated with lenvatinib to inhibit the growth of HCC tumors ( P 〈 .05). Conclusions: Dicer cooperated with lenvatinib to inhibit HCC growth via downregulating IL-8, and Dicer displayed its potential capability to enhance the anti-tumor effect of lenvatinib.

    Abstract: Profile design plays an important role in improving the performance and reliability of scroll compressors. To expand design ideas of variable wall thickness profile (VWTP) of electric scroll compressor used for automobile air conditioner, a method to generate and modify VWTP is developed in this paper. The VWTP is generated according to the generalized profile theory and modified with symmetrical arc modification (SAM) and symmetrical arc plus line modification (SAPLM) at the central part. To make sure that the generated VWTP can form scroll profile, the generation conditions for coefficients of swing radius are discussed in detail. For the VWTP modified with SAM and SAPLM at the central part, a new way to classify scroll chambers and analyze the evolution of scroll chambers is proposed. Geometry analyses such as conjugate points on involute curves and arc segments, volume and its derivative with respect to orbiting angle are conducted as well. The generation and geometry analysis method of VWTP is validated through a design example of VWTP for electric scroll compressor with a displacement of 34 mL. The performance of the electric scroll compressor was tested through experiment and the results show that the cooling capacity can reach 7.5 kW and the COP can reach 3.0.
