    Abstract: The ability to express and purify modified recombinant proteins, so they retain their biological function in a cell-free format, has provided a basis for development of molecular biosensors. Here we utilize recombinant G Protein-coupled receptors (GPCRs) and their G proteins for cell-free detection of various binding partners. Fusion peptides were used to improve surface-attachment and fluorescent-labelling capabilities. A novel homogeneous fluorescence resonance energy transfer (FRET)-based assay was developed to detect rearrangements in the G protein heterotrimer. By using this heterotrimeric ‘molecular switch’, we are developing a generic technology such that multiple GPCRs could be assayed for ligand-mediated activation while tethered to surfaces or in solution, with increased throughput compared to current assay platforms.

    Abstract: A simple, disposable and inexpensive electrochemical DNA biosensor based on a zirconia (ZrO2) modified thin film screen-printed electrode (ZrO2/SPE) has been developed. Short DNA sequences (21 monomer units) from the Escherichia coli pathogen, modified with a phosphate group at the 5′ end, were attached to the surface of the electrode through the affinity of the phosphate group for zirconia, to produce an effective DNA probe (ssDNA/ZrO2/SPE). DNA immobilization and hybridization were characterized using differential pulse voltammetry by employing methylene blue as redox indicator. Target sequences hybridized with the probe resulted in a decrease of the reduction peak current of methylene blue intercalated into the probe. The response of a non-complementary sequence and a single base pair mismatch sequence were both clearly distinguished from that of a complementary sequence. The developed biosensor had a high selectivity and sensitivity towards hybridization detection (10–10 M complementary DNA detectable). Making use of screen-printed technology, the fabrication of the biosensors exhibited satisfactory reproducibility, investigated by cyclic voltammetry and differential pulse voltammetry. The relative standard deviation was found to be 〈 3.0% for six bare SPEs and six ssDNA-modified SPEs (ssDNA/ZrO2/SPE) from a batch.

    Abstract: The effect of methyl substitution on 15- and 17-membered macrocycles (incorporating N2O2- and N3O2-donors, respectively) on their interaction with cobalt(II), nickel(II), copper(II), zinc(II), cadmium(II) and lead(II) has been investigated. The results are compared with those obtained for the corresponding complexes of the unsubstituted (parent) rings. Binding constants for the formation of the metal complexes have been determined potentiometrically in 95% methanol (I = 0·1; Et4NClO4). An X-ray crystallographic study of [NiL(H2O)] (ClO4)2 (where L is the dimethyl derivative of a 17-membered, N3O2-donor macrocycle) confirms that the nickel ion is six-coordinate, with the complex cation exhibiting a distorted octahedral geometry defined by all five donor atoms of the O2N3-macrocycle (in its meso form) and the oxygen of the water molecule. In this structure the macrocyclic backbone incorporating the N3-donor fragment is arranged facially. Molecular mechanics modelling of this complex predicts that the observed facial configuration will be favoured over an alternative facial arrangement or a meridional one.

    Abstract: Methanol induced oxidative stress (OS) models in mice were successfully established and evaluated by the electron paramagnetic resonance (EPR) spin trapping technique. The capacity for removal of reactive oxygen species (ROS) free radicals by rhubarb and vitamin C (Vc) as candidate materials was also investigated. EPR was employed to determine the free radicals generated from a spin trapping agent, α-phenyl-N-tert-butylnitrone (PBN), that reacted with the ROS. The activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX), and the level of malondialdehyde (MDA) were also evaluated by enzyme assays. The results indicated that methanol clearly promoted the generation of ROS free radicals in the liver of mice. The activity of SOD and GSH-PX was reduced significantly, although the level of MDA was increased as a result of the harmful effect of methanol. In addition, rhubarb and Vc exhibited a protective effect on the mice liver under acute OS.

    Abstract: Using an established method, we isolated a large quantity of cucurbit[10]uril (Q[10] ) and prepared Q[10]-based supramolecular assemblies via different approaches. For example, the structure-directing agent [CdCl4] 2– was used or the Q[10] molecule itself acted as a self-structure-directing agent to form different Q[10] -based supramolecular assemblies through the outer surface interaction of Q[10]. Generally, the Q[10] -based supramolecular assemblies possess porous features that suggest that Q[n]-based compounds could be used to manufacture molecular sieves and sensors and applied in absorption and separation technologies.

    Abstract: Two new oxovanadium(iv) compounds containing 1,10-bathophenanthroline (Bphen) and amino Schiff base derivatives [VO(hnd-napha)(Bphen)] (1) and [VO(o-van-met)(Bphen)] (2) were synthesised (where hnd-napha and o-van-met are N-Schiff bases derived from the reaction of 2-hydroxy-1-naphthaldehyde with 3-(1-naphthyl)-l-alanine and o-vanillin with l-methionine, respectively). These compounds were characterised by elemental analysis, infrared spectroscopy, high-resolution mass spectrometry, and single-crystal X-ray diffraction (XRD). Both compounds showed low molar conductance values, indicating that they are non-electrolytes. The XRD results showed that the VIV atoms in both compounds existed in the VO3N3 coordination geometry with Schiff base and Bphen ligands. The in vitro anticancer activities of compounds 1 and 2 were evaluated against A549 human lung carcinoma and HepG2 human hepatoma cell lines using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, and the results revealed that both compounds were cytotoxic with half maximal inhibitory concentration (IC50) values in the range of 8.22 ± 1.0 to 94.89 ± 3.2 μmol L−1. Notably, compound 2 exhibited much better anticancer activity in vitro against A549 cells (8.22 ± 1 μmol L−1) than [VO(acac)2] (24 ± 6 μmol L−1) or any of our previously reported oxovanadium(iv) compounds, making it comparable in activity to cisplatin (3.1 ± 0.5 μmol L−1). These results therefore suggest that compound 2 could be used as a promising lead for the development of anticancer agents for the treatment of lung cancer.
