    Kurzfassung: Objective. This study aimed to explore follow-up mode changes for peritoneal dialysis (PD) patients and their effects on PD quality during the COVID-19 pandemic. Methods. A retrospective single-center study was conducted. All patients who received PD treatment at the Second Affiliated Hospital of Soochow University between January 2018 and March 2020 were enrolled in this study. Patient data during the first quarter of 2018 (Q1-2018), the first quarter of 2019 (Q1-2019), and the first quarter of 2020 (Q1-2020) were collected. Results. No significant differences were observed for any serum examinations in different follow-up periods ( P 〉 0.05 ). A significantly reduced outpatient follow-up rate was observed in Q1-2020 compared with Q1-2018 and Q1-2019 (71.6% Vs 78.9% Vs 84.7%, P 〈 0.001 ), accompanied by a significantly increased remote follow-up rate (28.4% Vs 21.1% Vs 15.3%, P 〈 0.001 ). Compared with Q1-2018 and Q1-2019, the hospitalization rate (27.7% Vs 30.9% Vs 15.7%, P 〈 0.001 ) and the incidence of peritonitis (0.162 Vs 0.186 Vs 0.08 per patient-year, P 〈 0.001 ) decreased significantly in Q1-2020. PD patients had a significant decline in the drop-out rate for Q1-2020 compared with Q1-2019 (4.4% Vs 7.3% Vs 2.2%, P 〈 0.001 ). No differences in the incidence of catheter-related infections were observed. No significant differences were observed for any peritoneal dialysis key performance indicators (KPIs) between outpatient follow-up and remote follow-up patients. Conclusion. During the COVID-19 pandemic (Q1-2020), our center practiced more remote follow-up procedures in PD patients. The hospitalization rate and peritonitis incidence were significantly decreased compared with the same time in previous years. No statistical differences were observed in other KPIs for peritoneal dialysis. This study shows that telehealth methods are a reasonable alternative to in-person care in the care/management of PD patients.

    Kurzfassung: Honeydew melon ( Cucumis melo L.) is an oval-shaped delicious fruit of one cultivar group of the muskmelon with immense nutritional importance and is extensively consumed by many tropical countries. The effect of various organic solvents on the recovery of phytochemicals from honeydew melon plant fruits and seeds was assessed. Further, High-Performance Liquid Chromatography (HPLC) was used to examine and assess the contents of phenolic acid (gallic acid) and flavonoid (rutin) compounds. The use of gas chromatography–mass spectrometry (GC-MS) analysis explained the presence of volatile phytocompounds in the extracts. The use of organic solvents had a substantial impact on the total dry weight and extract yield. In general, the solvent-extracted constituents remained in the order of methanol 〉 chloroform 〉 distilled water for both honeydew melon seeds and whole fruit. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) was used to assess the cytotoxicity effect against PC3, HCT116, HeLa, and Jurkat cell lines. The chloroform extract exhibited a good cytotoxic activity against all cell lines as compared to other solvent extracts. HPLC analysis revealed the occurrence of gallic acid content of 0.102 ± 0.23  mg/10 mg of dry whole fruit extract, while 10 mg of dry seed extract contained only 0.022 ± 0.12  mg of gallic acid content. Likewise, rutin content was observed to be 0.224 ± 0.31  mg and 0.1916 ± 0.82  mg/10 mg of dry whole fruit and seed extract, respectively. Further, GC-MS analysis revealed the presence of a total of 37 compounds in chloroform extract of whole fruit, while only 14 compounds were found in seed extract. Nevertheless, more examinations are needed to identify and characterize other metabolites from honeydew melon and evaluate their pharmacological importance.

    Kurzfassung: Background. This study is aimed at investigating the relationship between neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR), and hyperhomocysteinemia (HHCY) in patients with ischemic stroke (IS). Methods. This study retrospectively analyzed the clinical data and laboratory results of 110 IS patients. According to the cut-off value of HCY = 15   μ mol / L , the included patients were divided into the normal HCY (NHCY, n = 69 ) and the high HCY (HHCY, n = 41 ) groups. The relationship between NLR, PLR, and HCY between these groups was then compared. Results. NLR and PLR levels in IS patients with HHCY were significantly higher than those in IS patients without HHCY ( P 〈 0.05 ). Correlation analysis showed that NLR and PLR were positively correlated with hs-CRP, RDW, N, L, and HCY ( P 〈 0.05 ). NLR and PLR were the largest predictors of IS-related HHCY, followed by WBC, hs-CRP, RDW, VLDL, and TG. Additionally, NLR and PLR were independent risk factors for IS-related HHCY. Conclusions. NLR and PLR may be fast, cheap, and easy-to-analyze biomarkers for predicting IS-related HHCY.

    Kurzfassung: Kiaa1867 (human Kirre, hKirre) has a critical role in brain development and/or maintenance of the glomerular slit diaphragm in kidneys. Murine homolog of this gene, mKirre expressed in OP9 and AFT024 cells could support hematopoietic stem cells/hematopoietic progenitor cells (HSC/HPC) expansion in vitro. HKirre is also expressed in human FBMOB-hTERT cell line and fetal liver fibroblast-like cells but its function has remained unclear. In this paper, we cloned a hKirre gene from human fetal liver fibroblast-like cells and established a stably overexpressing hKirre-AFT024 cell line. Resultant cells could promote self-renewal and ex vivo expansion of HSCs/HPCs significantly higher than AFT024-control cells transformed with mock plasmid. The Expanded human umbilical cord blood (hUCB) CD34 + cells retained the capacity of multipotent differentiation as long as 8 weeks and successfully repopulated the bone marrow of sublethally irradiated NOD/SCID mice, which demonstrated the expansion of long-term primitive transplantable HSCs/HPCs. Importantly, hkirre could upregulate the expressions of Wnt-5A, BMP4, and SDF-1 and downregulate TGF- β with other hematopoietic growth factors. By SDS-PAGE and Western Blot analysis, a ~89 kDa protein in total lysate of AFT024-hKirre was identified. Supernatants from AFT024-hkirre could also support CD34 + CD38 − cells expansion. These results demonstrated that the AFT024-hKirre cells have the ability to efficiently expand HSCs/HPCs.
