    Abstract: The amygdala plays an important role in stress responses and stress-related psychiatric disorders. It is possible that amygdala connectivity may be a neurobiological vulnerability marker for stress responses or stress-related psychiatric disorders and will be useful to precisely identify the vulnerable individuals before stress happens. However, little is known about the relationship between amygdala connectivity and subsequent stress responses. The current study investigated whether amygdala connectivity measured before experiencing stress is a predisposing neural feature of subsequent stress responses while individuals face an emergent and unexpected event like the COVID-19 outbreak. Methods Data collected before the COVID-19 pandemic from an established fMRI cohort who lived in the pandemic center in China (Hubei) during the COVID-19 outbreak were used to investigate the relationship between amygdala connectivity and stress responses during and after the pandemic in 2020. The amygdala connectivity was measured with resting-state functional connectivity (rsFC) and effective connectivity. Results We found the rsFC of the right amygdala with the dorsomedial prefrontal cortex (dmPFC) was negatively correlated with the stress responses at the first survey during the COVID-19 outbreak, and the rsFC between the right amygdala and bilateral superior frontal gyri (partially overlapped with the dmPFC) was correlated with SBSC at the second survey. Dynamic causal modeling suggested that the self-connection of the right amygdala was negatively correlated with stress responses during the pandemic. Discussion Our findings expand our understanding about the role of amygdala in stress responses and stress-related psychiatric disorders and suggest that amygdala connectivity is a predisposing neural feature of subsequent stress responses.

    Abstract: Late Paleozoic magmatic rock outcrops are common in the Northern Alxa, Southern Central Asian Orogenic Belt (CAOB), which is a key area for understanding tectonic processes and defining the final closure time of the Paleo-Asian Ocean (PAO). We present zircon U‒Pb chronology and whole-rock geochemistry data for late Paleozoic magmatic rocks from the Yagan area of northern Alxa. This paper reveals two periods of magmatism: syenogranite (374.8 Ma) and bimodal intrusive rocks, which consist of gabbro (261.4 Ma), diabase (262.9 Ma) and biotite monzogranite (263.4 Ma). The syenogranite is high-K calc-alkaline and shows enrichments in Th, Zr, Hf and LREEs; depletions in Sr, Nb, Ta, and Ti; and low Mg# values (6.9–13.2); the syenogranite was derived from partial melting of the crust and has volcanic arc characteristics. The gabbro and diabase have similar geochemical characteristics, such as enrichments in Pb, Rb, Sr, Zr, and Hf and depletions in Nb, Ta, and Ti, with positive ε Hf (t) values (+0.9–+2.7 and +2.6–+3.6, respectively), indicating that they originated from partial melting of depleted mantle and experienced crustal contamination during magma emplacement. The biotite monzogranite shows depletions in Nb, Ta, and Ti and ε Nd (t) values of -2.6 to −2.4 and resulted from partial melting of the lower crust caused by asthenospheric underplating. The bimodal intrusive rocks formed in a postcollision extensional setting. Combined with previous data, we conclude that northern Alxa was an active continental margin during the late Devonian and that the final closure of the Yagan branch ocean of the PAO occurred prior to the middle Permian.

    Abstract: This study aimed to assess the host immune signatures associated with EBV infection and its clinical value in indicating the severity of children with acute infectious mononucleosis (IM). Methods Twenty-eight pediatric patients with IM aged 3–8 years were enrolled. The immune phenotypes and cytokine secretion capability of T cells were detected. Results The percentages and absolute numbers of CD3 + and CD8 + T cells were significantly increased in IM patients compared with HCs. The percentages of Naïve CD4 + and CD8 + T cells were decreased but with increased percentages of memory CD4 + and CD8 + T subsets. Our results showed the upregulation of active marker HLA-DR, TCR-αβ, and inhibitory receptors PD-1, TIGIT in CD8 + T cells from IM patients, which suggested that effective cytotoxic T cells were highly against EBV infection. However, EBV exposure impaired the cytokine (IFN-γ, IL-2, and TNF-α) secretion capability of CD4 + and CD8 + T cells after stimulation with PMA/ionomycin in vitro . Multivariate analysis revealed that the percentage of HLA-DR + CD8 + T cells was an independent prognostic marker for IM. The percentage of HLA-DR + CD8 + T cells was significantly correlated with high viral load and abnormal liver function results. Conclusion Robust expansion and upregulation of HLA-DR in CD8 + T cells, accompanied with impaired cytokine secretion, were typical characteristics of children with acute IM. The percentage of HLA-DR + CD8 + T cells might be used as a prominent marker not only for the early diagnosis but also for indicating the severity of IM.

    Abstract: Objective: Internet addiction has emerged as a growing concern worldwide. This study aimed to compare the prevalence of Internet addiction between left-behind children (LBC) and non-left-behind children (non-LBC), and explore the role of paternal and maternal parent-child communication on LBC. Methods: We conducted a cross-sectional survey in rural areas in Anhui, China. The complete data were available from 699 LBC and 740 non-LBC. Multivariable logistic regression was used to examine 1) whether LBC were more likely to develop Internet addiction, and 2) the association between parent-child communication and Internet addiction among LBC. Results: LBC had a higher likelihood to report Internet addiction when compared to non-LBC (OR = 2.03, 95%CI = 1.43–2.88, p & lt; 0.001). Among LBC, parent-child communication (both mother-child and father-child) was protective factor for children’s Internet addiction. The role of mother-child communication played well among male LBC. Conclusions: The lack of parental supervision may lead to Internet addiction. It is highly recommended for migrant parents to improve the quality of communication with their children. Also, gender-matching effects should be considered in the relationship between children’s behavior and parental factors.

    Abstract: Neuroimaging studies have highlighted gender differences in brain functions, but conclusions are not well established. Few studies paid attention to the influence of phase-encoding (PE) direction in echo-planar imaging on gender differences, which is a commonly used technique in functional magnetic resonance imaging (fMRI). A disadvantage of echo-planar images is the geometrical distortion and signal loss due to large susceptibility effects along the PE direction. The present research aimed to clarify how PE direction can affect the outcome of a specific research on gender differences. Methods We collected resting-state fMRI using anterior to posterior (AP) and posterior to anterior (PA) directions from 113 healthy participants. We calculated several commonly used indices for spontaneous brain activity including amplitude of low frequency fluctuations (ALFF), fractional ALFF (fALFF), regional homogeneity (ReHo), degree centrality (DC), and functional connectivity (FC) of posterior cingulate cortex for each session, and performed three group comparisons: (i) AP versus PA; (ii) male versus female; (iii) interaction between gender and PE direction. Results The estimated indices differed substantially between the two PE directions, and the regions that exhibited differences were roughly similar for all the indices. In addition, we found that multiple brain regions showed gender differences in these estimated indices. Further, we observed an interaction effect between gender and PE direction in the bilateral middle frontal gyrus, right precentral gyrus, right postcentral gyrus, right lingual gyrus, and bilateral cerebellum posterior lobe. Conclusion These apparent findings revealed that PE direction can partially influence gender differences in spontaneous brain activity of resting-state fMRI. Therefore, future studies should document the adopted PE direction and appropriate selection of PE direction will be important in future resting-state fMRI studies.

    Abstract: Neuroinflammation is considered to have a prominent role in the pathogenesis of Alzheimer’s disease (AD). Microglia are the resident macrophages of the central nervous system, and modulating microglia activation is a promising strategy to prevent AD. Essential oil of Jasminum grandiflorum L. flowers is commonly used in folk medicine for the relief of mental pressure and disorders, and analyzing the volatile compound profiles and evaluating the inhibitory effects of J. grandiflorum L. essential oil (JGEO) on the excessive activation of microglia are valuable for its application. This study aims to explore the potential active compounds in JGEO for treating AD by inhibiting microglia activation-integrated network pharmacology, molecular docking, and the microglia model. A headspace solid-phase microextraction combined with the gas chromatography–mass spectrometry procedure was used to analyze the volatile characteristics of the compounds in J. grandiflorum L. flowers at 50°C, 70°C, 90°C, and 100°C for 50 min, respectively. A network pharmacological analysis and molecular docking were used to predict the key compounds, key targets, and binding energies based on the detected compounds in JGEO. In the lipopolysaccharide (LPS)-induced BV-2 cell model, the cells were treated with 100 ng/mL of LPS and JGEO at 7.5, 15.0, and 30 μg/mL, and then, the morphological changes, the production of nitric oxide (NO) and reactive oxygen species, and the expressions of tumor necrosis factor-α, interleukin-1β, and ionized calcium-binding adapter molecule 1 of BV-2 cells were analyzed. A total of 34 compounds with significantly different volatilities were identified. α-Hexylcinnamaldehyde, nerolidol, hexahydrofarnesyl acetone, dodecanal, and decanal were predicted as the top five key compounds, and SRC, EGFR, VEGFA, HSP90AA1, and ESR1 were the top five key targets. In addition, the binding energies between them were less than −3.9 kcal/mol. BV-2 cells were activated by LPS with morphological changes, and JGEO not only could clearly reverse the changes but also significantly inhibited the production of NO and reactive oxygen species and suppressed the expressions of tumor necrosis factor-α, interleukin-1β, and ionized calcium-binding adapter molecule 1. The findings indicate that JGEO could inhibit the overactivation of microglia characterized by decreasing the neuroinflammatory and oxidative stress responses through the multi-compound and multi-target action modes, which support the traditional use of JGEO in treating neuroinflammation-related disorders.

    Abstract: Somatostatin-positive (SOM + ) neurons have been proposed as one of the key populations of excitatory interneurons in the spinal dorsal horn involved in mechanical pain. However, the molecular mechanism for their role in pain modulation remains unknown. Here, we showed that the T-type calcium channel Cav3.2 was highly expressed in spinal SOM + interneurons. Colocalization of Cacna1h (which codes for Cav3.2) and SOM tdTomato was observed in the in situ hybridization studies. Fluorescence-activated cell sorting of SOM tdTomato cells in spinal dorsal horn also proved a high expression of Cacna1h in SOM + neurons. Behaviorally, virus-mediated knockdown of Cacna1h in spinal SOM + neurons reduced the sensitivity to light touch and responsiveness to noxious mechanical stimuli in naïve mice. Furthermore, knockdown of Cacna1h in spinal SOM + neurons attenuated thermal hyperalgesia and dynamic allodynia in the complete Freund’s adjuvant-induced inflammatory pain model, and reduced both dynamic and static allodynia in a neuropathic pain model of spared nerve injury. Mechanistically, a decrease in the percentage of neurons with Aβ-eEPSCs and Aβ-eAPs in superficial dorsal horn was observed after Cacna1h knockdown in spinal SOM + neurons. Altogether, our results proved a crucial role of Cav3.2 in spinal SOM + neurons in mechanosensation under basal conditions and in mechanical allodynia under pathological pain conditions. This work reveals a molecular basis for SOM + neurons in transmitting mechanical pain and shows a functional role of Cav3.2 in tactile and pain processing at the level of spinal cord in addition to its well-established peripheral role.

    Abstract: This retrospective study aimed to investigate the clinical features of optic neuritis associated with COVID-19 (COVID-19 ON), comparing them with neuromyelitis optica-associated optic neuritis (NMO-ON), myelin oligodendrocyte glycoprotein-associated optic neuritis (MOG-ON), and antibody-negative optic neuritis (antibody-negative ON). Methods Data from 117 patients (145 eyes) with optic neuritis at the Shantou International Eye Center (March 2020–June 2023) were categorized into four groups based on etiology: Group 1 (neuromyelitis optica-related optic neuritis, NMO-ON), Group 2 (myelin oligodendrocyte glycoprotein optic neuritis, MOG-ON), Group 3 (antibody-negative optic neuritis, antibody-negative ON), and Group 4 (optic neuritis associated with COVID-19, COVID-19 ON). Characteristics of T2 and enhancement in orbital magnetic resonance imaging (MRI) were assessed. Best-corrected visual acuity (BCVA) was compared before treatment, at a short-term follow-up (14 days), and at the last follow-up after treatment. Results The COVID-19-associated optic neuritis (COVID-19 ON) group exhibited 100% bilateral involvement, significantly surpassing other groups ( P & lt; 0.001). Optic disk edema was observed in 100% of COVID-19 ON cases, markedly differing from neuromyelitis optica-related optic neuritis (NMO-ON) ( P = 0.023). Orbital magnetic resonance imaging (MRI) revealed distinctive long-segment lesions without intracranial involvement in T1-enhanced sequences for the COVID-19 ON group compared to the other three groups ( P & lt; 0.001). Discrepancies in optic nerve sheath involvement were noted between the COVID-19 ON group and both NMO-ON and antibody-negative optic neuritis (antibody-negative ON) groups ( P = 0.028). Before treatment, no significant difference in best-corrected visual acuity (BCVA) existed between the COVID-19 ON group and other groups. At the 14-day follow-up, BCVA in the COVID-19 ON group outperformed the NMO-ON ( P & lt; 0.001) and antibody-negative ON ( P = 0.028) groups, with no significant difference observed compared to the myelin oligodendrocyte glycoprotein optic neuritis (MOG-ON) group. At the last follow-up after treatment, BCVA in the COVID-19 ON group significantly differed from the NMO-ON group ( P & lt; 0.001). Conclusion Optic neuritis associated with COVID-19 (COVID-19 ON) predominantly presents with bilateral onset and optic disk edema. Orbital magnetic resonance imaging (MRI) demonstrates that COVID-19 ON presents as long-segment enhancement without the involvement of the intracranial segment of the optic nerve in T1-enhanced images. Glucocorticoid therapy showed positive outcomes.
