    Abstract: The impacts of thyroid hormones (TH) on lipid profile in euthyroid adults have gained much attention. It is currently unknown whether BMI influences such interaction. In the present study, we investigate the role of BMI in modulating the association between TH and lipid parameters in 1372 euthyroid healthy adults. Our results show that thyroid parameters are differentially associated with lipid profile. FT3 is positively correlated with total cholesterol ( β = 0.176 ± 0.046 , P 〈 0.001 ) and LDL cholesterol levels ( β = 0.161 ± 0.040 , P 〈 0.001 ). FT4 is negatively correlated with TG ( β = − 0.087 ± 0.029 , P 〈 0.01 ) while positively correlated with HDL cholesterol levels ( β = 0.013 ± 0.005 , P 〈 0.01 ). TSH is positively associated with TG ( β = 0.145 ± 0.056 , P 〈 0.05 ) and total cholesterol levels ( β = 0.094 ± 0.030 , P 〈 0.01 ). Importantly, BMI modulates the effect of TH on lipid profile: the interaction of FT4 and BMI and the interaction of FT3 and BMI reach statistical significance in predicting TG and HDL cholesterol levels, respectively. Stratified according to BMI levels, most associations between TH and lipid profile are significant only in normal-weight group. In conclusion, in euthyroid adults, high normal FT3, TSH levels, and low normal FT4 levels are associated with unfavorable lipid profile. BMI mediates the effect of thyroid function on lipid profile in euthyroid adults.

    Abstract: Adverse drug reactions (ADRs) are responsible for drug candidate failure during clinical trials. It is crucial to investigate biological pathways contributing to ADRs. Here, we applied a large-scale analysis to identify overrepresented ADR-pathway combinations through merging clinical phenotypic data, biological pathway data, and drug-target relations. Evaluation was performed by scientific literature review and defining a pathway-based ADR-ADR similarity measure. The results showed that our method is efficient for finding the associations between ADRs and pathways. To more systematically understand the mechanisms of ADRs, we constructed an ADR-pathway network and an ADR-ADR network. Through network analysis on biology and pharmacology, it was found that frequent ADRs were associated with more pathways than infrequent and rare ADRs. Moreover, environmental information processing pathways contributed most to the observed ADRs. Integrating the system organ class of ADRs, we found that most classes tended to interact with other classes instead of themselves. ADR classes were distributed promiscuously in all the ADR cliques. These results reflected that drug perturbation to a certain pathway can cause changes in multiple organs, rather than in one specific organ. Our work not only provides a global view of the associations between ADRs and pathways, but also is helpful to understand the mechanisms of ADRs.

    Abstract: Objective . The transport and metabolism of glucose are important during mammalian development. High glucose can mediate the biological characteristics of mesenchymal stem cells (MSCs). However, the role of high glucose in the odonto/osteogenic differentiation of stem cells from apical papilla (SCAPs) is unclear. Materials and Methods . SCAPs were isolated and identified in vitro . Then, SCAPs were cultured in normal α -MEM and high glucose α -MEM separately. MTT assay was applied to observe the proliferation of SCAPs. ALP activity, alizarin red staining, real-time RT-PCR, and western blot were used to detect the odonto/osteogenic capacity of SCAPs as well as the participation of NF- κ B pathway. Results . SCAPs in 25mmol/L glucose group expressed the maximum proteins of RUNX2 and ALP as compared with those in 5, 10, and 15 mmol/L groups. MTT assay showed that 25 mmol/L glucose suppressed the proliferation of SCAPs. ALP assay, alizarin red staining, real-time RT-PCR, and western blot showed 25 mmol/L high glucose can obviously enhance the odonto/osteogenic capacity of SCAPs. Moreover, the NF- κ B pathway was activated in 25mmol/L glucose-treated SCAPs and the odonto/osteogenic differentiation was inhibited following the inhibition of NF- κ B signaling pathway. Conclusions . High glucose can enhance the odonto/osteogenic capacity of SCAPs via NF- κ B pathway.

    Abstract: Background. Oral and maxillofacial bone loss is highly prevalent among populations, and nowadays, increased attention has been focused on dentin derivatives serving as desirable graft materials for bone regeneration. In this study, dentin-derived inorganic mineral (DIM) was fabricated with a high-temperature calcination technique and the effects of DIM on the osteogenic differentiation of bone marrow-derived mesenchymal stem cells (BMMSCs) and the bone formation were elucidated. Methods. The effects of DIM on BMMSC proliferation and apoptosis capacity were evaluated by CCK-8, flow cytometry, and EdU assays. Alkaline phosphatase (ALP) activity detection, ALP staining, alizarin red staining, and osteogenic marker expression analysis were performed to investigate the influence of DIM on the osteogenic differentiation of BMMSCs, as well as the relevant signal mechanisms. The model of critical-sized defects in the calvarium of rats was constructed for exploring the in vivo efficiency of DIM on bone regeneration. Results. Cell viability assays indicated that DIM had no cytotoxicity. BMMSCs cultured with DIM presented a higher level of osteogenic differentiation ability than those in the control group. The activation in ERK and p38 signals was detected in DIM-treated BMMSCs, and both pathways and osteogenic process were suppressed while using ERK inhibitor U0126 and p38 inhibitor SB203580, respectively. Furthermore, the animal experiments revealed that DIM could dramatically enhance new bone formation compared to the control group. Conclusion. DIM could promote BMMSC osteogenic differentiation via triggering the ERK and p38 MAPK signaling pathways and might be a novel predictable material for facilitating bone formation.

    Abstract: When massive numbers of wireless IoT devices are being deployed, cognitive spectrum management is critical to satisfy the explosive broadband requirements of IoT applications. Heterogeneous the target of the spatial spectrum estimation which is part of array signal processing is to achieve the evaluation of space signal parameters and source location, which result in that the spatial spectrum estimation becomes the most basic content of the array signal processing. It needs a method by which the large dimensional array still has its consistency. Therefore, this paper studies an improved large dimensional array parameterized spatial spectrum estimation method based on Pisarenko method, named G-Pisarenko method. Firstly, an improved estimator about the logarithm of the covariance matrix of a certain bilinear form is analyzed which is based on the theory of large dimension random matrix. We can find out a relatively better method, i.e., MW method. The method will become the primitive method for us to improve. Then, aimed at the relating covariance matrix in MW, we use an improved large dimensional array estimation method which can improve the logarithm of the covariance matrix estimation. Finally, we compare the improved method and the original method by simulation, and it can be seen the clear advantage of G-Pisarenko method when the sample number and observed dimensions are in the same order of magnitude.

    Abstract: Yunnan Baiyao is a traditional Chinese herbal remedy that has long been used for its characteristics of wound healing, bone regeneration, and anti-inflammation. However, the effects of Yunnan Baiyao on the odonto/osteogenic differentiation of stem cells from apical papilla (SCAPs) and the potential mechanisms remain unclear. The aim of this study was to investigate the odonto/osteogenic differentiation effects of Yunnan Baiyao on SCAPs and the underlying mechanisms involved. SCAPs were isolated and cocultured with Yunnan Baiyao conditioned media. The proliferation ability was determined by cell counting kit 8 and flow cytometry. The differentiation capacity and the involvement of NF- κ B pathway were investigated by alkaline phosphatase assay, alizarin red staining, immunofluorescence assay, real-time RT-PCR, and western blot analyses. Yunnan Baiyao conditioned medium at the concentration of 50  μ g/mL upregulated alkaline phosphatase activity, induced more mineralized nodules, and increased the expression of odonto/osteogenic genes/proteins (e.g., OCN /OCN, OPN /OPN, OSX /OSX, RUNX2 /RUNX2, ALP /ALP, COL-I /COL-I, DMP1, DSP /DSPP) of SCAPs. In addition, the expression of cytoplasmic phos-I κ B α , phos-P65, and nuclear P65 was significantly increased in Yunnan Baiyao conditioned medium treated SCAPs in a time-dependent manner. Conversely, the differentiation of Yunnan Baiyao conditioned medium treated SCAPs was obviously inhibited when these stem cells were cocultured with the specific NF- κ B inhibitor BMS345541. Yunnan Baiyao can promote the odonto/osteogenic differentiation of SCAPs via the NF- κ B signaling pathway.

    Abstract: The latest developments in edge computing have paved the way for more efficient data processing especially for simple tasks and lightweight models on the edge of the network, sinking network functions from cloud to edge of the network closer to users. For the reform of English teaching mode, this is also an opportunity to integrate information technology, providing new ideas and new methods for the optimization of English teaching. It improves the efficiency of English reading teaching, stimulates the interest of English learning, enhances students’ autonomous learning ability, and creates favorable conditions for students’ learning and development. This paper designs a MEC-based GNN (GCN-GAN) user preference prediction recommendation model, which can recommend high-quality video or picture text content to the local MEC server based on user browsing history and user preferences. In the experiment, the LFU-LRU joint cache placement strategy used in this article has a cache hit rate of up to 99%. Comparing the GCN-GAN model with other traditional graph neural network models, it performs caching experiments on the Douban English book data and Douban video data sets. The GCN-GAN model has a higher score on the cache task, and the highest speculation accuracy value F1 can reach 86.7.

    Abstract: The calcined tooth powder (CTP), a type of allogeneic biomimetic mineralized material, has been confirmed that can promote new bone formation when obtained at high temperature. The aim of this study was to investigate effects of the conditioned medium of calcined tooth powder (CTP-CM) on the osteogenic and odontogenic differentiation of human dental pulp stem cells (hDPSCs) and the underlying mechanisms involved. First, ALP activity assay determined that 200  μ g/mL was the optimal concentration of CTP-CM for the following experiments. CTP-CM had no significant effect on the proliferation of hDPSCs as indicated by CCK-8 and FCM analysis. Both the gene and protein ( DSPP /DSPP, RUNX 2/RUNX2, OCN /OCN, OSX /OSX, OPN /OPN, ALP /ALP, and COL -1/COL-1) expression levels increased in the CTP-CM-induced hDPSC group as compared with those in the control group at day 3 or 7, showing the positive regulation of CTP-CM on the osteo/odontogenic differentiation of hDPSCs. Mechanistically, MAPK signaling pathways were activated after the CTP-CM treatment, and the inhibitors targeting MAPK were identified which weakened the effects of CTM-CM on the committed differentiation of hDPSCs. These findings could lead to the creation of stem cell therapies for dental regeneration.
