    Abstract: Many post-transcriptional mRNA processing steps play crucial roles in tumorigenesis and the progression of cancers, such as N6-methyladenosine (m6A) modification and alternative splicing. Upregulation of methyltransferase-like 3 (METTL3), the catalytic core of the m6A methyltransferase complex, increases m6A levels and results in significant effects on the progression of hepatocellular carcinoma (HCC). However, alternative splicing of METTL3 has not been fully investigated, and the functions of its splice variants remain unclear. Here, we analyzed both our and online transcriptomic data, obtaining 13 splice variants of METTL3 in addition to canonical full-length METTL3-A in HCC cell lines and tissues. Validated by RT–qPCR and Western blotting, we found that METTL3-D, one of the splice variants expressing a truncated METTL3 protein, exhibits higher levels than METTL3-A in normal human livers but lower levels than METTL3-A in HCC tumor tissues and cell lines. Further functional assays demonstrated that METTL3-D expression decreased cellular m6A modification, inhibited the proliferation, migration, and invasion of HCC cells, and was negatively associated with the malignancy of patient tumors, exhibiting functions opposite to those of full-length METTL3-A. This study demonstrates that the METTL3-D splice variant is a tumor suppressor that could potentially be used as a target for HCC therapy.

    Abstract: Rice blast is one of the main diseases in rice and can occur in different rice growth stages. Due to the complicated procedure of panicle blast identification and instability of panicle blast infection influenced by the environment, most cloned rice resistance genes are associated with leaf blast. In this study, a rice panicle blast resistance gene, Pb2, was identified by genome-wide association mapping based on the panicle blast resistance phenotypes of 230 Rice Diversity Panel 1 (RDP1) accessions with 700,000 single-nucleotide polymorphism (SNP) markers. A genome-wide association study identified 18 panicle blast resistance loci (PBRL) within two years, including 9 reported loci and 2 repeated loci (PBRL2 and PBRL13, PBRL10 and PBRL18). Among them, the repeated locus (PBRL10 and PBRL18) was located in chromosome 11. By haplotype and expression analysis, one of the Nucleotide-binding domain and Leucine-rich Repeat (NLR) Pb2 genes was highly conserved in multiple resistant rice cultivars, and its expression was significantly upregulated after rice blast infection. Pb2 encodes a typical NBS-LRR protein with NB-ARC domain and LRR domain. Compared with wild type plants, the transgenic rice of Pb2 showed enhanced resistance to panicle and leaf blast with reduced lesion number. Subcellular localization of Pb2 showed that it is located on plasma membrane, and GUS tissue-staining observation found that Pb2 is highly expressed in grains, leaf tips and stem nodes. The Pb2 transgenic plants showed no difference in agronomic traits with wild type plants. It indicated that Pb2 could be useful for breeding of rice blast resistance.

    Abstract: Copper (II) complexes containing mixed ligands were synthesized in dimethyl formamide (DMF). The intense cyan emission at an ambient temperature is observed for solid copper (II) complexes with salicylic acid and a 12% quantum yield with a fluorescent lifetime of approximately 10 ms. Hence, copper (II) complexes with salicylic acid are excellent candidates for photoactive materials. Fourier transform infrared spectroscopy (FTIR) and X-ray photoelectron spectroscopy (XPS) reveal that the divalent copper metal centers coordinate with the nitrogen and oxygen lone pairs of conjugate ligands. XPS binding energy trends for core electrons in lower-lying orbitals are similar for all three copper (II) complexes: nitrogen 1s and oxygen 1s binding energies increase relative to those for undiluted ligands, and copper 2p3/2 binding energies decrease relative to that for CuCl2. The thermal behavior of these copper complexes reveals that the thermal stability is characterized by the following pattern: Cu(1,10-phenanthroline)(salicylic acid) 〉 Cu(1,10-phenanthroline)(2,2’-bipyridine) 〉 Cu(1,10-phenanthroline)(1-benzylimidazole)2.

    Abstract: Rice blast is a worldwide fungal disease that seriously affects the yield and quality of rice. Identification of resistance genes against rice blast disease is one of the effective ways to control this disease. However, panicle blast resistance genes, which are useful in the fields, have rarely been studied due to the difficulty in phenotypic identification and the environmental influences. Here, panicle blast resistance-3 (Pb3) was identified by a genome-wide association study (GWAS) based on the panicle blast resistance phenotypes of 230 Rice Diversity Panel I (RDP-I) accessions with 700,000 single-nucleotide polymorphism (SNP) markers. A total of 16 panicle blast resistance loci (PBRLs) within three years including one repeated locus PBRL3 located in chromosome 11 were identified. In addition, 7 genes in PBRL3 were identified as candidate genes by haplotype analysis, which showed significant differences between resistant and susceptible varieties. Among them, one nucleotide-binding domain and Leucine-rich Repeat (NLR) gene Pb3 was highly conserved in multiple resistant rice cultivars, and its expression was significantly induced after rice blast inoculation. Evolutionary analysis showed that Pb3 was a typical disease resistance gene containing coiled-coil, NB-ARC, and LRR domains. T-DNA insertion mutants and CRISPR lines of Pb3 showed significantly reduced panicle blast resistance. These results indicate that Pb3 is a panicle blast resistance gene and GWAS is a rapid method for identifying panicle blast resistance in rice.

    Abstract: In this research contribution, nano-aggregates have been fabricated by introducing lanthanide (Ln3+) ions into solutions of amphiphilic diblock copolymers of polystyrene-b-poly (acrylic acid) (PS-b-PAA). The coordination of acrylic acid segments to lanthanide cations induces diblock copolymer (BCPs) self-assembly in order to design stable white luminescent hybrid nanoparticles with fine uniform particle size. The introduction of Ln3+ ions (Eu3+ and Tb3+) bestows the micelles, precisely white light, upon excitation of 342 nm. Lanthanide coordination cross-linking of poly (acrylic acid) segments, or blocks, endows the micelles higher thermal stability than that of BCPs micelles without cross-linking. As the most important key point of this work, the regular and stable nano-particles with high emission quality can make fully flexible electroluminescent devices with self-formation or uncoordinated into polymer hosts. Instead of inorganic luminescent nanoparticles with hard cores, this method can potentially apply for fully flexible white-light emitting diodes (FFWLEDs).

    Abstract: To examine the impacts of various straw mulching techniques, this study used the indoor soil column test as the primary research method and the field test as the validation test on the salinity dynamics of saline and alkaline soils. The experiment in this study was designed with five treatments: SC means for straw covered on the soil surface; DB means for straw buried 40 cm below the soil surface; S1D1, S2D1, and S1D2 represent the ratio of soil surface cover to the amount of straw buried 40 cm below the soil surface as 1:1, 2:1, and 1:2, respectively. The results of the indoor soil column test showed that all kinds of straw mulching techniques could effectively reduce soil moisture evaporation, and the straw mulching and deep burial mode was more effective: after 45 days of evaporation, compared with that of CK, the cumulative evaporation of soil moisture were reduced by 29.61%, 27.49%, 37.87%, 65.85%, and 54.58% for SC, DB, S1D1, S2D1, and S1D2, respectively; the straw mulching and deep burial mode could reduce the soil evaporation intensity more effectively than the single-layer straw mulching mode: the mean soil evaporation rates of CK, SC, DB, S1D1, S2D1 and S1D2 after 45 days of evaporation were 1.27 mm/day, 0.90 mm/day, 0.92 mm/day, 0.80 mm/day, 0.43 mm/day, and 0.58 mm/day; various straw mulching techniques could inhibit the accumulation of salts in the surface soil and effectively regulate the distribution of salts in the soil profile, among which the straw mulching and deep burial mode had the best effect of salinity suppression: after 30 days of evaporation, the re-salinization levels of the 0–40 cm soil layer of SC, DB, S1D1, S2D1, and S1D2 were reduced by 66.78%, 43.08%, 33.95%, 92.04% and 45.94% compared with that in the CK, respectively; there was a significant positive correlation between cumulative evaporation of soil moisture and cumulative soil salinity, which implied that cumulative soil salinity increased with the increase in cumulative evaporation of soil moisture. The results of the field experiment justified the results of the indoor soil column test: after four months of evaporation, the field moisture contents of CK, SC, DB, S1D1, S2D1, and S1D2 in the 0–20 cm soil layer were 14.77%, 3.51%, 15.10%, 15.26%, 18.73%, and 2.94%, respectively; during the experimental period, the salt inhibition rate of SC, DB, S1D1, S2D1 and S1D2 in 0–20 cm soil layer were 35.46%, 44.76%, 50.98%, 54.80% and 37.30%, respectively. Therefore, in a comprehensive view, S2D1 treatment had the best effect of salt and vapor suppression on saline soil. This study is of great significance for the resource utilization of straw waste, the improvement of water utilization and efficiency, and the management of soil salinization.

    Abstract: In response to the fast-waning immune response and the great threat of the Omicron variant of concern (VOC) to the public, we report the pilot-scale production of an inactivated Omicron vaccine candidate that induces high levels of neutralizing antibody titers to protect against the Omicron virus. Here, we demonstrate that the inactivated Omicron vaccine is safe and effective in recalling immune responses to the HB02, Omicron, and Delta viruses after one or two doses of BBIBP-CorV. In addition, the efficient productivity and good genetic stability of the manufactured inactivated vaccine is proved. These results support the further evaluation of the Omicron vaccine in a clinical trial.

    Abstract: A hybrid polymer of SiO2@Tb3+(poly(ethylene terephthalate)-tetraglycol)3 phenanthroline (SiO2@Tb3+(PET–TEG)3Phen) was synthesized by mixing of inorganic SiO2 nanoparticles with polymeric segments of PET–TEG, whereas PET–TEG was achieved through multi-step functionalization strategy. Tb3+ ions and β-diketonate ligand Phen were added in resulting material. The experimental results demonstrated that it was well blended with PET as a robust additive, and not only promoted the crystallinity, but also possessed excellent luminescence properties. An investigation of the mechanism revealed that the SiO2 nanoparticles functioned as a crystallization promotor; the Tb3+ acted as the fluorescent centre; and the PET–TEG segments played the role of linker and buffer, providing better compatibility of PET matrix with the inorganic component. This work demonstrated that hybrid polymers are appealing as multifunctional additives in the polymer processing and polymer luminescence field.

    Abstract: The 2021 Mw 7.4 Maduo (Madoi) earthquake that struck the northern Tibetan Plateau resulted in widespread coseismic deformation features, such as surface ruptures and soil liquefaction. By utilizing the unmanned aerial vehicle (UAV) photogrammetry technology, we accurately recognize and map 39,286 liquefaction sites within a 1.5 km wide zone along the coseismic surface rupture. We then systematically analyze the coseismic liquefaction distribution characteristics and the possible influencing factors. The coseismic liquefaction density remains on a higher level within 250 m from the surface rupture and decreases in a power law with the increasing distance. The amplification of the seismic waves in the vicinity of the rupture zone enhances the liquefaction effects near it. More than 90% of coseismic liquefaction occurs in the peak ground acceleration (PGA) 〉 0.50 g, and the liquefaction density is significantly higher in the region with seismic intensity 〉 VIII. Combined with the sedimentary distribution along-strike of the surface rupture, the mapped liquefaction sites indicate that the differences in the sedimentary environments could cause more intense liquefaction on the western side of the epicenter, where loose Quaternary deposits are widely spread. The stronger coseismic liquefaction sites correspond to the Eling Lake section, the Yellow River floodplain, and the Heihe River floodplain, where the soil is mostly saturated with loose fine-grained sand and the groundwater level is high. Our results show that the massive liquefaction caused by the strong ground shaking during the Maduo (Madoi) earthquake was distributed as the specific local sedimentary environment and the groundwater level changed.
