    Abstract: Isoflavones are a class of flavonoids that belong to a large family of polyphenols and synthesized predominantly in legume, and they play important roles including acting as antioxidant, preventing osteoporosis, reducing the risk of atherosclerosis, and protecting against cardiovascular disease. This study focused on the accumulation and synthetic metabolism of isoflavone in soybean hypocotyl and cotyledon calluses under UV-B radiation. The results showed that UV-B radiation significantly up-regulated the gene expression of phenylalanine ammonia lyase (PAL), cinnamate-4-hydroxylase (C4H), 4-coumarate-CoA ligase (4CL), chalcone ketone synthase (CHS), chalcone isomerase (CHI), and isoflavone synthase (IFS), and enhanced their activity in soybean hypocotyl and cotyledon calluses. As a result, isoflavones content increased by 21.23 and 21.75% in soybean hypocotyl and cotyledon calluses, respectively. Among the isoflavones produced, malonyldaidzin was the dominant one in hypocotyl callus, while malonylglycitin and daidzein were the main isoflavones in cotyledon calluses. This study revealed that UV-B radiation induced isoflavone accumulation in soybean calluses, which could be an efficient strategy to improve the nutritional value of food and produce high levels of bioactive secondary metabolites.

    Abstract: The collapse or plugging of gas extraction borehole can result in partial or complete failure of the extraction function, affecting the efficiency of gas extraction and increasing project costs. The integrated friction coefficient k indicates the resistance of the borehole repair system per unit length. The current repairing methods are mainly self-propelled and rotary drilling technology, which ignores the influence of drilling angle and return water flow on integrated friction coefficient k . In this paper, wellbore repair model based on Newton’s variable mass law is established. The relationship between repair length and hydraulic parameters, wellbore parameters and pipeline parameters is derived. The system resistance test experiment is carried out to clarify the influence of well length, angle, backwater and movement speed on the integrated friction coefficient k . Main conclusions are as follows: the integral friction coefficient k is the key parameter of the repair model, the system resistance increases linearly with the increase of drilling repair distance. Backwater exerts backward thrust on the front end of the drill bit and the high-pressure hose, resulting in an increase in the integrated friction coefficient k, the increase of drilling angle and jet pressure can lead to the increase of backwater flow and the increase of integral friction coefficient k . The improved repair model can more accurately predict the maximum repair distance of self-propelled and rotary drill bits. At the repair distance of 104.7 m, the error between the repair value and the experimental value is 5.7 m, which verifies the feasibility of the application of drilling repair in self-propelled and rotary drilling technology.

    Abstract: Wheat stripe rust is a destructive disease in many cool and temperate regions around the world. Exploiting novel sources of resistance can provide wheat cultivars with robust and durable resistance to stripe rust. The wheat– Thinopyrum intermedium addition line TAI-14 was proven to carry a stripe rust resistance gene (named as YrT14 ) on the alien Th. intermedium chromosome. In order to transfer the resistance gene to wheat, wheat– Th. intermedium translocation lines were created by irradiating the pollen of the line TAI-14. We totally obtained 153 wheat– Th. intermedium translocation lines, among which the long alien segmental translocation line Zhongke 78 and the intercalary translocation line Zhongke 15 not only showed good integrated agronomic traits but also were identified as highly resistant to stripe rust in both seedling and adult plant stages. The alien chromatin in Zhongke 15 was identified as an insertion into the satellite of chromosome 6B, a type of translocation never reported before in chromosome engineering. By screening Simple Sequence Repeat (SSR) and Expressed Sequence Tag (EST) markers as well as the markers developed from RNA-sequencing (RNA-Seq) data, 14 markers were identified specific for the alien chromosome and a physical map was constructed. Both Zhongke 78 and Zhongke 15 could be used as a novel source of stripe rust resistance for wheat breeding, and the linked marker T14K50 can be used for molecular marker–assisted breeding. Finally, based on the karyotype, reaction to stripe rust, and genome resequencing data of different wheat– Th. intermedium translocation lines, the stripe rust resistance gene YrT14 was located to an 88.1 Mb interval from 636.7 to 724.8 Mb on Th. intermedium chromosome 19 corresponding to 7J or 7J s .
