    Abstract: The never-ending emergence of SARS-CoV-2 variations of concern (VOCs) has challenged the whole world for pandemic control. In order to develop effective drugs and vaccines, one needs to efficiently simulate SARS-CoV-2 spike receptor-binding domain (RBD) mutations and identify high-risk variants. We pretrain a large protein language model with approximately 408 million protein sequences and construct a high-throughput screening for the prediction of binding affinity and antibody escape. As the first work on SARS-CoV-2 RBD mutation simulation, we successfully identify mutations in the RBD regions of 5 VOCs and can screen millions of potential variants in seconds. Our workflow scales to 4096 NPUs with 96.5% scalability and 493.9× speedup in mixed-precision computing, while achieving a peak performance of 366.8 PFLOPS (reaching 34.9% theoretical peak) on Pengcheng Cloudbrain-II. Our method paves the way for simulating coronavirus evolution in order to prepare for a future pandemic that will inevitably take place. Our models are released at https://github.com/ZhiweiNiepku/SARS-CoV-2_mutation_simulation to facilitate future related work.

    Abstract: Poly(3-hydroxybutyrate-co-4-hydroxybutyrate), poly(ε-caprolactone), and poly(1actide-co-glycolide) films were prepared by solution casting and electrospinning. The degradability of cast films was evaluated in vitro and in vivo by recording the weight remaining, observing the morphology, and measuring change in molecular weight and M w / M n polydispersity. In addition, cell proliferation on electrospun films was tested by MTT assay, and cell infiltration into electrospun films was assessed through subcutaneous implantation in rats. By analyzing data obtained from these tests, we discovered that the degradation rates of poly(3-hydroxybutyrate-co-4-hydroxybutyrate) copolymers were between those of poly(ε-caprolactone) and poly(1actide-co-glycolide) and could be controlled by adjusting 4-hydroxybutyrate content. Moreover, tissue responses to materials made from poly(3-hydroxybutyrate-co-4-hydroxybutyrate) were milder compared with those elicited by poly(ε-caprolactone) and poly(1actide-co-glycolide), and poly(3-hydroxybutyrate-co-4-hydroxybutyrate) films exhibited a good and moderate cell infiltration. Therefore, our data demonstrate that poly(3-hydroxybutyrate-co-4-hydroxybutyrate) copolymers are promising candidates for the construction of scaffolds designed for various types of tissue regeneration with special biodegradability requirements.

    Abstract: Biodegradable pH-sensitive amphiphilic block polymer (mPEG-Hyde-PLGA) was synthesized via ring-opening polymerization, initiated from a hydrazone-containing macro-initiator. In this way, a pH-sensitive hydrazone bond was inserted into the backbone of block copolymer, linking hydrophilic poly(ethylene glycol) segment and hydrophobic poly(lactic-co-glycolic acid) segment. The copolymer self-assembled to form stable micelles with mean diameters below 100 nm and served as a drug delivery system for doxorubicin, with drug loading content of 5.3%. pH sensitivity of the hydrazone-containing micelles was investigated by changes in diameter and size distribution observed by dynamic light scattering measurements when the micelles were encountered to acidic medium. Small pieces and larger aggregates were found by transmission electron microscopy resulting from the disassociation of the micelles in acidic conditions. It was also noted that doxorubicin release from the pH-sensitive micelles is significantly faster at pH 4.0 and pH 5.0 compared to pH 7.4, while almost no difference was detected in the case of pH non-sensitive micelles. MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assays on HepG-2 and MCF-7 cells revealed that doxorubicin-loaded pH-sensitive micelles had higher antitumor activity than pH-insensitive ones. This pH-sensitive drug delivery system based on hydrazone-containing block copolymer has been proved as a promising drug formulation for cancer therapy.

    Abstract: The Prognostic Nutritional Index (PNI) has been reported to be correlated with long-term outcomes after gastrointestinal tumor surgery. However, to our knowledge, only a few studies have shown that the PNI is related to cardiovascular diseases. Therefore, we aimed to assess the association between the PNI and long-term outcomes in patients with coronary artery disease (CAD) undergoing percutaneous coronary intervention (PCI). Methods This was retrospective observational study. A total of 3561 patients with CAD after PCI were retrospectively enrolled in the CORFCHD-ZZ study from January 2013 to December 2017. The patients (3519) were divided into three groups according to PNI tertiles: the first tertile (PNI  〈  47.12, n = 1173), the second tertile (47.12 ≤ PNI  〈  51.50, n = 1185), and the third tertile (PNI ≥ 51.50, n = 1161). The mean follow-up time was 37.59 ± 22.24 months. The primary endpoint long-term mortality, including all-cause mortality (ACM) and cardiac mortality (CM).Secondary endpoints were major adverse cardiovascular events (MACEs) and major adverse cardiovascular and cerebrovascular events (MACCEs). Result In our study, the incidences of ACM in the first, second, and third tertiles were 3.8%, 1.8% and 1.4%, respectively ( P  〈  0.001). The incidences of CM occurring in the first, second, and third tertiles were 1.7%, 3.1% and 2.1%, respectively ( P  〈  0.001).There was statistically significant different in primary endpoints incidence. MACEs occurred in 139 patients (11.8%) in the first tertile, 121 patients(11.1%) in the second tertile and 123 patients(10.8%) in the third tertile( P = 0.691). MACCEs occurred in 183 patients (15.6%) in the first tertile, 174 patients(14.7%) in the second tertile and 160 patients(13.85%) in the third tertile( P = 0.463).There was no statistically significant different in secondary endpoints incidence. Kaplan–Meier analyses showed that elevated PNI was significantly related to long-term CM (log rank, P   〈  0.001) and long-term ACM (log-rank, P   〈  0.001). Cox regression analyses suggested that compared with the patients in the first tertile, the risk of ACM was decreased to 60.9% (HR = 0.609, 95% CI: 0.398–0.932, P = 0.029) in the second tertile and 40.3%(HR = 0.403, 95% CI: 0.279–0.766, P = 0.003) in the third tertile, while the risk of CM was decreased to 58.8%(HR = 0.588, 95% CI: 0.321–0.969, P = 0.038) in the second tertile and 46.6%(HR = 0.466, 95% CI: 0.250–0.870, P = 0.017) in the third tertile. Multivariate Cox regression analyses showed that the PNI was an independent predictor of long-term ACM and CM. Conclusion Our finding shown that PNI is an independent predictor in CAD patients after PCI,the higher the PNI, the less occurring adverse event. Therefore,PNI may be an new biomarker to predict long-term outcome of CAD patients after PCI.

    Abstract: Monocyte to lymphocyte ratio (MLR) has been confirmed as a novel marker of poor prognosis in patients with coronary heart disease (CAD). However, the prognosis value of MLR for patients with CAD after percutaneous coronary intervention (PCI) needs further studies. In present study, we aimed to investigate the correlation between MLR and long-term prognosis in patients with CAD after PCI. A total of 3,461 patients with CAD after PCI at the First Affiliated Hospital of Zhengzhou University were included in the analysis. According to the cutoff value of MLR, all of the patients were divided into 2 groups: the low-MLR group ( 〈 0.34, n = 2338) and the high-MLR group (≥0.34, n = 1123). Kaplan–Meier curve was performed to compare the long-term outcome. Multivariate COX regression analysis was used to assess the independent predictors for all-cause mortality, cardiac mortality and MACCEs. Multivariate COX regression analysis showed that the high MLR group had significantly increased all-cause mortality (ACM) [hazard ratio (HR) = 1.366, 95% confidence interval (CI): 1.366-3.650, p = 0.001] and cardiac mortality (CM) (HR = 2.379, 95%CI: 1.611-3,511, p 〈 0.001) compared to the low MLR group. And high MLR was also found to be highly associated with major adverse cardiovascular and cerebrovascular events (MACCEs) (HR = 1.227, 95%CI: 1.003-1.500, p = 0.047) in patients with CAD undergoing PCI. MLR was an independent predictor of ACM, CM and MACCEs in CAD patients who underwent PCI.

    Abstract: The training efficiency of the existing innovative thinking training methods are not high. In order to improve the ability of optimizing learning and searching for innovative entrepreneurial thinking, and to improve the efficiency of cultivation innovative entrepreneurial thinking, a method of optimizing and controlling the cultivation path of innovative entrepreneurial thinking based on cognitive learning theory was proposed. Used particle swarm optimization method to construct the virtual space of path learning of innovative entrepreneurial thinking cultivation, and to simulate and construct the optimized learning environment model of innovative entrepreneurial thinking cultivation. By abstracting the space coordinate of innovative entrepreneurial thinking cultivation entered the virtual world of particle swarm filter population, the map grid model of innovative entrepreneurial thinking cultivation path tracking was obtained. Through particle swarm filter evolution to found a new mode of innovative entrepreneurial thinking cultivation, combined with global stability control and particle swarm cognitive learning algorithm, to realized adaptive optimization and global optimization control of innovative entrepreneurial thinking cultivation path. Improve the learning performance of entrepreneurial thinking. The simulation results show that the learning efficiency of path optimization is higher and the cost of path evolution is lower by using this method, which improves the efficiency of innovative entrepreneurial thinking cultivation. The feasibility of the proposed method is verified.

    Abstract: In order to improve the maintenance training effect of complex mechanical product, a virtual maintenance system was developed. The system framework was proposed, and the main functional modules were elaborated. A multilevel information representation model for complex mechanical product was put forward, and the flowchart of model transformation technology was designed. Moreover, a collision detection method based on hierarchical bounding volume was proposed, and the maintainability analysis and evaluation solution based on maintenance knowledge was presented. Finally, a prototype system was developed, and the proposed system was provedto be efficient through an example of hydraulic winch.

    Abstract: Previous evidences have been proved that age, N-terminal pro-B-type natriuretic peptide (NT-proBNP), and ejection fraction are tightly associated with the long-term outcomes in patients suffered from coronary artery disease (CAD). Therefore, the present study aimed to assess the prognosis value of age, NT-proBNP, and ejection fraction (ABEF) score in CAD patients who underwent percutaneous coronary intervention (PCI). Methods Observational cohort methodology was used in this study which enrolled totally 3561 patients. And the patients were followed up regularly for 37.59 ± 22.24 months. Patients were classed into three groups based on the tertiles of ABEF sore: first tertile ( 〈 5.06, n = 831), second tertile (5.06-6.25, n = 839), and third tertile (≥ 6.25, n = 834). The ABEF score was calculated as follows: age (years)/ejection fraction (%) + NT-proBNP (NT-proBNP 〈 177pg/mL was 1, 177≤NT-proBNP≥524pg/mL was 2 and NT-proBNP  〉  524pg/mL is 3). The association between ABEF score and adverse prognosis, including all-cause death (ACD), cardiac death (CD), major adverse cardiovascular events (MACEs) and major adverse cardiac and cerebrovascular events (MACCEs), in patients who underwent PCI was analyzed. Results According to the risk category of ABEF score, the incidences of ACD ( P  〈  .001), CD ( P  〈  .001) and MACCEs ( P = .021) among the three groups showed significant differences. Multivariate Cox regression analysis suggested that the respective risks of ACD and CD were increased 3.013 folds (hazard risk [HR] = 4.013 [95% confidence interval [CI] : 1.922-8.378], P  〈  .001) and 4.922 folds ([HR] = 5.922 [95% [CI] : 2.253-15.566], P  〈  .001) in the third tertile compared with those in the first tertile. Kaplan-Meier survival analyses showed that the cumulative risks of ACD,CD and MACCEs in patients with the high ABEF score tended to increase. Conclusion The present study indicated ABEF score was a novel biomarker suitable for predicting adverse prognosis in patients after PCI, which may be used for early recognition and risk stratification.

    Abstract: In order to improve the interfacial adhesion of stainless-steel mesh reinforced poly(ether-ether-ketone) (SSM/PEEK) heating elements (HEs) used in the resistance welding process for continuous carbon fiber reinforced PEEK composites (CF/PEEK composites), three kinds of surface treatments, sandblasting, aryl diazonium grafting and silane grafting, were adopted on SSM surfaces. The functional groups were grafted successfully according to the Fourier Transform Infrared (FTIR) results and Energy Dispersive Spectrometer (EDS) results, and the surface morphologies of SSM changed significantly after different treatments. Although the tensile performances of sandblasted stainless-steel (SS) wire and sandblasted SSM were distinctly reduced, the lap shear strength (LSS) of joints with sandblasted SSM still slightly improved. Besides, the interfacial shear strength (IFSS) increased from 28 MPa for untreated SSM to 34 MPa for diazonium grafted SSM and 38 MPa for silane grafted SSM. Furthermore, the LSS results were in agreement with this trend, representing a 9% improvement for joints with diazonium grafted SSM and a 23% improvement for joints with silane grafted SSM compared with untreated CF/PEEK joints. After comparison, silane grafting was the most suitable surface modification for resistance-welded CF/PEEK joints. The interfacial bonding mechanism of CF/PEEK joints with silane grafted SSM was compared with that of untreated CF/PEEK joints, suggesting that the mechanical interlocking contributed to improved interfacial bonding.

    Abstract: Peripherally inserted central catheters (PICCs) have been placed through the peripheral veins, and the best location for the tip of the PICCs is the lower third of the superior vena cava (SVC) and cavo-atrial junction. PICCs are commonly used in intravenous administration, parenteral nutrition therapy, chemotherapy, as well as in critical care units. The success rates in venipuncture are enhanced when ultrasonographic guides are used by the bedside PICC teams. There have been few reports of PICCs placed in super elderly patients with permanent cardiac pacemakers. We share a case of two successful PICC insertions in a 97-year-old patient with bilateral pacemaker placement, that have not been reported previously.
