    Abstract: As the main detector of the Gravitational wave high-energy Electromagnetic Counterpart All-sky Monitor mission (GECAM), the calibration of the energy response and detection efficiency of the gamma-ray detector (GRD) is the main content of the ground-based calibration. This article mainly focuses on the calibration of the energy response and detection efficiency in the 8–160 keV with a refined measurement around the absorption edges of the lanthanum bromide crystal ($\rm {LaBr_3}$). The GRD performances for different crystal types, data acquisition modes, working modes, and incident positions are also analysed in detail. We show that the calibration campaign is comprehensive, the detector performance meets the flight requirements, and the calibration results generally agree with simulations as expected. The detector’s model was corrected by the ground-based calibration, which led to the establishment of the calibration data base.

    Abstract: Pseudobagrus ussuriensis is an aquaculture catfish with significant sexual dimorphism. In this study, a chromosome-level genome with a size of 741.97 Mb was assembled for female P. ussuriensis. A total of 26 chromosome-level contigs covering 97.34% of the whole-genome assembly were obtained with an N50 of 28.53 Mb and an L50 of 11. A total of 24,075 protein-coding genes were identified, with 91.54% (22,039) genes being functionally annotated. Based on the genome assembly, four chromosome evolution clusters of catfishes were identified and the formation process of P. ussuriensis chromosomes was predicted. A total of 55 sex-related quantitative trait loci (QTLs) with a phenotypic variance explained value of 100% were located on chromosome 8 (chr08). The QTLs and other previously identified sex-specific markers were located in a sex-determining region of 16.83 Mb (from 6.90 to 23.73 Mb) on chr08, which was predicted as the X chromosome. The sex-determining region comprised 554 genes, with 135 of which being differently expressed between males and females/pseudofemales, and 16 candidate sex-determining genes were screened out. The results of this study provided a useful chromosome-level genome for genetic, genomic and evolutionary studies of P. ussuriensis, and also be useful for further studies on sex-determination mechanism analysis and sex-control breeding of this fish.

    Abstract: Muscle regeneration after damage or during myopathies requires a fine cooperation between myoblast proliferation and myogenic differentiation. A growing body of evidence suggests that microRNAs play critical roles in myocyte proliferation and differentiation transcriptionally. However, the molecular mechanisms underlying the orchestration are not fully understood. Here, we showed that miR-130b is able to repress myoblast proliferation and promote myogenic differentiation via targeting Sp1 transcription factor. Importantly, overexpression of miR-130b is capable of improving the recovery of damaged muscle in a freeze injury model. Moreover, miR-130b expression is declined in the muscle of muscular dystrophy patients. Thus, these results indicated that miR-130b may play a role in skeletal muscle regeneration and myopathy progression. Together, our findings suggest that the miR-130b/Sp1 axis may serve as a potential therapeutic target for the treatment of patients with muscle damage or severe myopathies.

    Abstract: Sex differences in human emotion and related decision-making behaviors are recognized, which can be traced back early in development. However, our understanding of their underlying neurodevelopmental mechanisms remains elusive. Using developmental functional magnetic resonance imaging and computational approach, we investigated developmental sex differences in latent decision-making dynamics during negative emotion processing and related neurocognitive pathways in 243 school-aged children and 78 young adults. Behaviorally, girls exhibit higher response caution and more effective evidence accumulation, whereas boys show more impulsive response to negative facial expression stimuli. These effects parallel sex differences in emotion-related brain maturity linking to evidence accumulation, along with age-related decrease in emotional response in the basolateral amygdala and medial prefrontal cortex (MPFC) in girls and an increase in the centromedial amygdala (CMA) in boys. Moreover, girls exhibit age-related decreases in BLA–MPFC coupling linked to evidence accumulation, but boys exhibit increases in CMA–insula coupling associated with response caution. Our findings highlight the neurocomputational accounts for developmental sex differences in emotion and emotion-related behaviors and provide important implications into the neurodevelopmental mechanisms of sex differences in latent emotional decision-making dynamics. This informs the emergence of sex differences in typical and atypical neurodevelopment of children’s emotion and related functions.

    Abstract: Tigecycline is a last-resort antibiotic used to treat lethal infections caused by carbapenem-resistant Enterobacterales; however, plasmid-borne tigecycline resistance tmexCD-toprJ gene clusters can confer tigecycline resistance. The aim of the study was to identify novel subtypes and the spread of tmexCD-toprJ. Methods Five non-duplicate isolates of different species, carrying tmexCD-toprJ gene clusters or novel subtypes, were isolated from patients across China between November 2018 and June 2019. WGS was performed using Illumina and Nanopore platforms. A phylogenetic tree was constructed using a dataset of 77 sequences carrying the tmexCD-toprJ gene clusters, 72 of which were downloaded from NCBI with a blastn identity cut-off of 95%. Results We detected six different transfer units and two novel subtypes (tmexC1D1.2-toprJ1 and tmexC2D2.2-toprJ2) of the tmexCD-toprJ gene clusters. Among the six transfer units, three were mediated by IS26, while the rest were presumably mediated by Tn5393, hypothetical integrases (xerD-hp clusters-umuC-integrases-tnfxB2-tmexC2D2-toprJ2-umuC) and hypothetical units (hp-hp-hp-tnfxB2-tmexC2D2.2-toprJ2-ΔTn5393-Tn6292). Moreover, two tmexCD-toprJ-like gene clusters co-located on the same plasmid with blaNDM in five isolates. Phylogenetic analysis revealed that tmexCD-toprJ gene clusters may have originated in Pseudomonas spp., being mainly distributed in Pseudomonas spp. and Klebsiella spp. (64/77). Most tmexCD-toprJ gene clusters in Enterobacterales were located on plasmids, indicating that the gene clusters have a high inter-species transfer risk after transfer to Enterobacterales. Conclusions In summary, to the best of our knowledge, this is the first report of tmexCD-toprJ gene clusters being isolated from Enterobacter cloacae and Klebsiella oxytoca, revealing that these multiple transfer units should be further studied because of their clinical significance.

    Abstract: Inflammaging refers to low-grade, chronically activated innate immunity that has deleterious effects on healthy lifespan. However, little is known about the intrinsic signaling pathway that elicits innate immune genes during aging. Here, using Drosophila melanogaster, we profile the microRNA targetomes in young and aged animals, and reveal Dawdle, an activin-like ligand of the TGF-β pathway, as a physiological target of microRNA-252. We show that microRNA-252 cooperates with Forkhead box O, a conserved transcriptional factor implicated in aging, to repress Dawdle. Unopposed Dawdle triggers hyperactivation of innate immune genes coupled with a decline in organismal survival. Using adult muscle tissues, single-cell sequencing analysis describes that Dawdle and its downstream innate immune genes are expressed in distinct cell types, suggesting a cell nonautonomous mode of regulation. We further determine the genetic cascade by which Dawdle signaling leads to increased Kenny/IKKγ protein, which in turn activates Relish/NF-κB protein and consequentially innate immune genes. Finally, transgenic increase of microRNA-252 and Forkhead box O pathway factors in wild-type Drosophila extends lifespan and mitigates the induction of innate immune genes in aging. Together, we propose that microRNA-252 and Forkhead box O promote healthy longevity by cooperative inhibition on Dawdle-mediated inflammaging.

    Abstract: This study sought to evaluate the association of coronary angiography-derived index of microcirculatory resistance (angio-IMR) measured after primary percutaneous coronary intervention (PPCI) with the evolution of infarct pathology during 3-month follow-up after ST-segment-elevation myocardial infarction (STEMI). Methods and Results Patients with STEMI undergoing PPCI were prospectively enrolled between October 2019 and August 2021. Angio-IMR was calculated using computational flow and pressure simulation immediately after PPCI. Cardiac magnetic resonance (CMR) imaging was performed at a median of 3.6 days and 3 months. A total of 286 STEMI patients (mean age 57.8 years, 84.3% men) with both angio-IMR and CMR at baseline were included. High angio-IMR ( & gt;40 U) occurred in 84 patients (29.4%) patients. Patients with angio-IMR & gt;40 U had a higher prevalence and extent of MVO. An angio-IMR & gt;40 U was a multivariable predictor of infarct size with a three-fold higher risk of final infarct size & gt;25% (adjusted OR 3.00, 95% CI 1.23–7.32, P = 0.016). Post-procedure angio-IMR & gt;40 U significantly predicted presence (adjusted OR 5.52, 95% CI 1.65–18.51, P = 0.006) and extent (beta coefficient 0.27, 95% CI 0.01–0.53, P = 0.041) of myocardial iron at follow-up. Compared with patients with angio-IMR ≤40 U, those with angio-IMR & gt;40 U had less regression of infarct size and less resolution of myocardial iron at follow-up. Conclusions Angio-IMR immediately post-PPCI showed a significant association with the extent and evolution of infarct pathology. An angio-IMR & gt;40 U indicated extensive microvascular damage with less regression of infarct size and more persistent iron at follow-up.

    Abstract: Central venous stenosis (CVS) is a common but underestimated complication of vascular access in hemodialysis (HD) patients. The objective of this study was to compare the diagnostic efficacy of echocardiography and computed tomography angiography (CTA) for CVS in HD patients with access dysfunction. METHOD This was a retrospective study. HD patients with access dysfunction admitted to our institute between 1 April 2021 and 31 October 2021 were enrolled. Echocardiography, CTA and digital subtraction angiography (DSA) were performed to assess CVS. Receiver operator characteristic analysis was performed to compare the diagnostic efficacy of echocardiographic parameters and CTA for CVS, with DSA results as the reference standard. RESULTS A total of 167 HD patients (70 males, age 63 ± 13 years) were enrolled, among whom 99 (59%) were diagnosed with CVS based on DSA and only 9 presented with significant clinical symptoms of CVS. Among all of the echocardiographic parameters, superior vena cava (SVC) diameter showed the highest efficacy for CVS diagnosis (cut-off value ≤12 mm, specificity 70%, sensitivity 77%), which was equivalent to CTA (AUC 0.79 versus 0.81; P = 0.646) (Figure 1A). Some of the other echocardiographic parameters, including maximal and minimal right atrial volume (RAV), also showed moderated diagnostic efficacy for CVS. The specificity and sensitivity increased when echocardiography and CTA were used to diagnose brachiocephalic vein and SVC stenosis instead of total CVS (Figure 1B). Patients with CVS had a smaller SVC diameter (9.8 ± 3.5 mm versus 13.7 ± 3.3 mm;  P & lt; 0.001) and a reduced right RAV [RAV max median 20 (interquartile range 15–27) mL versus 31(24–39) mL; P & lt; 0.001; RAV min 8 (5–10) mL versus 14 (10–17) mL; P & lt; 0.001] compared with non-CVS patients. Spearman's rank correlation showed a significant negative correlation between RAV and hemodialysis vintage. CONCLUSION With equivalent diagnostic power for CVS to CTA, echocardiography has the potential to be used as a screening and monitoring tool for CVS in HD patients with access dysfunction.
