    Abstract: Porcine epidemic diarrhea virus (PEDV) is currently detected as the main pathogen causing severe diarrhea in pig farms. The phenotypic alterations induced by pathogenic infections are usually tightly linked with marked changes in epigenetic modification and gene expression. We performed global mapping of H3K4 trimethylation (H3K4me3) and transcriptomic analyses in the jejunum of PEDV-infected and healthy piglets using chromatin immunoprecipitation sequencing and RNA-seq techniques. A total of 1885 H3K4me3 peaks that are associated with 1723 genes were characterized. Moreover, 290 differentially expressed genes were identified, including 104 up-regulated and 186 down-regulated genes. Several antiviral genes including 2’-5’-oligoadenylate synthetase 1 (OAS1), 2’-5’-oligoadenylate synthetase 2 (OAS2), ephrin B2 (EFNB2), and CDC28 protein kinase regulatory subunit 1B (CKS1B) with higher H3K4me3 enrichment and expression levels in PEDV-infected samples suggested the potential roles of H3K4me3 deposition in promoting their expressions. Transcription factor annotation analysis highlighted the potential roles of two transcription factors interferon regulatory factor 8 (IRF8) and Kruppel like factor 4 (KLF4) in modulating the differential expression of genes involved in PEDV infection. The results provided novel insights into PEDV infection from the transcriptomic and epigenetic layers and revealed previously unknown and intriguing elements potentially involved in the host responses.

    Abstract: The milk fat globule ( MFG ) is one of the most representative of mammary gland tissues and can be utilized to study gene expression of lactating cows during lactation. In this study, RNA ‐seq technology was employed to detect differential expression of genes in MFG s at day 10 and day 70 after calving between two groups of cows with extremely high (H group) and low (L group) 305‐day milk yield, milk fat yield and milk protein yield. In total, 1232, 81, 429 and 178 significantly differentially expressed genes (false discovery rate q   〈  0.05) were detected between H10 and L10, H70 and L70, H10 and H70, and L10 and L70 respectively. Gene Ontology enrichment and pathway analysis revealed that these differentially expressed genes were enriched in biological processes involved in mammary gland development, protein and lipid metabolism process, signal transduction, cellular process, differentiation and immune function. Among these differentially expressed genes, 178 (H10 vs. L10), 4 (H70 vs. L70), 68 (H10 vs. H70) and 22 (L10 vs. L70) were found to be located within previously reported QTL regions for milk production traits. Based on these results, some promising candidate genes for milk production traits in dairy cattle were suggested.

    Abstract: Reproductive performance of livestock is an economically important aspect of global food production. The Chinese Meishan pig is a prolific breed, with an average of three to five more piglets per litter than European breeds; however, the genetic basis for this difference is not well understood. Results In this study, we investigated copy number variations (CNVs) of 32 Meishan pigs and 29 Duroc pigs by next-generation sequencing. A genome-wide analysis of 61 pigs revealed 12,668 copy number variable regions (CNVRs) that were further divided into three categories based on copy number (CN) of the whole population, i.e., gain ( n  = 7,638), and loss ( n  = 5,030) CNVRs. We then compared Meishan and Duroc pigs and identified 17.17 Mb of 6,387 CNVRs that only existing in Meishan pigs CNVRs that overlapped the reproduction-related gene encoding the aryl hydrocarbon receptor ( AHR ) gene. We found that normal AHR CN was more frequent than CN loss in four different pig breeds. An association analysis showed that AHR CN had a positive effect on litter size ( P   〈  0.05) and that a higher CN was associated with higher total number born ( P   〈  0.05), number born alive ( P   〈  0.05), number of weaned piglets, and birth weight. Conclusions The present study provides comprehensive CNVRs for Meishan and Duroc pigs through large-scale population resequencing. Our results provide a supplement for the high-resolution map of copy number variation in the porcine genome and valuable information for the investigation of genomic structural variation underlying traits of interest in pig. In addition, the association results provide evidence for AHR as a candidate gene associated with reproductive traits that can be used as a genetic marker in pig breeding programs.

    Abstract: • Premise of the study: Sequence tagged site (STS) primers were developed for Vicia faba , based on amplified bands by ISSR primers. The usefulness of these STS markers was validated for size polymorphism among fava bean accessions. • Methods and Results: Based on the sequences derived from intersimple sequence repeat (ISSR) amplification, 66 sequence tagged site (STS) primer pairs were developed and screened, and 20 of them were polymorphic. The polymorphism of these markers was identified in 32 fava bean germplasm from different global geographical locations. Alleles ( N a ) per locus numbered 2 to 4, and expected heterozygosity ( H E ) per locus ranged from 0.000 to 0.714. There was significant variation in H E among germplasm from different regions for individual primers. • Conclusions: These novel polymorphic STS markers may be useful and convenient for further studies of population genetics, cultivar identification, and evolution in fava bean.

    Abstract: • Premise of the study: Expressed sequence tag (EST)‐derived microsatellite simple sequence repeat (SSR) markers were developed in Vicia faba L. by screening the NCBI database. Markers were validated and explored for size polymorphism among fava bean accessions. • Methods and Results: Twenty‐one EST–SSR primer pairs were identified, and the loci characterized were size polymorphic among 32 fava bean genotypes from diverse geographical locations. The number of alleles ( N a ) per locus ranged from 2 to 9, and expected heterozygosity ( H E ) ranged from 0.0476 to 0.8304, respectively. • Conclusions: These markers will be useful to study genetic diversity, genetic mapping, and molecular breeding in fava bean.

    Abstract: In order to solve the problems of high energy consumption, high noise and pollution gas emission existing in the mechanical transmission system of loader, a research on the mechanical transmission system of loader based on hydraulic hybrid technology is proposed. The mechanical energy and heat energy are generated by the mechanical operation of the loader, which are converted into hydraulic energy and output to the drive system. According to the fast response characteristics and high power density characteristics of the hydraulic power system relative to the thermal engine, the dynamic model of the hydraulic hybrid drive system of the loader is established. The double fuzzy PID control method is used to identify and modify the unknown load and power parameters of the drive system. Through the experiment, it is concluded that the power parameter matching of the transmission system using the hydraulic hybrid technology is more optimized, the application effect is good, and the efficiency of the power transmission system can be maximized.

    Abstract: Toll-like receptor 5 (TLR5) plays an important role in immune system. In this study, we performed transcriptome analysis of the duodenum in E. coli F18-resistant and -sensitive Sutai weaned piglets and analyzed the differential expression of TLR5. The cellular localization of TLR5 was investigated, and the effect of TLR5 expression on E. coli invasion was evaluated after pig small intestinal epithelial cell lines (IPEC-J2) were stimulated by E. coli. The results showed that TLR5 expression level in duodenum and jejunum were significantly higher in E. coli F18-sensitive than in E. coli F18-resistant piglets. TLR5 protein was mainly expressed in the cytoplasm and cell membrane. The expression of genes associated with the TLR5 signaling pathway were significantly higher in TLR5-overexpressed cells than in control cells. Bacterial adhesion was higher in TLR5-overexpressed cells than in blank cells and lower in TLR5 interference than in blank cells. The core promoter region of TLR5 included two CpG islands and 16 acting elements. The methylation of the mC-6 site in the second CpG island of the promoter region had a regulatory effect on TLR5 expression. Therefore, TLR5 plays an important regulatory role on E. coli invasion. Low expression of TLR5 inhibited the immune response and decreased cell damage, which was conducive to the resistance to E. coli stimulation. In conclusion, this study preliminarily revealed the molecular mechanism of TLR5 gene regulating the resistance of piglets to Escherichia coli, and provided a new candidate gene for screening Escherichia coli resistance markers in pigs.

    Abstract: The study of the performance of dynamic hydraulic throttling under the condition of stable fluid is of great significance. The effect of a step change in pressure differences on the throttling performance of a hydraulic valve is studied. This paper studies the dynamic and static performance of a hydraulic-valve-outlet throttling-speed regulation system, builds a more accurate mathematical model, considers the linear factors of the flow of hydraulic-valve throttling, analyzes the influence of the step-load change in pressure difference on the stability of the hydraulic-valve movement speed, and constructs a nonlinear mathematical model of the speed-regulation system of the outlet throttling. A pressure sensor is used to measure the change in pressure overshoot, and the effect of a pressure-difference step change on the throttling performance of the hydraulic valve is studied under steady-fluid conditions. The theory is analyzed and verified by experiment, and the parameters of hydraulic components are modified using the dynamic-change rule of the hydraulic valve’s two-chamber pressure.
