    Kurzfassung: Aimed at investigating the differentiation of acoustic emission (AE) signals and fractal precursory characteristics between strong, weak, and no bursting liability coals under uniaxial compression, as well as improving the accuracy of rockburst monitoring and early warning by AE techniques, we experimentally studied the evolution law and differences of AE ring count rate, energy rate, and correlation dimension between different loaded bursting liability coals by the YAW4306 electric mechanical test system and CTA-1 AE monitor. Our experimental results indicated that the AE count and energy of coal samples with different bursting liabilities showed a similar evolution law of “sharp increase-calm-sharp increase” before their main rupture. The active points of AE signals emitted from coal with strong, weak, and no bursting liability appeared at about 85∼90%, 75∼78%, and 51∼55% of the peak stress, respectively. The stronger the bursting liability of coal, the shorter the duration of main rupture and postpeak failure stage, and the greater the AE energy rate in the main rupture. The AE counts of different coals had obvious fractal characteristics, and the AE correlation dimension values of strong and weak bursting liability coal samples presented the phenomenon of “fluctuating rise to a peak value-sharp drop-continuous decrease,” which can be used as a precursory information of coal failure.

    Kurzfassung: The health management of weather radar plays a key role in achieving timely and accurate weather forecasting. The current practice mainly exploits a fixed threshold prespecified for some monitoring parameters for fault detection. This causes abundant false alarms due to the evolving working environments, increasing complexity of the modern weather radar, and the ignorance of the dependencies among monitoring parameters. To address the above issues, we propose a deep learning-based health monitoring framework for weather radar. First, we develop a two-stage approach for problem formulation that address issues of fault scarcity and abundant false fault alarms in processing the databases of monitoring data, fault alarm record, and maintenance records. The temporal evolution of weather radar under healthy conditions is represented by a long short-term memory network (LSTM) model. As such, any anomaly can be identified according to the deviation between the LSTM-based prediction and the actual measurement. Then, construct a health indicator based on the portion of the occurrence of deviation beyond a user-specified threshold within a time window. The proposed framework is demonstrated by a real case study for the Chinese S-band weather radar (CINRAD-SA). The results validate the effectiveness of the proposed framework in providing early fault warnings.

    Kurzfassung: Several different methods exist for reconstructing the mandibular body and ramus defect with the use of a free vascularized fibula flap, but none have adequately addressed the long-term mechanical stability and osseointegration. The aim of this study is to compare the biomechanics of different surgical methods and to investigate the best approach for reconstructing the mandibular body and ramus defect. Five finite element models based on different reconstructive methods were simulated. Stress, strain, and displacement of connective bone sections were calculated for five models and compared. The models were printed using a 3D printer, and stiffness was measured using an electromechanical universal testing machine. The postoperative follow-up cone beam computed tomography (CBCT) was taken at different time points to analyze bone mineral density of connective bone sections. The results showed that the “double up” (DU) model was the most efficient for reconstructing a mandibular body and ramus defect by comparing the mechanical distribution of three sections under vertical and inclined loading conditions of 100 N. The stiffness detection showed that stiffness in the DU and “double down” (DD) models was higher compared with the “single up” (SU), “single down” (SD), and “distraction osteogenesis” (DO) models. We used the DU model for the surgery, and postoperative follow-up CBCT showed that bone mineral density of each fibular connective section increased gradually with time, plateauing at 12 weeks. We conclude that a free vascularized fibula flap of the DU type was the best approach for the reconstruction of the mandibular body and ramus defect. Preoperative finite element analysis and stiffness testing were shown to be very useful for maxillofacial reconstruction.

    Kurzfassung: Background/Aims . It is known that chronic low-grade inflammation contributes to the initiation and development of both diabetes and diabetic nephropathy (DN), so we designed this study to investigate the role of P2X7R and NLRP3 inflammasome in DN pathogenesis and the antagonistic effects of artificially cultivated Ophiocordyceps sinensis (ACOS). Methods . A rat model of DN caused by high-fat-diet feeding and low-dose streptozotocin injection and a mouse podocyte injury model induced by high-glucose (HG) stimulation were established, and the intervention effects of ACOS on them were observed. The biological parameters of serum and urine and the pathological manifestations of kidney tissue were examined. The expression of mRNA and protein of P2X7R and NLRP3 inflammasome (NLRP3, ASC, and caspase-1) and downstream effectors (IL-1 β and IL-18), as well as podocyte-associated molecules, was determined by real-time quantitative PCR and Western blot assay, respectively. Results . The DN rats showed to have developed insulin resistance, elevated fasting blood glucose, increased urinary protein excretion, and serum creatinine level as well as corresponding glomerular pathological alterations including podocyte damages. ACOS significantly antagonized the above changes. The experiments in vivo and in vitro both displayed that the mRNA and protein expression of P2X7R, NLRP3, ASC, caspase1 (procaspase-1 mRNA in the gene level and active caspase-1 subunit P10 in the protein level), IL-1 β , and IL-18 was significantly upregulated and the mRNA and protein expression of podocyte-associated molecules was significantly changed (downregulation of nephrin, podocin, and WT-1 expression and upregulation of desmin expression) indicating podocyte injury in the kidney tissue of DN rats and in the HG-stressed mouse podocytes, respectively. ACOS also significantly antagonized all the above changes. Conclusion . Our research work suggests that P2X7R and NLRP3 inflammasome are involved in the pathogenesis of DN, and ACOS can effectively inhibit the high expression of P2X7R and the activation of NLRP3 inflammasome, which may contribute to the therapeutic effects of Ophiocordyceps sinensis .

    Kurzfassung: Background. There are few biomarkers with an excellent predictive value for postacute myocardial infarction (MI) patients who developed heart failure (HF). This study aimed to screen candidate biomarkers to predict post-MI HF. Methods. This is a secondary analysis of a single-center cohort study including nine post-MI HF patients and eight post-MI patients who remained HF-free over a 6-month follow-up. Transcriptional profiling was analyzed using the whole blood samples collected at admission, discharge, and 1-month follow-up. We screened differentially expressed genes and identified key modules using weighted gene coexpression network analysis. We confirmed the candidate biomarkers using the developed external datasets on post-MI HF. The receiver operating characteristic curves were created to evaluate the predictive value of these candidate biomarkers. Results. A total of 6,778, 1,136, and 1,974 genes (dataset 1) were differently expressed at admission, discharge, and 1-month follow-up, respectively. The white and royal blue modules were most significantly correlated with post-MI HF (dataset 2). After overlapping dataset 1, dataset 2, and external datasets (dataset 3), we identified five candidate biomarkers, including FCGR2A, GSDMB, MIR330, MED1, and SQSTM1. When GSDMB and SQSTM1 were combined, the area under the curve achieved 1.00, 0.85, and 0.89 in admission, discharge, and 1-month follow-up, respectively. Conclusions. This study demonstrates that FCGR2A, GSDMB, MIR330, MED1, and SQSTM1 are the candidate predictive biomarker genes for post-MI HF, and the combination of GSDMB and SQSTM1 has a high predictive value.

    Kurzfassung: The acupuncture case registry study is focusing on acupuncture therapy data from patient cases. The main objective is to collect real-world data and integrate clinically meaningful outcome evaluation indicators to uncover and evaluate real-world acupuncture efficacy and safety, explore factors affecting acupuncture efficacy, and provide real-world evidence to complement RCTs. Since the International Acupuncture Case Registry data collection system’s establishment in 2017, 16 projects have been underway, including two acupuncture specialty therapies and 15 diseases. Data from 3404 patients included extensive information on the diagnosis and treatment of acupuncture and the evaluation of its efficacy. In order to serve as a guide for future studies, this article discusses the value of and rationale for establishing acupuncture case registry studies, how to distinguish them from patient registries, and crucial techniques for implementing registry studies in terms of applications, patient recruitment, costakeholder collaboration, data collection and management, study quality control, and ethics.

    Kurzfassung: In the Uxin Banner area of the Inner Mongolia Autonomous Region, mines that have experienced rock bursts have all been affected by the presence of confined water in the roof strata. Strong mine pressure appears in the working face with about 30 m wide coal pillar in this area, and the research and practice of prevention and control of rock burst by small coal pillar are carried out. Mining-related events linked to the preservation of small coal pillars in the 2-2 coal seam of this region, and studies of the phenomenon, have shown that head-on rock bursts occur during gob-side roadway driving at the edge of the drained area, making it necessary to identify the sources of pressure triggering the events, to determine the related threshold criteria, and to identify methods to reduce the occurrence and magnitude of the rock burst events. This study used the gob-side entry driving in the drained area of the 2202 auxiliary haulage roadway of a mine as a case study. The paper built a structural and computational model for overburdens (including the drained stratum) before mining disturbance, based on the theory for a continuously distributed Winkler elastic foundation. The models used and developed in our study were used to identify, quantify, and describe the changes to stress and the transfer of stress during the drainage process. On the basis of the effects of drainage stress induced by the gob-side entry driving, we outlined a method for ranking the level of rock burst hazard associated with the areas in and adjacent to the drained area. The level of rock burst hazard associated with the entry driving could be arranged in descending order as follows: parallel boundary 〉 vertical boundary 〉 undrained area 〉 drained area. Stress threshold criteria for head-on ejection were also proposed in the paper, namely, (1) the occurrence (or not) of coal ejection during loading and (2) the critical stress for the occurrence of ejection. Field monitoring indicated that microseismic events and energy are mainly concentrated in the driving region parallel to the boundary of the drained area. We concluded that head-on large-diameter borehole destressing would effectively reduce the likelihood of a rock burst occurrence.

    Kurzfassung: Purpose. Systemic lupus erythematosus (SLE) is a systemic and multifactorial autoimmune disease, and its diverse clinical manifestations affect molecular diagnosis and drug benefits. Our study was aimed at defining the SLE subtypes based on blood transcriptome data, analyzing functional patterns, and elucidating drug benefits. Methods. Three data sets were used in this paper that were collected from the Gene Expression Omnibus (GEO) database, which contained two published data sets of pediatric and adult SLE patients (GSE65391, GSE49454) and public longitudinal data (GSE72754) from a cohort of SLE patients treated with IFN-α Kinoid (IFN-K). Based on disease activity scores and gene expression data, we defined a global SLE signature and merged three clustering algorithms to develop a single-sample subtype classifier (SSC). Systematic analysis of coexpression networks based on modules revealed the molecular mechanism for each subtype. Results. We identified 92 genes as a signature of the SLE subtypes and three intrinsic subsets (“IFN-high,” “NE-high,” and “mixed”), which varied in disease severity. We speculated that IFN-high might be due to the overproduction of interferons (IFNs) caused by viral infection, leading to the formation of autoantibodies. NE-high might primarily result from bacterial and fungal infections that stimulated neutrophils (NE) to produce neutrophil extracellular traps (NETs) and induced individual autoimmune responses. The mixed type contained both of these molecular mechanisms and showed an intrinsic connection. Conclusions. Our research results indicated that identifying the molecular mechanism associated with different SLE subtypes would benefit the molecular diagnosis and stratified therapy. Moreover, repositioning of IFN-K based on subtypes also revealed an improved therapeutic effect, providing a new direction for disease treatment and drug development.
