    Abstract: Previous studies have reported a relatively low utilization of family doctor contract services (FDCS) in China, while the associated factors are unknown. The current study aimed to explore the factors associated with the utilization of FDCS, and then developed and validated a predictive model based on these identified factors. Methods We conducted a nationwide cross-sectional study using an online questionnaire, from March 2019 to April of 2019. Routinely collected variables in daily practice by family doctors were used to develop a derivation model to determine the factors associated with FDCS utilization, and then the external performance of the model was tested. Results A total of 115,717 and 49,593 participants were included in the development and validation datasets, respectively. Nearly 6.8% of the participants who signed a contract with FDCS received healthcare services from family doctors in China. Factors associated with the utilization of FDCS included age, male sex, self-reported household income, education attainment, insurance status, self-reported health status, smoking, drinking, self-reported physical activity status, chronic disease, walking distance from the nearest community center, and illness in the last 2 weeks, with an area under the receiver operating characteristic curve (AUC) of 0.660 [95% confidence interval (CI), 0.653–0.667] and good calibration. Application of this nomogram in the validation dataset also showed acceptable diagnostic value with an AUC of 0.659 (95% CI, 0.649–0.669) and good calibration. Conclusion Twelve easily obtainable factors in daily practice of family doctors were used to develop a model to predict the utilization of FDCS, with a moderate performance.

    Abstract: The burden of hepatocellular carcinoma (HCC) worldwide is increasing over time, while the underlying molecular mechanism of HCC development is still under exploration. Pseudogenes are classified as a special type of long non-coding RNAs (lncRNAs), and they played a vital role in regulating tumor-associated gene expression. Here, we report that a pseudogene peptidylprolyl isomerase A pseudogene 22 (PPIAP22) and its parental gene peptidylprolyl isomerase A (PPIA) were upregulated in HCC and were associated with the clinical outcomes of HCC. Further investigation revealed that PPIAP22 might upregulate the expression of PPIA through sponging microRNA (miR)-197-3p, behaving as competing endogenous RNA (ceRNA). PPIA could participate in the development of HCC by regulating mRNA metabolic process and tumor immunity based on the functional enrichment analysis. We also found a strong correlation between the expression levels of PPIA and the immune cell infiltration or the expression of chemokines, especially macrophage, C-C motif chemokine ligand 15 (CCL15), and C-X-C motif chemokine ligand 12 (CXCL12). Our findings demonstrate that the PPIAP22/miR-197-3p/PPIA axis plays a vital role in the progression of HCC by increasing the malignancy of tumor cells and regulating the immune cell infiltration, especially macrophage, through CCL15-CCR1 or CXCL12-CXCR4/CXCR7 pathways.

    Abstract: Integrated ecological farming combines rice growing with aquaculture, and is an effective way to improve soil productivity by increasing soil nutrient supply. However, the long-term effects of such integrated farming on phosphorus fractions and phosphorus availability of paddy soils in the Pearl River Delta (PRD) remain unknown. A four-year field experiment compared the phosphorus fractions with paddy field in rice-fish-duck system (RFD), rice-vegetable cropping system (RVS) and conventional rice system (CRS) in the PRD. SOC and phosphorus fractions were significantly influenced by cropping systems. RFD significantly increased SOC and phosphorus in the soil. Soil phosphorus was dominated by moderately labile P (40.67–49.41%). RFD also significantly increased soil microbial biomass carbon, microbial biomass phosphorus, and acid phosphatase activity (ACP) by 67.68, 46.68, and 15.87% compared to RVS, and by 134.14, 65.99, and 30.20% compared to CRS, respectively. SOC and ACP were the primary factors influencing the conversion and effectiveness of soil phosphorus. The RFD can alleviate low phosphorus activity in PRD paddy soils through the combined effect of chemical and biological process, while promoting a sustainable soil nutrient cycle within the ecosystem and guiding the sustainable development of rational soil fertilization in the PRD.

    Abstract: Pyrite is widely distributed in the Longmaxi marine shale in the Upper Yangtze area of China. Pyrite, one of the important components of shale, has an important influence on the enrichment of shale gas. However, there are currently only a few studies on this topic. Based on shale samples from drilling cores using field emission scanning electron microscopy, the pore characteristics of pyrite from the Longmaxi Formation in the Upper Yangtze area of China are studied. The results showed that the intergranular pores of pyrite and abundant organic pores in the organic matter pyrite assemblages are developed in the Longmaxi Formation shale in the study area. Most pyrite framboid pores have triangular or irregular quadrilateral shapes, with pore diameters ranging from 0 to 240 nm. Pores with diameters of 80–240 nm are the major reservoir spaces for shale gas within the pyrite framboids. The average contribution rate of pyrite framboids to shale pores reaches 3.21%, and the highest contribution is 5.66%, indicating that the pyrite pores may have a favorable contribution to the shale reservoir pore system, but the contribution degree is low.

    Abstract: Few studies have incorporated the tools of stable isotopes and optical properties to study the biogeochemical process of organic matter (OM), including particulate organic matter (POM) and dissolved organic matter (DOM), which prevents our comprehension of the interactions between POM and DOM in the marine environment. In this study, the origin, distribution, and fate of POM and DOM in Tieshangang Bay, a weak dynamic bay were investigated by measuring δ 13 C and δ 15 N of POM, dissolved organic carbon (DOC), and absorption and fluorescence of DOM (CDOM and FDOM). In the upper bay, POM source was mainly originated from terrestrial discharge, whereas the high fluorescence index (FI) indicated that external aquatic organism activity dominated DOM sources. In contrast, in the outer bay, the slightly increased δ 13 C and enriched δ 15 N of POM and enhanced fluorescence intensities of protein-like DOM components indicated the considerable contributions of the in situ biological activities to OM. A net addition of DOC and optical components of DOM occurred in both the upper and outer bays. The decomposition of terrestrial POM was responsible for the addition of DOM due to the weak dynamics in the upper bay, whereas the enhanced primary production and the strong decomposition of freshly produced POM jointly contributed to the addition of DOM in the outer bay. Our study suggests that hydrodynamics regulate the biogeochemistry and interactions of POM and DOM in the weak dynamic bay.

    Abstract: Non-traumatic osteonecrosis of the femoral head (NONFH) remains a common refractory disease with poorly understood pathogenesis. Macrophage M1/M2 imbalance and chronic inflammatory microenvironment have been suggested to be closely related to osteonecrosis. Here we describe direct visual evidence for the involvement of dynamic changes in macrophages and the chronic inflammatory microenvironment in human NONFH. Osteonecrosis induces inflammatory responses and macrophage enrichment in the reparative area, and the number of inflammatory cells and macrophages falls during progressive-to end-stage NONFH. Multiplex immunohistochemistry demonstrated that macrophage M1/M2 ratio increased from 3 to 10 during progressive-to end-stage. During the progressive-stage, new blood vessels formed in the reparative area, M2 macrophages accumulated in perivascular (M1/M2 ratio ∼0.05), while M1 macrophages were enriched in avascular areas (M1/M2 ratio ∼12). Furthermore, inflammatory cytokines were detected in synovial fluid and plasma using cytometric bead arrays. Interleukin (IL)-6 and IL-1β were persistently enriched in synovial fluid compared to plasma in patients with NONFH, and this difference was confirmed by immunohistochemistry staining. However, only IL-6 levels in plasma were higher in patients with progressive-stage NONFH than in osteoarthritis. Moreover, fibrosis tissues were observed in the necrotic area in progressive-stage and end-stage NONFH based on Sirius Red staining. Together, these findings indicate that macrophage M1/M2 imbalance facilitates the progression of NONFH, a chronic inflammatory disease characterized by chronic inflammation, osteonecrosis and tissue fibrosis in the local lesion. Inhibiting inflammation, promoting the resolution of inflammation, switching macrophages to an M2 phenotype, or inhibiting their adoption of an M1 phenotype may be useful therapeutic strategies against NONFH.
