    Abstract: Previous study showed that low protein diet-fed pigs are characterized by lower histidine concentration in the serum and muscle, suggesting that histidine may involve in protein-restricted response. Thus, the current study mainly investigated the effects of dietary histidine on growth performance, blood biochemical parameters and amino acids, intestinal morphology, and microbiota communities in low protein diet-challenged-piglets. The results showed that protein restriction inhibited growth performance, blood biochemical parameters and amino acids, and gut microbiota but had little effect on intestinal morphology. Dietary supplementation with histidine markedly enhanced serum histidine level and restored tryptophan concentration in low protein diet-fed piglets, while growth performance and intestinal morphology were not markedly altered in histidine-treated piglets. In addition, histidine exposure failed to affect bacterial diversity (observed species, Shannon, Simpson, Chao1, ACE, and phylogenetic diversity), but histidine-treated piglets exhibited higher abundances of Butyrivibrio and Bacteroides compared with the control and protein-restricted piglets. In conclusion, dietary histidine in low protein diet enhanced histidine concentration and affected gut microbiota (Butyrivibrio and Bacteroides) but failed to improve growth performance and intestinal morphology.

    Abstract: Wen-Dan Decoction (WDD), a formula of traditional Chinese medicine, has been clinically used for treating insomnia for approximately 800 years. However, the therapeutic mechanisms of WDD remain unclear. Orexin-A plays a key role in the sleep-wake cycle, while leptin function is opposite to orexin-A. Thus, orexin-A and leptin may be important factors in sleep disorders. In this study, 48 rats were divided into control, model, WDD-treated, and diazepam-treated groups. The model of insomnia was produced by sleep deprivation (SD) for 14 days. The expressions of orexin-A, leptin, and their receptors in blood serum, prefrontal cortex, and hypothalamus were detected by enzyme-linked immunosorbent assay, immunohistochemistry, and real time PCR. Open field tests showed that SD increased both crossing movement (Cm) and rearing-movement (Rm) times. Orexin-A and leptin levels in blood serum increased after SD but decreased in brain compared to the control group. mRNA expressions of orexin receptor 1 and leptin receptor after SD were decreased in the prefrontal cortex but were increased in hypothalamus. WDD treatment normalized the behavior and upregulated orexin-A, leptin, orexin receptor 1 and leptin receptor in brain. The findings suggest that WDD treatment may regulate SD-induced negative emotions by regulating orexin-A and leptin expression.

    Abstract: The traditional Chinese medicine (TCM) decoction Si-Ni-San (SNS) has been utilised for millennia to improve physiological coordination of the functions of the liver and spleen, which are regarded as the main pathological organs of central fatigue in TCM. This study evaluates the effect of a modified SNS (MSNS) formula on central fatigue in rats and explores molecular changes associated with hippocampal mitochondrial biogenesis. Central fatigue was induced through a 21-day sleep deprivation protocol. We assessed MSNS’s effects on behaviour, blood and liver biomarkers, and mitochondrial ultrastructure. We found that MSNS could reverse various signs of central fatigue such as its effects on hippocampal gene and protein expression levels of sirtuin 1 (SIRT1), peroxisome proliferator-activated receptor gamma coactivator-1 α (PGC-1 α ), and nuclear respiratory factor 1 (NRF1). We also observed evidence of MSNS decreasing central fatigue, such as decreasing creatine kinase activity, decreasing levels of malondialdehyde and blood urea nitrogen, increasing lactate dehydrogenase and superoxide dismutase activities, increasing mitochondrial DNA copy number, and reversing mitochondrial ultrastructure changes. These findings suggest that MSNS can ameliorate central fatigue and that its molecular mechanism involves mitochondrial biogenesis enhancement mediated by hippocampal SIRT1, PGC-1 α , and NRF1.

    Abstract: Deconvolution-related methods are the mainstream choice when it comes to enhancing the pulse impact of bearing fault and reducing noise interference. Kurtogram algorithm is used to optimize the minimum generalized Lp/Lq deconvolution to improve the nonconvexity of other optimization criteria. However, it has low computational efficiency and poor diagnostic accuracy under strong background noise. The paper proposes an optimized method using protrugram algorithm that combines fast iterative filter decomposition (FIFD) with minimum generalized Lp/Lq deconvolution (OMGD) for the 1.5-dimension Teager energy spectrum demodulation. Here is the specific process of the application: Fast iterative filtering (FIF) was used to reduce noise interference before using the maximum kurtosis to obtain the center frequency and frequency band and optimize the filter design, which was for the MGD initialization operation to prevent the result from falling into the local optimal solution and check the interference of impulse noise to a certain extent. The 1.5-dimension Teager energy spectrum was then used for demodulation analysis to extract small fault features of rolling bearings. The verification of simulation signals and actual data showed that this method was better in terms of extraction effect and efficiency than the use of fast kurtogram algorithm to optimize minimum generalized Lp/Lq deconvolution when it comes to extracting microfault features with high interference of background noise.

    Abstract: Oxymatrine (OMT), an alkaloid derived from the traditional Chinese medicine herb Sophora flavescens Aiton, has been shown to exhibit anticancer properties on various types of cancer cells. In this study, we investigate the anticancer properties of OMT on human glioblastoma (GBM) cells and evaluate their underlying mechanisms. MTT assays were performed and demonstrated that OMT significantly inhibits the proliferation of GBM cells. Flow cytometry suggested that OMT at a concentration of 10 −5  M may induce apoptosis in U251 and A172 cells. Western blot analyses demonstrated a significant increase in the expression of Bax and caspase-3 and a significant decrease in expression of Bcl-2 in both U251 and A172 cells. Additionally, OMT was found by transwell and high-content screening assays to decrease the migratory ability of the evaluated GBM cells. These findings suggest that the antitumor effects of OMT may be the result of inhibition of cell proliferation and migration and the induction of apoptosis by regulating the expression of apoptosis-associated proteins. OMT may represent a novel anticancer therapy for the treatment of GBM.

    Abstract: The fault signals of planetary gears are nonstationary and nonlinear signals. It is difficult to extract weak fault features under strong background noise. This paper adopts a new filtering method, fractional Wavelet transform (FRWT). Compared with the traditional fractional Fourier transform (FRFT), it can improve the effect of noise reduction. This paper adopts a planetary gear fault diagnosis method combining fractional wavelet transform (FRWT) and two-dimensional convolutional neural network (2D-CNN). Firstly, several intrinsic mode component functions (IMFs) are obtained from the original vibration signal by AFSA-VMD decomposition, and the two components with the largest correlation coefficient are selected for signal reconstruction. Then, the reconstructed signal is filtered in fractional wavelet domain. By analyzing the wavelet energy entropy of the filtered signal, a two-dimensional normalized energy characteristic matrix is constructed and the two-dimensional features are input into the two-dimensional convolution neural network model for training. The simulation results show that the training effect of this method is better than that of FRFT-2D-CNN. Through the verification of the test set, we can know that the fault diagnosis of planetary gears can be realized accurately based on FRWT and 2D-CNN.
