    Abstract: Background Information . Stem cells from apical papilla (SCAPs) are a potent candidate for the apexogenesis/apexification due to their multiple differentiation capacity. During the orthodontic treatment of developing teeth, SCAPs in vivo are usually subjected to the cyclic stress induced by compression forces. However, it remains unclear whether mechanical stress can affect the proliferation and differentiation of human SCAPs. Results . Human SCAPs were isolated and stimulated by 200 g mechanical stimuli for 30 min and their proliferation and differentiation capacity were evaluated in vitro at different time points. MTT and FCM results demonstrated that cell proliferation was enhanced, while TEM findings showed the morphological and ultrastructural changes in stress-treated SCAPs. ALP activity and mineralization capacity of stress-treated SCAPs were upregulated . In the meantime, higher odontogenic and osteogenic differentiation were found in stress-treated SCAPs by real-time RT-PCR and Western blot, as indicated by the expression of related markers at both mRNA and protein levels. Moreover, the protein expressions of pJNK and pERK MAPK pathways were upregulated. Conclusion . Together, these findings suggest that mechanical stress is an important factor affecting the proliferation and differentiation of SCAPs via the activation of ERK and JNK signaling pathway.

    Abstract: The traditional Chinese medicine (TCM) decoction Si-Ni-San (SNS) has been utilised for millennia to improve physiological coordination of the functions of the liver and spleen, which are regarded as the main pathological organs of central fatigue in TCM. This study evaluates the effect of a modified SNS (MSNS) formula on central fatigue in rats and explores molecular changes associated with hippocampal mitochondrial biogenesis. Central fatigue was induced through a 21-day sleep deprivation protocol. We assessed MSNS’s effects on behaviour, blood and liver biomarkers, and mitochondrial ultrastructure. We found that MSNS could reverse various signs of central fatigue such as its effects on hippocampal gene and protein expression levels of sirtuin 1 (SIRT1), peroxisome proliferator-activated receptor gamma coactivator-1 α (PGC-1 α ), and nuclear respiratory factor 1 (NRF1). We also observed evidence of MSNS decreasing central fatigue, such as decreasing creatine kinase activity, decreasing levels of malondialdehyde and blood urea nitrogen, increasing lactate dehydrogenase and superoxide dismutase activities, increasing mitochondrial DNA copy number, and reversing mitochondrial ultrastructure changes. These findings suggest that MSNS can ameliorate central fatigue and that its molecular mechanism involves mitochondrial biogenesis enhancement mediated by hippocampal SIRT1, PGC-1 α , and NRF1.

    Abstract: Aim . The purpose of this study was to investigate the relationship between upper limbs’ three functional partitions and the golden curve. Materials and Methods . We measured 30 subjects’ right or left upper limb data and investigate the relationship between them and the golden curve by use of SPSS version 20.0 statistical software (SPSS, Inc., Chicago, Illinois), one-sample t -test. Results . There are four points on human’s upper limbs which have no difference with the four points on the golden curve. And there is one point of which the difference is obvious. But we still could draw the conclusion that human upper limbs are accordant with the golden curve. Conclusion . Human upper limbs are accordant with the golden curve.

    Abstract: Encounter risk prediction is critical for safe ship navigation, especially in congested waters, where ships sail very near to each other during various encounter situations. Prior studies on the risk of ship collisions were unable to address the uncertainty of the encounter process when ignoring the complex motions constituting the dynamic ship encounter behavior, which may seriously affect the risk prediction performance. To fill this gap, a novel AIS data-driven approach is proposed for ship encounter risk prediction by modeling intership behavior patterns. In particular, multidimensional features of intership behaviors are extracted from the AIS trace data to capture spatial dependencies between encountering ships. Then, the challenging task of risk prediction is to discover the complex and uncertain relationship between intership behaviors and future collision risk. To address this issue, we propose a deep learning framework. To represent the temporal dynamics of the encounter process, we use the sliding window technique to generate the sequences of behavioral features. The collision risk level at a future time is taken as the class label of the sequence. Then, the long short-term memory network, which has a strong ability to model temporal dependency and complex patterns, is extended to establish the relationship. The benefit of our approach is that it transforms the complex problem for risk prediction into a time series classification task, which makes collision risk prediction reliable and easier to implement. Experiments were conducted on a set of naturalistic data from various encounter scenarios in the South Channel of the Yangtze River Estuary. The results show that the proposed data-driven approach can predict future collision risk with high accuracy and efficiency. The approach is expected to be applied for the early prediction of encountering ships and as decision support to improve navigation safety.

    Abstract: Lung cancer is still the leading cause of cancer-related death worldwide. Of lung cancer, lung adenocarcinoma (LUAD) is the most common subtype. Most patients with LUAD would develop into metastasis, which limits the available treatment. Targeted therapy and immunotherapy provided options for those advanced patients. But they also broached up challenges to identify the appropriate patients. This study aims to reveal the landscapes of genomic mutations in primary and metastatic LUAD and their actionability. This study enrolled 636 patients with LUAD, of whom 85 and 551 were from patients with and without metastasis, respectively. Next-generation sequencing technology was used to retrieve their genomic information. Genomic mutations including short nucleotide variation, long variation, copy number variations, and fusions were called. The corresponding actionability was revealed. A comparison of genomic mutations and actionability between primary and metastatic LUAD was performed. In primary tumors, BRCA2 and FAT3 were significantly mutated in older patients; while in metastases, ALK and NOTCH2 were significantly mutated in younger patients. Primary tumors in male patients were significantly mutated in LRP1B and KRAS. Compared to primary tumors, metastases harbored less short nucleotide variations but more copy number variations and fusions. In metastases, chromosome 1 and chromosome 9 had less short nucleotide variations and more CNV than in primary tumors. Genomic variations of activated dendritic cells were more frequently mutated in metastases. EGFR genomic variations were negatively associated with PD-L1 and TMB. Patients with EGFR inhibitor treatment tend to have lower PD-L1 expression. The revealed discrepancy between primary and metastatic lung cancer could help guide the treatment strategies and the development of novel drugs.

    Abstract: Star-like nano-ZrO 2 has been synthesized using Zr(NO 3 ) 4 ·5H 2 O as zirconium source by a hydrothermal process without any template and surfactant. The structure of the as-prepared ZrO 2 powder was investigated by multiple advanced analytical methods. The results showed that CH 3 COO − and NO 3 − had great effects on the formation of star-like ZrO 2 nanostructures. The as-prepared ZrO 2 had a superior catalytic activity, and the reason for it was analyzed by UV-Vis diffuse reflectance spectroscopy. The effect of raw material ratios on the photocatalytic property of ZrO 2 was studied. The synthesized ZrO 2 showed a narrow bandgap (3.50–3.85 eV) and an excellent photocatalytic activity, and the degradation of RhB was up to nearly 100% in 30 min with this photocatalyst.

    Abstract: Oral squamous cell carcinoma (OSCC) is the most common head and neck cancer with a poor prognosis. Therefore, it is crucial to explore molecular prognostic biomarkers for OSCC. ZEB1 (also known as δEF1) is a member of the zinc finger E-box binding protein family of transcription factors involved in various biological processes, including tumorigenesis, progression, and metastasis. Recent evidence suggests that ZEB1 has a role in the tumorigenicity of oral epithelial cells, although its mode of action needs to be investigated further. To better understand the relationship between ZEB1 and OSCC, we transfected the ZEB1-overexpressing oral squamous cell lines SCC9 and SCC25 with lentivirus and then extracted RNA from the cells for gene expression analysis. Furthermore, the GSE30784 dataset was downloaded from the Gene Expression Omnibus (GEO) database to identify potential biomarkers of OSCC and to assess the potential mechanisms. The criteria for identification of their DEGs were |logFC|  〉  1 and P   〈  0.05. Gene Ontology (GO) and Kyoto Encyclopaedia of Genes and Genomes (KEGG) analyses were also carried out. Integrating the data from the PPI network and survival analysis identified that ZEB1 might be an independent prognostic biomarker in OSCC. In conclusion, integrated bioinformatics and microarray analysis identified the critical gene ZEB1 linked to the overall survival (OS) of patients with OSCC. ZEB1 could be applied as a prognostic biomarker to forecast the survival of patients with OSCC and might indicate innovative therapeutic indicators for OSCC.

    Abstract: Longitudinal liquid sloshing in partially filled clear-bore tanks causes extensive degradation of tankers braking performance. To reduce the negative effect of longitudinal liquid sloshing on tankers, three kinds of transverse baffles were designed, namely, the conventional baffle, the circular baffle, and the staggered baffle. Each kind of baffle took several forms to investigate the impact of baffle installation angle, the sizes of holes pierced on the baffle, and their arrangement on the antisloshing effect. FLUENT software was used to simulate liquid sloshing in tanks equipped with different kinds of transverse baffles and subject to constant braking deceleration. A time-series analysis of the forces act on tank walls and transverse baffles was carried out. It was drawn that the baffle shape and its installation angle have great impact on the antisloshing effect of baffles. The study on the antisloshing effect of different transverse baffles is of great significance for tank vehicle driving and braking safety, as well as for the design of optimal transverse baffles.

    Abstract: Background . Identifying diagnostic and prognostic biomarkers that could be targeted in the therapy of pancreatic cancer is essential. Objective . Investigations were conducted with respect to plasma miRNA (miR-21, miR-210, miR-155, miR-196a, miR-20a, and miR-25) expression and clinicopathologic factors to evaluate the prognostic value of miRNAs in pancreatic ductal adenocarcinoma (PDAC). Methods . Plasma miRNAs were detected by real-time quantitative PCR, and the association with clinicopathologic factors was subsequently performed by univariate and multivariate analyses. Results . Six miRNAs expressed significantly higher in PDAC patients than in normal individuals were identified. Receiver operating characteristic (ROC) curves were constructed. It was evident that miRNA expression associated with PDAC, lymph node metastasis, serosal infiltration, and comprehensive therapy reached significance for overall survival. High miR-196a expression was associated with poor survival ( P = 0.001 ), whereas high miR-210 expression was significantly associated with improved survival ( P = 0.003 ). Multivariate survival analysis indicated that the miR-210 and miR-196a expression signature, lymph node metastasis, and comprehensive therapy were independent factors affecting overall survival. Conclusions . MiRNA expression profile is distinctive in PDAC. Aberrant expression of certain miRNAs was remarkably involved in shaping the overall survival time, which include miR-196a overexpression and decreased miR-210 expression.
