    Abstract: Astragalus polysaccharides (APS), the main effective component of Astragalus membranaceus , can inhibit tumor growth, but the underlying mechanisms remain unclear. Previous studies have suggested that APS can regulate the gut microenvironment, including the gut microbiota and fecal metabolites. In this work, our results showed that APS could control tumor growth in melanoma‐bearing mice. It could reduce the number of myeloid‐derived suppressor cells (MDSC), as well as the expression of MDSC‐related molecule Arg‐1 and cytokines IL‐10 and TGF‐β, so that CD8 + T cells could kill tumor cells more effectively. However, while APS were administered with an antibiotic cocktail (ABX), MDSC could not be reduced, and the growth rate of tumors was accelerated. Consistent with the changes in MDSC, the serum levels of IL‐6 and IL‐1β were lowest in the APS group. Meanwhile, we found that fecal suspension from mice in the APS group could also reduce the number of MDSC in tumor tissues. These results revealed that APS regulated the immune function in tumor‐bearing mice through remodeling the gut microbiota. Next, we focused on the results of 16S rRNA, which showed that APS significantly regulated most microorganisms, such as Bifidobacterium pseudolongum , Lactobacillus johnsonii and Lactobacillus . According to the Spearman analysis, the changes in abundance of these microorganisms were related to the increase of metabolites like glutamate and creatine, which could control tumor growth. The present study demonstrates that APS attenuate the immunosuppressive activity of MDSC in melanoma‐bearing mice by remodeling the gut microbiota and fecal metabolites. Our findings reveal the therapeutic potential of APS to control tumor growth.

    Abstract: Promoting the use of recycled water is an effective way to solve the problem of urban water shortage. In order to promote the utilization of recycled water, this study identified the influential factors determining the differences between willingness and behavior to use recycled water for toilet flushing. Binary logistic regression models of willingness and behavior were analyzed and the data came from 1195 Beijing residents in communities where recycled water was available for toilet flushing. The results are as follows: First, the proportion of those willing to use recycled water (92%) was significantly higher than those who actually did so (35.2%); thus, higher willingness to use recycled water did not necessarily lead to higher using behavior. Second, different factors influenced willingness and behavior, with the willingness mainly influenced by cognitive and attitudinal factors, and the behavior dependent on external environmental factors such as the convenience of installation and promotion measures. Third, the convenience of the installation of recycled water facilities is the most important factor influencing both willingness and behavior, and inconvenient facilities are the main factor hindering the use of recycled water.

    Abstract: Purpose: To investigate the effect of adipose-derived stem cells (ADSCs) on the survival rate of long random pattern skin flaps in rats and the likely underlying mechanism. Methods: Twenty (20) SPF-grade SD rats were randomly divided into control group and study group, with 10 rats in each group. Rats in the control group were given phosphate buffered saline (PBS), while rats in the study group received ADSCs. The survival rate of the skin flaps was compared between the 2 groups on day 7 after operation, while the levels of indices related to oxidative stress and inflammatory response were compared on the 7th and 14th days after operation. Results: Necrosis in the study group was milder (p 〈 0.05). On the 7th post-operation day, the survival rate of rat skin flap in the study group was significantly higher than that in the control group (p 〈 0.05), but the MDA level in the study group was lower than that of the control group (p 〈 0.05). On the 14th day after the operation, the MDA level was decreased in the two groups (p 〈 0.05), with a lower MDA level in the study group than in the control group (p 〈 0.05). Conclusion: ADSCs inhibit inflammatory response and reduces the level of oxidative stress after long random pattern skin flap transplantation in rats, thereby enhancing blood supply to the flap transplantation area and improving its survival rate. Thus, ADSCs have a potential clinical application in enhancing blood supply in skin transplantation.

    Abstract: TNF receptor-associated factors (TRAFs) are signaling adaptor proteins that play a crucial role in regulating cellular receptors’ signaling transduction to downstream pathways and exert multifaceted roles in regulating signaling pathways, cell survival, and carcinogenesis. The 13-cis-retinoic acid (RA), an active metabolite of vitamin A, exhibits anti-cancer properties, but the development of retinoic acid resistance poses a challenge in clinical application. This study aimed to investigate the relationship between TRAFs and retinoic acid sensitivity in various cancers. Here, we revealed that TRAFs’ expression varied significantly across The Cancer Genome Atlas (TCGA) cancer cohorts and human cancer cell lines. Additionally, inhibiting TRAF4, TRAF5, or TRAF6 improved retinoic acid sensitivity and reduced colony formation in ovarian cancer and melanoma cells. Mechanistically, knocking down TRAF4, TRAF5, or TRAF6 in retinoic acid-treated cancer cell lines increased the levels of procaspase 9 and induced cell apoptosis. Further in vivo studies using the SK-OV-3 and MeWo xenograft models confirmed the anti-tumor effects of TRAF knockdown combined with retinoic acid treatment. These findings support that combination therapy with retinoic acid and TRAF silencing may offer significant therapeutic advantages in treating melanoma and ovarian cancers.

    Abstract: Worldwide water scarcity has prompted governments to develop alternative water resources, and public acceptance plays a critical role in the implementation of recycled water projects. The aim of this study was to explore how public acceptance can be affected by people’s emotional responses to recycled water. In study 1, the general population was divided into four groups, clustering analysis with the intensity of various emotions was conducted, and the association between emotions and acceptance was explored. In study 2, the emotions, people’s acceptance, and “objects of care” of water treatment experts were compared with those of members of the general population for further study. We found that the objects of care shortened the psychological distance between individuals and recycled water, which led people to generate specific emotions to recycled water. In a broader context, the study suggested that various emotions motivate people’s behavior to accept or oppose the use of recycled water.

    Abstract: As neoadjuvant chemotherapy is widely used in breast cancer patients, the lymph node ratio has not been fully validated as a prognostic indicator of breast cancer received neoadjuvant chemotherapy. This study was conducted to investigate the prognostic value of lymph node ratio in breast cancer patients received neoadjuvant chemotherapy. Methods Systematic searches were performed in the PubMed, Embase, and Cochrane Library databases until 15 December 2021 for studies on the association between lymph node ratio and the prognosis of breast cancer after neoadjuvant chemotherapy. Overall survival and disease-free survival were used as outcome events, and hazard ratio was chosen as the parameter to evaluate the correlation. The dose-response relationship was assessed by restricted cubic splines. In the subgroup analyses, which were used to explore potential heterogeneity among the included studies according to study region and sample size. Sensitivity analysis was performed to assess the stability of individual studies, and publication bias was determined with funnel plots, Begg’s test, and Egger & amp;apos;s test. All statistical analyses were performed using Stata 15.1. Results A total of 12 studies with 4,864 patients were included in this meta-analysis. In this study, high lymph node ratio was significantly associated with decreased overall survival (HR: 4.74; 95%CI: 3.36–6.67; P  & lt; 0.001) and disease-free survival (HR: 4.77; 95%CI: 3.69–6.17; P  & lt; 0.001). Moreover, the dose-response meta-analysis showed a linear association between higher lymph node ratio and shorter overall survival and disease-free survival in breast cancer patients after neoadjuvant chemotherapy. Conclusions The meta-analysis suggested that high lymph node ratio was significantly associated with short overall survival and disease-free survival in breast cancer patients after neoadjuvant chemotherapy. Therefore, lymph node ratio is an independent predictive factor for the prognosis of breast cancer patients after neoadjuvant chemotherapy, which may better refine the cancer staging system.

    Abstract: Activated microglia is considered to be major mediators of the neuroinflammatory environment in demyelinating diseases of the central nervous system (CNS). Activated microglia are mainly polarized into M1 type, which plays a role in promoting inflammation and demyelinating. However, the proportion of microglia polarized into M2 type is relatively low, which cannot fully play the role of anti-inflammatory and resistance to demyelinating. Our previous study found that Astragalus polysaccharides (APS) has an immunomodulatory effect and can inhibit neuroinflammation and demyelination in experimental autoimmune encephalomyelitis (EAE), which is a classic animal model of CNS demyelinating disease. In this study, we found that APS was effective in treating EAE mice. It restored microglia balance by inhibiting the polarization of microglia to M1-like phenotype and promoting the polarization of microglia to M2-like phenotype in vivo and in vitro. miR-155 is a key factor in regulating microglia polarization. We found that APS could inhibit the expression level of miR-155 in vivo and in vitro. Furthermore, we performed transfection overexpression and blocking experiments. The results showed that miR-155 mediated the polarization of microglia M1/M2 phenotype, while the selective inhibitor of miR-155 attenuated the inhibition of APS on microglia M1 phenotype and eliminated the promotion of APS on microglia M2 phenotype. Microglia can secrete IL-1α, TNF-α, and C1q after polarizing into M1 type and induce the activation of A1 neurotoxic astrocytes, further aggravating neuroinflammation and demyelination. APS reduced the secretion of IL-1α, TNF-α, and C1q by activated microglia, thus inhibited the formation of A1 neurotoxic astrocytes. In summary, our study suggests that APS regulates the polarization of microglia from M1 to M2 phenotype by inhibiting the miR-155, reduces the secretion of inflammatory factors, and inhibits the activation of neurotoxic astrocytes, thus effectively treating EAE.
