    Beschreibung: Pharmaceutically active compounds (PhACs) released into the environment have an adverse impact on the soil and water ecosystem as well as human health. Sorption of PhACs by soils and its potential modification through introduced DOM in the applied animal manure or treated wastewater (TWW) determines the mobility and environmental relevance of PhACs. Sulfadiazine, caffeine and atenolol were selected as target PhACs to investigate their sorption behaviors by five selected arable soils in the absence and presence of pig manure DOM. Sulfadiazine was least sorbed, followed by caffeine and atenolol according to the Freundlich sorption isotherm fit (soil average Kf [μg〈sup〉(1−n)〈/sup〉 mL〈sup〉n〈/sup〉 g〈sup〉−1〈/sup〉] 4.07, 9.06, 18.92, respectively). The addition of manure DOM (31.34 mg C L〈sup〉−1〈/sup〉) decreased the sorption of sulfadiazine and especially of caffeine and atenolol (average K〈sub〉f〈/sub〉 3.04, 6.17, 5.79, respectively). Freundlich sorption isotherms of the PhACs became more nonlinear in the presence of manure DOM (Freundlich exponent n changed from 0.74–1.40 to 0.62–1.12), implying more heterogeneous sorption of PhACs in soil–DOM binary systems. Sorption competition of DOM molecules with sulfadiazine and caffeine mostly contributed to their decreased soil sorption when DOM was present. In contrast, the formation of DOM–atenolol associates in the solution phase caused the largely decreased soil sorption of atenolol in the presence of DOM. It is suggested that DOM concentration (e.g., ≥ 60 mg C L〈sup〉−1〈/sup〉) and its interaction with PhACs should be taken into consideration when assessing the environmental impact of land application of animal manure or irrigation with TWW.

    Beschreibung: Background: EphA5 is a member of the Eph/ephrin family and plays a critical role in the regulation of carcinogenesis. A significant reduction of EphA5 transcripts in high-grade prostate cancer tissue was shown using a transcriptomic analysis, compared to the low-grade prostate cancer tissue. As less is known about the mechanism of EphA5 downregulation and the function of EphA5, here we investigated the expression and an epigenetic change of EphA5 in prostate cancer and determined if these findings were correlated with clinicopathologic characteristics of prostate cancer. Methods: Seven prostate cell lines (RWPE-1, LNCap, LNCap-LN3, CWR22rv-1, PC-3, PC-3M-LN4, and DU145),thirty-nine BPH, twenty-two primary prostate carcinomas, twenty-three paired noncancerous and cancerous prostate tissues were examined via qRT-PCR, methylation-specific PCR, bisulfite sequencing, immunohistochemistry and western blotting. The role of EphA5 in prostate cancer cell migration and invasion was examined by wound healing and transwell assay. Results: Downregulation or loss of EphA5 mRNA or protein expression was detected in 28 of 45 (62.2%) prostate carcinomas, 2 of 39 (5.1%) hyperplasias, and all 6 prostate cancer cell lines. Methylation of the EphA5 promoter region was present in 32 of 45 (71.1%) carcinoma samples, 3 of 39 (7.7%) hyperplasias, and the 6 prostate cancer cell lines. Among 23 paired prostate carcinoma tissues, 16 tumor samples exhibited the hypermethylation of EphA5, and 15 of these 16 specimens (93.8%) shown the downregulation of EphA5 expression than that of their respectively matched noncancerous samples. Immunostaining analysis demonstrated that the EphA5 protein was absent or down-regulated in 10 of 13 (76.9%) available carcinoma samples, and 8 of these 10 samples (80.0%) exhibited hypermethylation. The frequency of EphA5 methylation was higher in cancer patients with an elevated Gleason score or T3-T4 staging. Following the treatment of 6 prostate cancer cell lines with 5-aza-2?-deoxycytidine, the levels of EphA5 mRNA were significantly increased. Prostate cancer cells invasion and migration were significantly suppressed by ectopic expression of EphA5 in vitro. Conclusion: Our study provides evidence that EphA5 is a potential target for epigenetic silencing in primary prostate cancer and is a potentially valuable prognosis predictor and thereapeutic marker for prostate cancer.

    Beschreibung: This study assessed the feasibility of community-based comprehensive intervention on Type 2 diabetes mellitus (T2DM) on a large population in China. An intervention study was conducted on 7200 T2DM patients within one year and consisted of six lectures on health issues, and four times face-to-face lifestyle counseling delivered by general health practitioners, at local primary health centers (PHCs). A “knowledge, attitude and practice” (KAP) survey and fasting plasma glucose (FPG) measurement were conducted at baseline and after the intervention, respectively. A total of 6586 T2DM patients completed the intervention. After one year intervention, patients’ KAP level improved significantly (p 〈 0.001) and the average FPG has decreased from 8.53 mmol/L (standard deviation: 2.84) to 7.11 mmol/L (standard deviation: 1.34) (p 〈 0.001). Patients in rural areas and with lower education level showed higher FPG and poorer KAP level both before and after the intervention. In conclusion, community-based comprehensive intervention for T2DM is feasible on a large population. Improving and repeating the comprehensive strategy is greatly recommended in order to sustain the impact, especially in rural areas and for patients with lower education levels.
