    Abstract: Ischemia–reperfusion (I/R) injury is a type of pathological injury that commonly arises in various diseases. Various forms of immune response are involved in the process of I/R injury. As a member of the B7 costimulatory molecule family, programmed death 1-ligand 1 (PD-L1) is an important target for immune regulation. Therefore, PD-L1 may be implicated in the regulation of I/R injury. This review briefly describes the immune response during I/R injury and how PD-L1 is involved in its regulation by focusing on findings from various I/R models. Despite the limited number of studies in this field of research, PD-L1 has shown sufficient potential as a clinical therapeutic target.

    Abstract: Metformin is a first-line anti-diabetic agent with a powerful hypoglycemic effect. Several studies have reported that metformin can improve the prognosis of stroke patients and that this effect is independent of its hypoglycemic effect; however, the specific mechanism remains unclear. In this research, we explored the effect and specific mechanism of metformin in cerebral ischemia-reperfusion (I/R) injury by constructing a transient middle cerebral artery occlusion model in vivo and a glucose and oxygen deprivation/reoxygenation (OGD/R) model in vitro . The results of the in vivo experiments showed that acute treatment with low-dose metformin (10 mg/kg) ameliorated cerebral edema, reduced the cerebral infarction volume, improved the neurological deficit score, and ameliorated neuronal apoptosis in the ischemic penumbra. Moreover, metformin up-regulated the brain-derived neurotrophic factor (BDNF) expression and increased phosphorylation levels of AMP-activated protein kinase (AMPK) and cAMP-response element binding protein (CREB) in the ischemia penumbra. Nevertheless, the above-mentioned effects of metformin were reversed by Compound C. The results of the in vitro experiments showed that low metformin concentrations (20 μM) could reduce apoptosis of human umbilical vein endothelial cells (HUVECs) under OGD/R conditions and promote cell proliferation. Moreover, metformin could further promote BDNF expression and release in HUVECs under OGD/R conditions via the AMPK/CREB pathway. The Transwell chamber assay showed that HUVECs treated with metformin could reduce apoptosis of SH-SY5Y cells under OGD/R conditions and this effect could be partially reversed by transfection of BDNF siRNA in HUVECs. In summary, our results suggest that metformin upregulates the level of BDNF in the cerebral ischemic penumbra via the AMPK/CREB pathway, thereby playing a protective effect in cerebral I/R injury.

    Abstract: Intensive cereal production has brought about increasingly serious environmental threats, including global warming, environmental acidification, and water shortage. As an important grain producer in the world, the rice cultivation system in central China has undergone excessive changes in the past few decades. However, few articles focused on the environmental impacts of these shifts from the perspective of ecological footprints. In this study, a 2-year field trial was carried out in Hubei province, China, to gain insight into carbon footprint (CF), nitrogen footprint (NF), and water footprint (WF) performance. The three treatments were, namely, double-rice system (DR), ratoon rice system (RR), and rice-wheat system (RW). Results demonstrated that RR significantly increased the grain yield by 10.22–15.09% compared with DR, while there was no significant difference in the grain yield between RW and DR in 2018–2019. All of the calculation results by three footprint approaches followed the order: RR & lt; RW & lt; DR; meanwhile, RR was always significantly lower than DR. Methane and NH 3 field emissions were the hotspots of CF and NF, respectively. Blue WF accounts for 40.90–42.71% of DR, which was significantly higher than that of RR and RW, primarily because DR needs a lot of irrigation water in both seasons. The gray WF of RW was higher than those of DR and RR, mainly due to the higher application rate of N fertilizer. In conclusion, RR possesses the characteristics of low agricultural inputs and high grain yield and can reduce CF, NF, and WF, considering the future conditions of rural societal developments and rapid demographic changes; we highlighted that the RR could be a cleaner and sustainable approach to grain production.

    Abstract: The poor prognosis and fewer treatment option is a current clinical challenge for patients with lung adenosquamous carcinoma (ASC). The previous studies reported that tumor mutational burden (TMB, numbers of mutation per Megabase) is a predictor of clinical response in trials of multiple cancer types, while fewer studies assessed the relationship between TMB level and clinical features and outcomes of lung ASC. Herein, the present study enrolled Chinese patients with lung ASC. DNA was extracted from formalin-fixed paraffin-embedded tumor samples and subjected to next generation sequencing (NGS), and the 457 cancer related genes were evaluated. The results demonstrated that 95 unique genes with somatic variations were identified in the enrolled patients. The top three of high frequency gene mutations were TP53, EGFR, PIK3CA with rates of 62% (13 cases), 48% (10 cases), and 14% (3 cases), respectively. We identified TMB value was significantly correlated with pathological stages ( p & lt; 0.05) and invasion of lymph node ( p & lt; 0.05). However, TMB value was not significantly correlated to other clinicopathologic indexes, for examples, age, sex, smoking history, tumor size, as well as TP53 and EGFR mutations in lung ASC. Moreover, TMB value was associated with the overall survival ( p & lt; 0.01), but not with the relapse-free survival ( p = 0.23). In conclusion, this study indicated that lung ASC with high TMB might be associated with the invasion of lymph node and short overall survival. Immunotherapy might be a promising treatment option for lung ASC patients with high TMB.
