    Abstract: This paper proposes an optimal placement model of ultra-high frequency (UHF) sensors for accurate location of partial discharge (PD) in gas-insulated switchgear (GIS). The model is based on 0-1 program in consideration of the attenuation influence on the propagation of electromagnetic (EM) waves generated by PD in GIS. the optimal placement plan improves the economy, observability, and accuracy of PD locating. After synchronously acquiring the time of the initial EM waves reaching each UHF sensor, PD occurring time can be obtained. Then, initial locating results can be acquired by using the Euclidean distance measuring method and the extended time difference of arriving (TDOA) location method. With the information of all UHF sensors and the inherent topological structure of GIS, the locating accuracy can be further improved. The method is verified by experiment, showing that the method can avoid the influence of false information and obtain higher locating accuracy by revising initial locating results.

    Abstract: Zonula occludens-1 (ZO-1) is a tight junction protein in the cerebrovascular endothelium, responsible for blood–brain barrier function. Hydroxysafflor yellow A (HSYA) is a major ingredient of safflower (Carthamus tinctorius L.) with antioxidative activity. This study investigated whether HSYA protected ZO-1 by targeting ROS-generating NADPH oxidases (NOXs). HSYA administration reduced cerebral vascular leakage with ZO-1 protection in mice after photothrombotic stroke, largely due to suppression of ROS-associated inflammation. In LPS-stimulated brain microvascular endothelial cells, HSYA increased the ratio of NAD+/NADH to restore Sirt1 induction, which bound to Von Hippel–Lindau to promote HIF-1αdegradation. NOX2 was the predominant isoform of NOXs in endothelial cells and HIF-1α transcriptionally upregulated p47phox and Nox2 subunits for the assembly of the NOX2 complex, but the signaling cascades were blocked by HSYA via HIF-1α inactivation. When oxidate stress impaired ZO-1 protein, HSYA attenuated carbonyl modification and prevented ZO-1 protein from 20S proteasomal degradation, eventually protecting endothelial integrity. In microvascular ZO-1 deficient mice, we further confirmed that HSYA protected cerebrovascular integrity and attenuated ischemic injury in a manner that was dependent on ZO-1 protection. HSYA blocked HIF-1α/NOX2 signaling cascades to protect ZO-1 stability, suggestive of a potential therapeutic strategy against ischemic brain injury.

    Abstract: To explore the mechanistic origin that determines the binding affinity of SARS-CoV-2 spike receptor binding domain (RBD) to human angiotensin converting enzyme 2 (ACE2), we constructed the homology models of RBD-ACE2 complexes of four Omicron subvariants (BA.1, BA.2, BA.3 and BA.4/5), and compared them with wild type complex (RBDWT-ACE2) in terms of various structural dynamic properties by molecular dynamics (MD) simulations and binding free energy (BFE) calculations. The results of MD simulations suggest that the RBDs of all the Omicron subvariants (RBDOMIs) feature increased global structural fluctuations when compared with RBDWT. Detailed comparison of BFE components reveals that the enhanced electrostatic attractive interactions are the main determinant of the higher ACE2-binding affinity of RBDOMIs than RBDWT, while the weakened electrostatic attractive interactions determine RBD of BA.4/5 subvariant (RBDBA.4/5) lowest ACE2-binding affinity among all Omicron subvariants. The per-residue BFE decompositions and the hydrogen bond (HB) networks analyses indicate that the enhanced electrostatic attractive interactions are mainly through gain/loss of the positively/negatively charged residues, and the formation or destruction of the interfacial HBs and salt bridges can also largely affect the ACE2-binding affinity of RBD. It is worth pointing out that since Q493R plays the most important positive contribution in enhancing binding affinity, the absence of this mutation in RBDBA.4/5 results in a significantly weaker binding affinity to ACE2 than other Omicron subvariants. Our results provide insight into the role of electrostatic interactions in determining of the binding affinity of SARS-CoV-2 RBD to human ACE2.

    Abstract: Background: In microsatellite instability (MSI)/mismatch repair-deficient (dMMR) gastrointestinal cancers, the optimum therapy after the progression of immune checkpoint inhibitors (ICIs) is yet unknown. Here, we compared the efficacy of programmed death 1 (PD1)/programmed death ligand-1 (PD-L1) inhibitors plus other therapy and chemotherapy with or without targeted therapy in MSI/dMMR gastrointestinal cancer patients after progression on anti-PD1/PD-L1 monotherapy. Methods: We retrospectively recruited MSI/dMMR gastrointestinal cancer patients who had progressed on anti-PD1/PD-L1 monotherapy. Objective response rate (ORR), disease control rate (DCR), progression-free survival (PFS), overall survival (OS), and PFS ratio (PFSr) were compared between patients who received anti-PD1/PD-L1 plus other therapy (ICI-plus group) and patients who received chemotherapy with or without targeted therapy (chemo-targeted group). Results: In total, 26 and 25 patients were recruited in the ICI-plus group and chemo-targeted group, respectively. Significantly better DCR (80.8% vs. 44.0%, p = 0.007), PFS (median PFS 6.9 months vs. 3.0 months, p = 0.001), OS (median OS NR vs. 14.1 months, p = 0.043), and PFSr (2.4 vs. 0.9, p = 0.021), along with a numerically higher ORR (23.1% vs. 12.0%, p = 0.503) were observed in the ICI-plus group compared with the chemo-targeted group. Multivariate analyses identified the therapy regimen as an important prognostic factor in gastrointestinal cancers. Conclusions: Compared to conventional chemotherapy with or without targeted therapy, continuing anti-PD1/PD-L1 in combination with other treatments showed better clinical outcomes in MSI/dMMR gastrointestinal cancer patients who progressed on PD1/PD-L1 blockade, which should be validated prospectively in clinical trials.

    Abstract: Potential evapotranspiration (ETp) and reference crop evapotranspiration (ETo) are two key parameters in hydrology, meteorology, and agronomy. ETp and ETo are related to each other but have different meanings and applications. In this study, the ETp and ETo were distinguished and calculated with the Penman and FAO56 PM equations using the weather data of 551 stations in China from 1961 to 2018. The differences in their spatiotemporal variations were examined with an MMK test, an R/S test, and wavelet analysis. The monthly ETp and ETo were close but the ETp was always larger than the ETo, with values ranging from 1 to 356 mm and 2 to 323 mm, respectively. Their differences varied in different months and sub-regions. The maximum monthly difference transferred from south to north and then back to the south in a yearly cycle, showing spatiotemporal heterogeneity. The annual values of the ETp and ETo were also close, but the ETp was significantly higher than the ETo. The increasing future trends of ETp but decreasing trends of ETo were tested at most sites in China. Although the primary periods were almost the same, their spatial distribution was slightly different. In conclusion, ETp is different from ETo and they should be applied carefully. This study performs a thorough comparison and reveals the underlying basis of and discrepancy between ETp and ETo.

    Abstract: Studies on the population structure and variation of Magnaporthe oryzae in fields are of great significance for the control of rice blast disease. In this study, a total of 462 isolates isolated from different areas of Hunan Province in 2016 and 2018 were analyzed for their population structure and variation tendency. The results showed that from 2016 to 2018, the concentration of fungal races of M. oryzae increased and the diversity decreased; furthermore, 218 isolates in 2016 belonged to ZA, ZB, ZC, ZE, ZF and ZG, with a total of 6 groups and 29 races, in which the dominant-population ZB group accounted for 66.2%; meanwhile, in 2018, 244 isolates were classified into 4 groups and 21 races, including ZA, ZB, ZC and ZG, in which the dominant-population ZB group accounted for 72.54%. In 2018, isolates of ZD, ZE and ZF populations were absent, and the number of total races and isolates of the ZA and ZC groups decreased. Fungal pathogenicity was identified, with 24 monogenic lines (MLs) carrying 24 major R genes. The resistance frequency of R genes to fungal isolates in 2018 decreased significantly, in which except Pikm was 64.5%, the other monogenic lines were less than 50%. Rep-PCR analysis for isolates of Guidong in Hunan also showed that fungal diversity decreased gradually. The influence of R genes on fungal variation was analyzed. The pathogenicity of isolates purified from Xiangwanxian 11 planted with monogenic lines was significantly more enhanced than those without monogenic lines. All the results indicated that in recent years, the fungal abundance in Hunan has decreased while fungal pathogenicity has increased significantly. This study will greatly benefit rice-resistance breeding and the control of rice blast disease in Hunan Province.

    Abstract: Electrical Capacitance Tomography (ECT) image reconstruction has developed for decades and made great achievements, but there is still a need to find a new theoretical framework to make it better and faster. In recent years, machine learning theory has been introduced in the ECT area to solve the image reconstruction problem. However, there is still no public benchmark dataset in the ECT field for the training and testing of machine learning-based image reconstruction algorithms. On the other hand, a public benchmark dataset can provide a standard framework to evaluate and compare the results of different image reconstruction methods. In this paper, a benchmark dataset for ECT image reconstruction is presented. Like the great contribution of ImageNet that transformed machine learning research, this benchmark dataset is hoped to be helpful for society to investigate new image reconstruction algorithms since the relationship between permittivity distribution and capacitance can be better mapped. In addition, different machine learning-based image reconstruction algorithms can be trained and tested by the unified dataset, and the results can be evaluated and compared under the same standard, thus, making the ECT image reconstruction study more open and causing a breakthrough.
