    Abstract: Qingluoyin (QLY) is a representative herbal formula prescribed for hot symptom-related rheumatoid arthritis treatment. Among its derivatives, Xiaoyao-Qingluoyin (XYQLY) attracts increasing attention due to the notable clinical efficacy. In this study, we compared its effects with QLY on adjuvant-induced arthritis (AIA) in rats and partially elucidated the antirheumatic mechanism using a network pharmacology-based strategy. After continuous oral treatments, clinical outcomes were systematically evaluated by radiographic, histological, immunohistochemical, and serological analyses. Possibly altered pathways were predicted based on reported interactions between the related chemicals and proteins/genes. The obtained conclusion was further validated by experiments in vitro. QLY and XYQLY eased polyarthritis in AIA rats after repeated doses, which reflected in reduced inflammation and bone degradation and downregulated p-p65, MMP3, and TLR4 expressions in joints. Meanwhile, they restored oxidative stress (MDA, SOD, GSH, T-AOC, and NO) and inflammatory indicators (TNF-α and CO) in serum. Synovium-based immunoblotting assay revealed that QLY and XYQLY were similarly effective in downregulating MMP3 and COX-2, but XYQLY treatment exhibited notable merit in suppressing p-p65 expression. Network pharmacology analysis hinted that XYQLY should exert greater impacts on LPS signaling and the downstream. Based on results from LC-MS analysis, we treated AIA rat-derived peripheral blood mononuclear cells with either QLY or XYQLY-based chemical combinations and confirmed that XYQLY had the better potential in inhibiting TLR4/NF-κB-controlled IL-6 production. Consequently, it led to a more profound decrease in Th17 cells counts. Overall evidence demonstrated that XYQLY was especially effective in regulating innate immunity and, therefore, improved immune environment in AIA rats as a whole.

    Abstract: Background. Tuberculosis (TB), a multisystemic disease with protean presentation, remains a major global health problem. Although concurrent pulmonary tuberculosis (PTB) and extrapulmonary tuberculosis (EPTB) cases are commonly observed clinically, knowledge regarding concurrent PTB-EPTB is limited. Here, a large-scale multicenter observational study conducted in China aimed to study the epidemiology of concurrent PTB-EPTB cases by diagnostically defining TB types and then implementing association rules analysis. Methods. The retrospective study was conducted at 21 hospitals in 15 provinces in China and included all inpatients with confirmed TB diagnoses admitted from Jan 2011 to Dec 2017. Association rules analysis was conducted for cases with concurrent PTB and various types of EPTB using the Apriori algorithm. Results. Evaluation of 438,979TB inpatients indicated PTB was the most commonly diagnosed (82.05%) followed by tuberculous pleurisy (23.62%). Concurrent PTB-EPTB was found in 129,422 cases (29.48%) of which tuberculous pleurisy was the most common concurrent EPTB type observed. The multivariable logistic regression models demonstrated that odds ratios of concurrent PTB-EPTB cases varied by gender and age group. For PTB cases with concurrent EPTB, the strongest association was found between PTB and concurrent bronchial tuberculosis (lift = 1.09). For EPTB cases with concurrent PTB, the strongest association was found between pharyngeal/laryngeal tuberculosis and concurrent PTB (lift = 1.11). Confidence and lift values of concurrent PTB-EPTB cases varied with gender and age. Conclusions. Numerous concurrent PTB-EPTB case types were observed, with confidence and lift values varying with gender and age. Clinicians should screen for concurrent PTB-EPTB in order to improve treatment outcomes.

    Abstract: Background. The role of IgG subclass in idiopathic membranous nephropathy (IMN) was unclarified. Recent study found IgG subtype switches from IgG1 to IgG4 in the early pathological stage in IMN. The profile of IgG subclass in phospholipase A2 receptor- (PLA2R-) related and PLA2R-unrelated IMN was unrevealed. Our study is aimed at testifying whether IgG subclass switch existed in PLA2R-related and PLA2R-unrelated IMN, respectively. Methods. Our study retrospectively enrolled 157 Chinese patients with biopsy-confirmed IMN between September 2017 and November 2019. We measured glomerular PLA2R antigen and serum anti-PLA2R antibody to classify the patients into PLA2R-related ( n = 132 ) and PLA2R-unrelated ( n = 25 ) subgroup. We evaluated glomerular IgG subclass by immunofluorescence (IF) predominance. Our study defined IgG subclass deposition as predominant if the IF score was higher than the other three and ≥1 +, or as codominant if the IF intensity was equal to any other and ≥1 +. We explored the relationship between IF predominance of glomerular IgG subtype and electron microscopic (EM) stages of IMN. Results. We did not find statistical difference of predominant or codominant rate (pre/co-rate) among EM stages in any subclass ( P 〉 0.05 ). Pre/co-rate of IgG3 linearly associated with EM stage in total and PLA2R-related subgroup ( P = 0.044 , P = 0.013 ). PLA2R-related subgroup showed higher IgG4 intensity ( 2.1 ± 0.6 vs. 1.6 ± 0.7 , P = 0.001 ) and pre/co-rate of IgG4 in stage 1 (97% vs. 57%, P = 0.015 ) than PLA2R-unrelated group. We found no difference of IgG subclass pre/co-rate in different EM stages or linear association between pre/co-rate of IgG1, IgG2, IgG4, and EM stages ( P 〉 0.05 ). Conclusions. Pre/co-rate of IgG3 declined with EM stage in total and PLA2R-related subgroup. We did not find IgG subclass switches from IgG1 to IgG4 in either IMN patients or subgroups.

    Abstract: In this study, the porosities of C60 high-strength concrete after 0, 30, 60, and 90 freeze-thaw cycles determined via the water retention method are 1.30%, 3.65%, 5.14%, and 7.34%, respectively. Furthermore, a mathematical model of porosity varying with the number of freeze-thaw cycles is established. Using an artificial environment simulation experimental system and the natural diffusion method, the chloride diffusion law of C60 high-strength concrete after 0, 30, 60, and 90 freeze-thaw cycles is obtained. The corresponding diffusion coefficients are calculated based on the experimental results and Fick’s law, where 0.3431 × 10−12, 0.5288 × 10−12, and 0.6712 × 10−12, and 0.8930 × 10−12 m2/s are obtained, respectively, and a mathematical model of diffusion coefficient with freeze-thawing is established. Transport control equations comprising solution flow and solute migration control equations are established for chloride ions in concrete after freeze-thawing cycles. The equations consider the effects of freeze-thawing, solution pressure, solution concentration, solution density, convection, mechanical dispersion, and chemisorption on chloride ion transport in concrete. Using COMSOL numerical software, the transport control equations for chloride ions are solved using a real concrete numerical model, and the chloride ion corrosion process in concrete after freeze-thaw cycles is simulated. The simulation results are consistent with the experimental values.

    Abstract: Bakuchiol (BAK), a monoterpene phenol reported to have exerted a variety of pharmacological effects, has been related to multiple diseases, including myocardial ischemia reperfusion injury, pressure overload-induced cardiac hypertrophy, diabetes, liver fibrosis, and cancer. However, the effects of BAK on hyperglycemia-caused diabetic cardiomyopathy and its underlying mechanisms remain unclear. In this study, streptozotocin-induced mouse model and high-glucose-treated cell model were conducted to investigate the protective roles of BAK on diabetic cardiomyopathy, in either the presence or absence of SIRT1-specific inhibitor EX527, SIRT1 siRNA, or Nrf2 siRNA. Our data demonstrated for the first time that BAK could significantly abate diabetic cardiomyopathy by alleviating the cardiac dysfunction, ameliorating the myocardial fibrosis, mitigating the cardiac hypertrophy, and reducing the cardiomyocyte apoptosis. Furthermore, BAK achieved its antifibrotic and antihypertrophic actions by inhibiting the TGF-β1/Smad3 pathway, as well as decreasing the expressions of fibrosis- and hypertrophy-related markers. Intriguingly, these above effects of BAK were largely attributed to the remarkable activation of SIRT1/Nrf2 signaling, which eventually strengthened cardiac antioxidative capacity by elevating the antioxidant production and reducing the reactive oxygen species generation. However, all the beneficial results were markedly abolished with the administration of EX527, SIRT1 siRNA, or Nrf2 siRNA. In summary, these novel findings indicate that BAK exhibits its therapeutic properties against hyperglycemia-caused diabetic cardiomyopathy by attenuating myocardial oxidative damage via activating the SIRT1/Nrf2 signaling.

    Abstract: Background . Identification of potential molecular targets of acute myocardial infarction is crucial to our comprehensive understanding of the disease mechanism. However, studies of gene coexpression analysis via jointing multiple microarray data of acute myocardial infarction still remain restricted. Methods . Microarray data of acute myocardial infarction (GSE48060, GSE66360, GSE97320, and GSE19339) were downloaded from Gene Expression Omnibus database. Three data sets without heterogeneity (GSE48060, GSE66360, and GSE97320) were subjected to differential expression analysis using MetaDE package. Differentially expressed genes having upper 25% variation across samples were imported in weighted gene coexpression network analysis. Functional and pathway enrichment analyses were conducted for genes in the most significant module using DAVID. The predicted microRNAs to regulate target genes in the most significant module were identified using TargetScan. Moreover, subpathway analyses using iSubpathwayMiner package and GenCLiP 2.0 were performed on hub genes with high connective weight in the most significant module. Results . A total of 1027 differentially expressed genes and 33 specific modules were screened out between acute myocardial infarction patients and control samples. Ficolin (collagen/fibrinogen domain containing) 1 ( FCN1 ), CD14 molecule ( CD14 ), S100 calcium binding protein A9 ( S100A9 ), and mitochondrial aldehyde dehydrogenase 2 ( ALDH2 ) were identified as critical target molecules; hsa-let-7d , hsa-let-7b , hsa-miR-124-3 , and hsa-miR-9-1 were identified as potential regulators of the expression of the key genes in the two biggest modules. Conclusions . FCN1 , CD14 , S100A9 , ALDH2 , hsa-let-7d , hsa-let-7b , hsa-miR-124-3, and hsa-miR-9-1 were identified as potential candidate regulators in acute myocardial infarction. These findings might provide new comprehension into the underlying molecular mechanism of disease.

    Abstract: Nanoindentation is an effective nondestructive method for small scale determination of mechanical properties of materials. However, indentation response of metallic materials is very sensitive to indenter tip roundness, size effects, loading rate, and so forth. This study will analyze the effect of indenter shape imperfections on hardness determination. For this purpose, experimental investigations and finite element simulations are carried out. At first, it is found that hardness values determined with Oliver and Pharr’s method are affected by errors caused by imperfect indenter tip: errors increase for imperfect indenters with larger tip radii. Afterwards, several commonly used methods accounting at different extents for tip radius variations are compared. However, most of those methods are found not to be accurate for shallow indentation. For this reason, a novel hardness determination method based on geometrical relations of the imperfect indenter tip is developed. Results show that the new approach is very effective even in the case of shallow indentation.

    Abstract: Marine corrosion and freeze-thaw environment will bring serious damage to marine concrete structures, leading to affect the safety and service life of structures. With the help of artificial climate and environment simulation laboratory, the variation of the compression strength and elastic modulus of concrete with the number of freeze-thaw cycles and corrosion time under the corrosion and freeze-thaw environment is studied. The results show that both of them firstly increase and then decrease with corrosion time. When the corrosion time is 270 d and the freeze-thaw time is 90 times, the strength of concrete decreases by 13% and the elastic modulus decreases by 5%. Then, based on the theory of damage mechanics, the damage evolution and constitutive model of concrete under the marine corrosion and freeze-thaw environment are established. Compared with the experimental results, it is found that the model can well describe the damage evolution characteristics of concrete under marine corrosion and freeze-thaw environment. Finally, a numerical model is established on the basis of elastic modulus and strength degradation model of concrete under marine corrosion and freeze-thaw environment. Elevated pile caps of concrete pile component are taken as an example to analyze the process of damage, and the change rules of displacement, deformation, and damage of concrete pile are obtained.

    Abstract: In order to study the influence of the softening and swelling of weakly cemented soft rock on the stability of roadway surrounding rock, based on the background of the general return airway in the south wing of Yuwu coal mine, 4 different schemes are designed according to whether the softening or swelling effect of weak cemented soft rock is considered. Based on the numerical simulation program, FLAC3D was used to analyze the distribution characteristics of the shear stress, displacement, and plastic zone of the surrounding rock of the roadway under these 4 designs. On this basis, in view of the geological conditions of the roadway, a new type of “grouting + anchor cable mesh + bottom arch” combined support plan is proposed. The research results show that, under the conventional anchor-net-cable-spraying support scheme, the surrounding rock of the roadway can remain stable without considering the softening and expansion of the weakly cemented soft rock in contact with water and its maximum displacement and plastic zone depth are only 66.9 mm and 2.8 m. When considering the softening and swelling of weakly cemented soft rock roadways, due to the synergistic effect of the two, the maximum displacement of the surrounding rock of the roadway and the depth of the plastic zone will reach 517.4 mm and 6.7 m, respectively, which will continue to increase with excavating along the roadway and lead to ultimately destabilization and destruction. After adopting the combined support of “grouting + anchor cable mesh + bottom arch,” the maximum displacement of the surrounding rock of the roadway and the depth of the plastic zone will be reduced by 77.1% ∼ 92.0% and 18.7% ∼ 72.4%, respectively, and they will basically remain stable after excavation of 5 days.

    Abstract: The reasonable sharing and effective integration of data is the premise of improving the comprehensive governance ability of society through data empowerment. It can be determined that the fine-grained splitting of microservice framework can promote the reasonable sharing and effective integration of data. However, how to determine the granularity of microservice splitting is a multiparameter and multiobjective decision-making problem, which is also a key basic problem to be solved urgently both in academic research and application. From the perspective of application, this paper puts forward the criteria and basic framework that can guide the microservice splitting, and for the first time, based on the perspective of maintainability, it gives the decision criteria and methods of microservice splitting granularity. After the theoretical research, this paper also takes the provincial microservice governance of food and drug regulation as an example: the example shows that the framework and methods proposed can effectively improve data sharing and system expandability. Through microservice governance, collaborative social governance featuring “one network to achieve management objectives, one network to realize comprehensive business processing, one network for comprehensive view of all information” can be achieved. It is an exemplary mode worth popularization.
