    Abstract: In retinal pigment epithelial (RPE) cells, transforming growth factor-beta (TGF-β) plays a critical role in epithelial-mesenchymal transition (EMT), which contributes to various fibrotic retinal disorders. In the present study, we investigated the effect of recombinant human cluster of differentiation 82 ( rh CD82), a tumor metastasis suppressor, on TGF-β-induced EMT in the human RPE cell line APRE-19. The results show that TGF-β1 significantly enhanced cell migration, invasion and the expression of EMT-mediate factors in ARPE-19 cells. However, rh CD82 markedly inhibited cell mobility and the expression of epithelial marker, zonula occludens-1, as well as increased the expression of mesenchymal markers, such as vimentin and α-smooth muscle actin in TGF-β1-treated APRE-19 cells. In addition, TGF-β1 upregulated the phosphorylation of Smad, extracellular signal regulated kinase (ERK) and glycogen synthase kinase-3β (GSK-3β), but only phosphorylation of Smad was suppressed by rh CD82. Noteworthy, rh CD82 greatly suppressed the expression of TGF-β receptor I (TGFRI), TGFRII and integrins in TGF-β1-treated APRE-19 cells. In particular, the result of molecular docking analysis and structural modeling show that rh CD82 partially interacts with the TGF-β1 binding sites of TGFRI, TGFRII, integrin β1 and integrin αv. Taken together, this finding suggested that rh CD82 suppressed TGF-β1-induced EMT of RPE by blocking of Smad-dependent pathway, which is caused by rh CD82 interaction with TGFRs and integrins, suggesting new insight into CD82 as a potential therapeutic strategy in fibrotic retinal disorders.

    Abstract: A dusty thermal vacuum chamber (DTVC) containing a regolith simulant bed is essential for testing equipment and techniques related to lunar surface exploration. Space agencies have been reluctant to operate a DTVC because of the challenge of controlling soil disturbance of the lunar regolith simulant bed during pumping down or depressurization, which may contaminate or even damage the chamber and vacuum equipment. There appears to be no previously available solution to this problem, or how to avoid it. We investigated the mechanism of soil disturbance during depressurization and established a criterion for evaluating its occurrence. The proposed criterion was validated by extensive experiments and numerical modelling to simulate air evacuation from soil voids. There is a critical pressure difference (CPD) between the top and bottom of the lunar regolith simulant bed that causes soil disturbance during depressurization. We found a simple equation estimating the CPD and further provided guideline on the optimum depressurization rate to avoid soil disturbance before the target vacuum level is achieved under varying soil conditions.

    Abstract: Nitric oxide (NO) is a signaling molecule that regulates various processes, including plant growth and development, immunity, and environmental interactions. Using high throughput RNA-seq data, we explored the role of the NO-induced ATILL6 gene in plant growth and defense using functional genomics. The atill6 mutant and wild-types were challenged with either oxidative (H 2 O 2 , MV) or nitro-oxidative (CySNO, GSNO) stress conditions, and the phenotypic results showed that ATILL6 gene differentially regulates cotyledon development frequency (CDF) as well as the root and shoot lengths of the plants. To investigate whether ATILL6 plays a role in plant basal or resistance ( R ) -gene -mediated defense, the plants were challenged with either virulent or avirulent strains of Pseudomonas syringae pathovar tomato (Pst) DC3000. The atill6 line showed a susceptible phenotype, higher pathogen growth, and highly reduced transcript accumulation of PR1 and PR2 genes. These results suggested that ATILL6 positively regulates plant basal defense. Furthermore, after the inoculation of atill6 with avirulent Pst (DC3000), the expressions of the PR1 and PR2 genes decreased, suggesting a positive role in R-gene -mediated resistance in protecting the plant from further spread of disease. We also investigated the role of ATILL6 in systemic acquired resistance (SAR), and the results showed that ATILL6 positively regulates SAR, as the mutant line atill6 has significantly ( p ≤ 0.05) lower transcript accumulation of PR, G3DPH , and AZI genes. Overall, these results indicate that the NO-induced ATILL6 gene differentially regulates plant growth and positively regulates plant basal defense, R-gene -mediated resistance, and SAR.

    Abstract: Excessive alcohol consumption is one of the most significant causes of morbidity and mortality worldwide. Alcohol is oxidized to toxic and carcinogenic acetaldehyde by alcohol dehydrogenase (ADH) and further oxidized to a non-toxic acetate by aldehyde dehydrogenase (ALDH). There are two major ALDH isoforms, cytosolic and mitochondrial, encoded by ALDH1 and ALDH2 genes, respectively. The ALDH2 polymorphism is associated with flushing response to alcohol use. Emerging evidence shows that Lactobacillus and Bifidobacterium species encode alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) mediate alcohol and acetaldehyde metabolism, respectively. A randomized, double-blind, placebo-controlled crossover clinical trial was designed to study the effects of Lactobacillus and Bifidobacterium probiotic mixture in humans and assessed their effects on alcohol and acetaldehyde metabolism. Here, twenty-seven wild types (ALDH2*1/*1) and the same number of heterozygotes (ALDH2*2/*1) were recruited for the study. The enrolled participants were randomly divided into either the probiotic (Duolac ProAP4) or the placebo group. Each group received a probiotic or placebo capsule for 15 days with subsequent crossover. Primary outcomes were measurement of alcohol and acetaldehyde in the blood after the alcohol intake. Blood levels of alcohol and acetaldehyde were significantly downregulated by probiotic supplementation in subjects with ALDH2*2/*1 genotype, but not in those with ALDH2*1/*1 genotype. However, there were no marked improvements in hangover score parameters between test and placebo groups. No clinically significant changes were observed in safety parameters. These results suggest that Duolac ProAP4 has a potential to downregulate the alcohol and acetaldehyde concentrations, and their effects depend on the presence or absence of polymorphism on the ALDH2 gene.

    Abstract: Alzheimer’s disease (AD) is characterised by amyloid-beta accumulation (A), tau aggregation (T) and neurodegeneration (N). Vascular (V) burden has been found concomitantly with AD pathology and has synergistic effects on cognitive decline with AD biomarkers. We determined whether cognitive trajectories of AT(N) categories differed according to vascular (V) burden. Methods We prospectively recruited 205 participants and classified them into groups based on the AT(N) system using neuroimaging markers. Abnormal V markers were identified based on the presence of severe white matter hyperintensities. Results In A+ category, compared with the frequency of Alzheimer’s pathological change category (A+T–), the frequency of AD category (A+T+) was significantly lower in V+ group (31.8%) than in V– group (64.4%) (p=0.004). Each AT(N) biomarker was predictive of cognitive decline in the V+ group as well as in the V– group (p 〈 0.001). Additionally, the V+ group showed more severe cognitive trajectories than the V– group in the non-Alzheimer’s pathological changes (A–T+, A–N+; p=0.002) and Alzheimer’s pathological changes (p 〈 0.001) categories. Conclusion The distribution and longitudinal outcomes of AT(N) system differed according to vascular burdens, suggesting the importance of incorporating a V biomarker into the AT(N) system.

    Abstract: To investigate the utility of preoperative multiparametric magnetic resonance imaging (mpMRI)-based clinical-radiomic analysis combined with machine learning (ML) algorithms in predicting the expression of the Ki-67 proliferative index and p53 tumor suppressor protein in patients with meningioma. Methods This multicenter retrospective study included 483 and 93 patients from two centers. The Ki-67 index was classified into high (Ki-67≥5%) and low (Ki-67 & lt;5%)-expressed groups, and the p53 index was classified into positive (p53≥5%) and negative (p53 & lt;5%)-expressed groups. Clinical and radiological features were analyzed using univariate and multivariate statistical analyses. Six ML models were performed with different types of classifiers to predict Ki-67 and p53 status. Results In the multivariate analysis, larger tumor volumes (p & lt;0.001), irregular tumor margin (p & lt;0.001), and unclear tumor-brain interface (p & lt;0.001) were independently associated with a high Ki-67 status, whereas the presence of both necrosis (p=0.003) and the dural tail sign (p=0.026) were independently associated with a positive p53 status. A relatively better performance was yielded from the model constructed by combined clinical and radiological features. The area under the curve (AUC) and accuracy of high Ki-67 were 0.820 and 0.867 in the internal test, and 0.666 and 0.773 in the external test, respectively. Regarding p53 positivity, the AUC and accuracy were 0.858 and 0.857 in the internal test, and 0.684 and 0.718 in the external test. Conclusion The present study developed clinical-radiomic ML models to non-invasively predict Ki-67 and p53 expression in meningioma using mpMRI features, and provides a novel non-invasive strategy for assessing cell proliferation.

    Abstract: Employees in the defense industry are required to enter test sites and maintenance hangars under military control for the operation tests and maintenance of military equipment. Despite the strengthening of safety standards for defense industry employees since 2019, they remain in a blind spot for safety accidents. However, the safety management of military facilities is conducted in accordance with the National Defense Safety Management Directive, necessitating a reassessment for the safety of defense industry employees. In this study, an analysis was conducted on the National Defense Safety Management Directive to examine the legal basis for preventing safety accidents among defense industry employees and for ensuring the obligation of contractors to secure safety and health under the Serious Accident Punishment Act. In addition, in-depth interviews were conducted to investigate and analyze the actual safety management status of defense industry employees. As a result of the analysis, it was found that the scope of the current military safety management instruction was limited to the Ministry of National Defense and institutional units, excluding defense industry workers, and it was confirmed that safety management of defense industry workers was not properly performed during test evaluation conducted within weapon system research institutions. In this study, the following implications and measures to strengthen safety standards were suggested. Firstly, the application scope of the Serious Accident Punishment Act should encompass employees in the defense industry. Secondly, a compensation system for accident victims needs to be implemented. Thirdly, substantive safety training for employees should be conducted, taking into account the risk factors at defense industry sites. These research findings are expected to contribute to enhancing the technological capabilities of the South Korean defense industry by preventing accidents at defense sites and actualizing compensation for victims.
