    Abstract: The effects of tree age on the growth of cutting seedlings propagated from ancient trees have been an important issue in plant breeding and cultivation. In order to understand seedling growth and stress resistance stability, phenotypic measurements, physiological assays, and high-throughput transcriptome sequencing were performed on sown seedlings propagated from 5-year-old donors and cutting seedlings propagated from 5-, 300-, and 700-year-old Platycladus orientalis donors. In this study, the growth of cutting seedlings propagated from ancient trees was significantly slower; the soluble sugar and chlorophyll contents gradually decreased with the increase in the age of donors, and the flavonoid and total phenolic contents of sown seedlings were higher than those of cutting seedlings. Enrichment analysis of differential genes showed that plant hormone signal transduction, the plant–pathogen interaction, and the flavone and flavonol biosynthesis pathways were significantly up-regulated with the increasing age of cutting seedlings propagated from 300- and 700-year-old donors. A total of 104,764 differentially expressed genes were calculated using weighted gene co-expression network analysis, and 8 gene modules were obtained. Further, 10 hub genes in the blue module were identified, which revealed that the expression levels of JAZ, FLS, RPM1/RPS3, CML, and RPS2 increased with the increase in tree age. The results demonstrated that the age of the donors seriously affected the growth of P. orientalis cutting seedlings and that cutting propagation can preserve the resistance of ancient trees. The results of this study provide important insights into the effects of age on asexually propagated seedlings, reveal potential molecular mechanisms, and contribute to an improvement in the level of breeding and conservation of ancient germplasm resources of P. orientalis trees.

    Abstract: Cajanolactone A (CLA) is a stilbenoid discovered by us from Cajanus cajan (L.) Millsp. In our study, CLA was found to promote osteoblast differentiation in human bone marrow mesenchymal stem cells (hBMSCs), as judged by increased cellular alkaline phosphatase activity and extracellular calcium deposits, and elevated protein expression of Runx2, collagen-1, bone morphogenetic protein-2, and osteopontin. Mechanistic studies revealed that hBMSCs undergoing osteoblast differentiation expressed upregulated mRNA levels of Wnt3a, Wnt10b, LRP5/6, Frizzled 4, β-catenin, Runx2, and Osterix from the early stage of differentiation, indicating the role of activated Wnt/β-catenin signaling pathway in osteoblast differentiation. Addition of CLA to the differentiation medium further increased the mRNA level of Wnt3a, Wnt10b, Frizzled 4, LRP5, and β-catenin, inferring that CLA worked by stimulating Wnt/LRP5/β-catenin signaling. Wnt inhibitor dickkopf-1 antagonized CLA-promoted osteoblastogenesis, indicating that CLA did not target the downstream of canonical Wnt signaling pathway. Treatment with CLA caused no changes in mRNA expression level, as well as protein secretion of osteoprotegerin (OPG) and receptor activator of nuclear factor kappa-B ligand (RANKL), indicating that CLA did not affect the OPG/RANKL axis. Our results showed that CLA, which promoted osteoblast differentiation in hBMSCs, through activating Wnt/LRP5/β-catenin signaling transduction, is a promising anti-osteoporotic drug candidate.

    Abstract: The Chinese Fengyun–4A geostationary meteorological satellite was successfully launched on 11 December 2016, carrying an Advanced Geostationary Radiation Imager (AGRI) to provide the observations of visible, near infrared, and infrared bands with improved spectral, spatial, and temporal resolution. The AGRI infrared observations can be assimilated into numerical weather prediction (NWP) data assimilation systems to improve the atmospheric analysis and weather forecasting capabilities. To achieve data assimilation, the first and crucial step is to characterize and evaluate the biases of the AGRI brightness temperatures in infrared channels 8–14. This study conducts the assessment of clear–sky AGRI full–disk infrared observation biases by coupling the RTTOV model and ERA Interim analysis. The AGRI observations are generally in good agreement with the model simulations. It is found that the biases over the ocean and land are less than 1.4 and 1.6 K, respectively. For bias difference between land and ocean, channels 11–13 are more obvious than water vapor channels 9–10. The fitting coefficient of linear regression tests between AGRI biases and sensor zenith angles manifests no obvious scan angle–dependent biases over ocean. All infrared channels observations are scene temperature–dependent over the ocean and land.

    Abstract: To evaluate the effects of donor ages on growth and stress resistance of 6-year-old seedlings propagated from 5-, 2000-, and 3000-year-old Platycladus orientalis donors with grafting, cutting, and seed sowing, growth indicators and physiological and transcriptomic analyses were performed in 6-year-old seedlings in winter. Results showed that basal stem diameters and plant heights of seedlings of the three propagation methods decreased with the age of the donors, and the sown seedlings were the thickest and tallest. The contents of soluble sugar, chlorophyll, and free fatty acid in apical leaves of the three propagation methods were negatively correlated with donor ages in winter, while the opposite was true for flavonoid and total phenolic. The contents of flavonoid, total phenolic, and free fatty acid in cutting seedlings were highest in the seedlings propagated in the three methods in winter. KEGG (Kyoto Encyclopedia of Genes and Genomes) enrichment analysis of differentially expressed genes showed phenylpropanoid biosynthesis and fatty acid metabolism pathways, and their expression levels were up-regulated in apical leaves from 6-year-old seedlings propagated from 3000-year-old P. orientalis donors. In addition, hub genes analysis presented that C4H, OMT1, CCR2, PAL, PRX52, ACP1, AtPDAT2, and FAD3 were up-regulated in cutting seedlings, and the gene expression levels decreased in seedlings propagated from 2000- and 3000-year-old donors. These findings demonstrate the resistance stability of cuttings of P. orientalis and provide insights into the regulatory mechanisms of seedlings of P. orientalis propagated from donors at different ages in different propagation methods against low-temperature stress.

    Abstract: The use of nanomedicines for cancer treatment has been widespread. Fullerenes have significant effects in the treatment of solid tumors. Here, we are going to study the effects of hydroxylated fullerene C60(OH)n(n = 18–22) treatment on chronic myeloid leukemia cell proliferation and investigate its toxicity. The results showed that hydroxylated fullerene C60(OH)n (n = 18–22) at low concentrations (less than 120 μM) not only had apparent toxic side effects, but also promoted the growth of K562 cells, while a high concentration of C60(OH)n had different degrees of inhibition on K562 cells. When the concentration is higher than 160 μM, the K562 cells showed morphological changes, the mitochondrial membrane potential decreased, the cell cycle was blocked in the stage of G2-phase, and cell apoptosis occurred, which may cause apoptosis, autophagy, and a variety of other damage leading to cell death. Meanwhile, it also indicated that its inhibition of solid tumors might be related to the tumor microenvironment; we verified the safety of fullerene without apparent cellular toxicity at a specific concentration.

    Abstract: Ground Penetrating Radar (GPR) is a geophysical method that uses antennas to transmit and receive high-frequency electromagnetic waves to detect the properties and distribution of materials in media. In this paper, geological observation, UAV detection and GPR technology are combined to study the recent sediments of the Yungang braided river study area in Datong. The application of the GPR technique to the description of fluvial facies and reservoir architecture and the development of geological models are discussed. The process of GPR detection technology and application includes three parts: GPR data acquisition, data processing and integrated interpretation of GPR data. The geological surface at different depths and scales can be identified by using different combinations of frequencies and antenna configurations during acquisition. Based on outcrop observation and lithofacies analysis, the Yandong Member of the Middle Jurassic Yungang Formation in the Datong Basin has been identified as a typical sandy braided river sedimentary system. The sandy braided river sandbody changes rapidly laterally, and the spatial distribution and internal structure of the reservoir are very complex, which has a very important impact on the migration and distribution of oil and gas as a reservoir. It is very important to make clear the characteristics of each architectural unit of the fluvial sand body and quantitatively characterize them. The architectural elements of the braided river sedimentary reservoir in the Datong-Yungang area can be divided into three types: Channel unit, bar unit and overbank assemblages. The geological radar response characteristics of different types of sedimentary units are summarized and their interfaces are identified. The channel sediments form a lens-shaped wave reflection with a flat at the top and convex-down at the bottom in the radar profile, and the angles of the radar reflection directional axes are different on both sides of the sedimentary interface. In the radar profile, the deposit of the unit bar is an upward convex reflection structure. The overbank siltation shows a weak amplitude parallel reflection structure. The flood plain sediments are distributed continuously and stably in the radar profile, showing weak reflection characteristics. Different sedimentary units are identified by GPR data and combined with Unmanned Aerial Vehicle (UAV) detection data, and the establishment of the field outcrop geological model is completed. The development pattern of the diara is clarified, and the swing and migration of the channel in different stages are identified.
