    Abstract: Natural products and metals play a crucial role in cancer research and the development of antitumor drugs. We designed and synthesized three new carboline-based cyclometalated iridium complexes [Ir(C-N)2(PPβC)](PF6), where PPβC = N-(1,10-phenanthrolin-5-yl)-1-phenyl-9H-pyrido[3,4-b] indole-3-carboxamide, C-N = 2-phenylpyridine (ppy, Ir1), 2-(2,4-difluorophenyl) pyridine (dfppy, Ir2), 7,8-benzoquinoline (bzq, Ir3), by combining iridium with β-carboline derivative. These iridium complexes exhibited high potential antitumor effects after being promptly taken up by A549 cells. Accumulating in mitochondria rapidly and preferentially, Ir1-3 caused a series of changes in mitochondrial events, including the loss of mitochondrial membrane potential, the depletion of cellular ATP, and the elevation of reactive oxygen species, leading to significant death of A549 cells. Moreover, the activation of intracellular caspase pathway and apoptosis was further validated to contribute to iridium complexes-induced cytotoxicity. These novel iridium complexes exerted a prominent inhibitory effect on tumor growth in a three-dimensional multicellular tumor spheroid model.

    Abstract: What are the factors influencing the occurrence of monozygotic (MZ) twins in offspring conceived by ART? SUMMARY ANSWER Parental ages, the transfer of fresh verses frozen embryos and the grade of blastocysts are all related to MZ twinning in ART offspring. WHAT IS KNOWN ALREADY Offspring conceived by ART have significantly increased risk of MZ twins, which may be due to the characters of infertility population. The objective of this study was to explore the incidence of monozygotic (MZ) twins after assisted reproductive technology (ART) and to clarify the risk factors for MZ twinning. STUDY DESIGN, SIZE, DURATION A total of 255 monozygotic twins were enrolled in this cohort study, and then matched with singletons at a ratio of 1:4 randomly (with 1020 in the control group). All offspring were conceived by single embryo transfer. PARTICIPANTS/MATERIALS, SETTING, METHODS The collected data were divided into the following three aspects for analysis: characteristics of the infertile population, gamete or embryo manipulations, and factors related to embryo development. MAIN RESULTS AND THE ROLE OF CHANCE The incidence of MZ twins was 1.638% (255 out of 15,567 pregnancies after single embryo transfers). Compared to singleton births, a significantly lower rate of frozen embryo transfers (FET; 78.0% vs 86.1% P = 0.002) was seen amongst the MZ twins. Amongst fresh ETs, the rate of blastocyst transfers in the MZ twins group was higher compared to that in the control group (92.9% vs 75.4%, P = 0.005). We also found that certain grades of blastocysts in terms of trophectoderm (TE) development, inner cell mass (ICM)+TE development and the classification of ′top-quality′ embryos were associated with the incidence of MZ twinning (P = 0.025, P = 0.012, P = 0.020, respectively). Logistic regression analysis revealed that higher paternal age (OR = 0.94, 95% CI = 0.89-1.00, P = 0.029) and FET (OR = 0.48, 95% CI = 0.33-0.68, P = 0.001) may be protective factors against MZ twinning. However, higher maternal age (OR = 1.07, 95% CI = 1.01-1.13, P = 0.027) and the transfer of blastocysts (OR = 4.31, 95% CI = 1.46-12.73, P = 0.008) appeared to be associated with an increased risk of MZ twinning. Amongst blastocyst transfers, a C grade TE may be protective factor against MZ twinning (B: OR = 1.90, 95% CI = 1.18-3.07, P = 0.009; A: OR = 1.58, 95% CI = 0.93-2.67, P = 0.089). LIMITATIONS, REASONS FOR CAUTION First, our definition of MZ twins was based on twins’ birth after single embryo transfers (SET), rather than ultrasound examination during early pregnancy. Second, the parental characteristics of the two groups were homogenous, so it was difficult to find any associations between infertility factors and the incidence of MZ twins. WIDER IMPLICATIONS OF THE FINDINGS This multifaceted analysis of the risk factors for MZ twinning provides some information for clinical interventions in high-risk populations.     Abstract: Multiple disease resistance (MDR) in maize has attracted increasing attention. However, the interplay between cell death and metabolite changes and their contributions to MDR remains elusive in maize. In this study, we identified a mutant named as lesion mimic 30 (les30) that showed ‘suicidal’ lesion formation in the absence of disease and had enhanced resistance to the fungal pathogen Curvularia lunata. Using map-based cloning, we identified the causal gene encoding pheophorbide a oxidase (PAO), which is known to be involved in chlorophyll degradation and MDR, and is encoded by LETHAL LEAF SPOT1 (LLS1). LLS1 was found to be induced by both biotic and abiotic stresses. Transcriptomics analysis showed that genes involved in defense responses and secondary metabolite biosynthesis were mildly activated in leaves of the les30 mutant without lesions, whilst they were strongly activated in leaves with lesions. In addition, in les30 leaves with lesions, there was overaccumulation of defense-associated phytohormones including jasmonic acid and salicylic acid, and of phytoalexins including phenylpropanoids, lignin, and flavonoids, suggesting that their biosynthesis was activated in a lesion-dependent manner. Taken together, our study implies the existence of an interactive amplification loop of interrupted chlorophyll degradation, cell death, expression of defense-related genes, and metabolite changes that results in suicidal lesion formation and MDR, and this has the potential to be exploited by genetic manipulation to improve maize disease resistance.

    Abstract: To analyze the relationship between gestational hypertension and P-wave dispersion and QTc interval dispersion. Methods From January 2017 to December 2019, 213 pregnant women who met the diagnosis of hypertension were selected as gestational hypertension with 248 healthy pregnant women as controls. The basic data, P-wave dispersion, and QTc dispersion were compared between the 2 groups, and binary logistic regression analysis was performed. Results The minimum time of P wave (78.59 ± 9.32 vs. 94.61 ± 7.03 ms) and the minimum time of QTc (384.65 ± 21.69 vs. 401.91 ± 15.49 ms) in gestational hypertension were significantly lower than those in the controls. The dispersion of P wave (42.75 ± 9.94 vs. 14.91 ± 4.03 ms), QTc (57.15 ± 16.10 vs. 21.42 ± 6.07 ms), the maximum time of P wave (121.34 ± 7.22 vs. 112.53 ± 6.43 ms), and the maximum time of QTc (441.80 ± 25.42 vs. 429.74 ± 27.83 ms) in gestational hypertension were significantly higher than those in the controls (all P & lt; 0.05). Logistic regression analysis showed that P-wave dispersion (odds ratio = 1.795, 95% confidence interval 1.266–2.546), QTc minimum time (odds ratio = 0.905, 95% confidence interval 0.833–0.983), and QTc dispersion (odds ratio = 1.216, 95% confidence interval 1.042–1.420) were correlated with gestational hypertension. Conclusions P-wave dispersion and QTc dispersion are associated with gestational hypertension.

    Abstract: More immune-related adverse events (irAEs) have emerged along with increased immune checkpoint inhibitor (ICI) treatment. ICI-induced myocarditis is a rare type of irAE with early onset, rapid progression, and high mortality. Its specific pathophysiological mechanism is not fully understood. In total, 46 patients with tumors and 16 patients with ICI-induced myocarditis were included. We performed single-cell RNA sequencing on CD3 + T cells, flow cytometry, proteomics, and lipidomics to improve our understanding of the disease. First, we demonstrate the clinical features of patients with PD-1 inhibitor–induced myocarditis. We then identified 18 subsets of T cells using single-cell RNA sequencing and performed comparative analysis and further verification. The composition of T cells in the peripheral blood of patients has changed remarkably. Compared with non-irAE patients, effector T cells were increased in irAE patients, while naive T cells, γδ T cells, and mucosal-associated invariant T cell cluster cells were decreased. Besides, reduced γδ T cells characterized with effector functions, increased natural killer T cells with high levels of FCER1G in patients may suggest an association with disease development. Meanwhile, the peripheral inflammatory response was exacerbated in patients, accompanied by upregulation of exocytosis as well as increased levels of multiple lipids. We provide a comprehensive overview of the composition, gene profiles, and pathway signatures of CD3+ T cells driven by PD-1 inhibitor–induced myocarditis, as well as illustrate clinical features and multi-omic characteristics, providing a unique perspective on disease progression and therapy in clinical practice.

    Abstract: The human cingulate cortex (CC) is a complex region that is characterized by heterogeneous cytoarchitecture, connectivity, and function, and it is associated with various cognitive functions. The adult CC has been divided into various subregions, and this subdivision is highly consistent with its functional differentiation. However, only a few studies have focused on the function of neonatal CC. The aim of this study was to describe the cingulate segregation and the functional connectivity of each subdivision in full-term neonates (n = 60) based on resting-state functional magnetic resonance imaging. The neonatal CC was divided into three subregions, and each subregion showed specific connectivity patterns. The anterior cingulate cortex was mainly correlated with brain regions related to the salience (affected) network and default mode network (DMN), the midcingulate cortex was related to motor areas, and the posterior cingulate cortex was coupled with DMN. Moreover, we found that the cingulate subregions showed distinct functional profiles with major brain networks, which were defined using independent component analysis, and exhibited functional lateralization. This study provided new insights into the understanding of the functional specialization of neonatal CC, and these findings may have significant clinical implications, especially in predicting neurological disorder.
