    Abstract: Root restriction cultivation leads to early maturation and quality improvement, especially in the anthocyanin content in grapevine. However, the molecular mechanisms that underlie these changes have not been thoroughly elucidated. In this study, four small RNA libraries were constructed, which included the green soft stage (GS) and ripe stage (RS) of ‘Muscat’ (Vitis vinifera L.) grape berries that were grown under root restriction (RR) and in traditional cultivation (no root restriction, CK). A total of 162 known miRNAs and 14 putative novel miRNAs were detected from the four small RNA libraries by high-throughput sequencing. An analysis of differentially expressed miRNAs (DEMs) revealed that 13 miRNAs exhibited significant differences in expression between RR and CK at the GS and RS stages, respectively. For different developmental stages of fruit, 23 and 34 miRNAs showed expression differences between the GS and RS stages in RR and CK, respectively. The expression patterns of the eight DEMs and their targets were verified by qRT-PCR, and the expression profiles of target genes were confirmed to be complementary to the corresponding miRNAs in RR and CK. The function of Vvi-miR828, which showed the down regulated expression in the RS stage under root restriction, was identified by gene transformation in Arabidopsis. The anthocyanin content significantly decreased in transgenic lines, which indicates the regulatory capacity of Vvi-miR828 in fruit coloration. The miRNA expression pattern comparison between RR and CK might provide a means of unraveling the miRNA-mediated molecular process regulating grape berry development under root restricted cultivation.

    Abstract: The quantitative characterization of microstructure is most desirable for the establishment of structure-property relationships in polymer nanocomposites. In this work, the effects of graphene on the microstructure, mechanical, electrical, and thermal properties of the obtained graphene/polyethylene (PE) composites were investigated. In order to reveal the structure-performance relationship of graphene/PE composites, especially for the effects of the relative free volume fraction (fr) and interfacial interaction intensity (β), positron annihilation lifetime spectroscopy (PALS) was employed for its quantitative description. The relative free volume fraction fr gives a good explanation of the variation for surface resistivity, melting temperature, and thermal stability, and the variation of tensile strength and thermal conductivity agree well with the results of interfacial interaction intensity β. The results showed that fr and β have a significant effect on the properties of the obtained graphene/PE composites, and the effect on the properties was revealed.

    Abstract: Giving a deep insight into the microstructure, and realizing the correlation between microstructure and properties is very important to the precise construction of high-performance graphene/polymer nanocomposites (GPN). For the promising application in microstructure characterization, much attention has been focused on the effective technique of positron annihilation lifetime spectroscopy (PALS). Based on the introduction of the basic principle, this review summarized the application progress of PALS in the correlation of microstructure and properties for GPN, especially for the characterization of free volume and interfacial interaction, and the correlation of these microstructures and properties.

    Abstract: The volatiles in coffee play an important part in the overall flavor profile. In this study, GC–TOF/MS and GC×GC–TOF/MS were used to detect the volatile organic compounds (VOCs) in coffee samples of three different brands at three states (bean, powder, and brew). The differences between the two methods in characterizing VOCs were analyzed using the Venn diagram and PCA (principal component analysis). The important aroma-contributing compounds were further compared and analyzed. The results of the venn diagrams of different coffee samples showed that most VOCs existed in 2–3 kinds of coffee. The PCA of VOCs in different coffee samples showed that the VOCs detected by GC–TOF/MS could distinguish the coffee samples in the different states. GC×GC–TOF/MS was suitable for the further identification and differentiation of the different brands of coffee samples. In addition, pyridine, pyrrole, alcohols, and phenols greatly contributed to distinguishing coffee in three states, and alcohols greatly contributed to distinguishing the three brands of coffee.

    Abstract: Endophyte Bacillus methylotrophicus NJ13 was isolated from Panax ginseng. Its sterile fermentation liquid showed a significant inhibitory effect against Ilyonectria robusta, causing the rusty root rot of P. ginseng and P. quinquefolius. The antifungal protein was obtained after precipitation by 20% saturated ammonium sulfate, desalted by Sephadex G-25, weak anion exchange chromatography, and gel filtration chromatography. SDS-PAGE showed that the purified protein was approximately 29 KDa. The antifungal protein after desalting was not resistant to temperatures higher than 100 °C, resistant to acid conditions, and did not tolerate organic solvents and protease K. The amino acid sequence of purified antifungal protein had an identity of 76% to flagellin from Bacillus velezensis. The isoelectric point of the protein was 4.97 and its molecular mass was 27 KDa. Therefore, a specific primer G1 was designed based on the flagellin gene sequence, and a 770 bp gene sequence was cloned in NJ13 genomic DNA, which shared the same size of flagellin. There were ten base differences between the gene sequences of flagellin and the cloned gene, however, the amino acid sequence encoded by the cloned gene was identical to the flagellin. In conclusion, the antifungal protein produced by biocontrol agent NJ13 contained a flagellin protein.

    Abstract: The incorporation of graphene oxide (GO) into a polymeric drug carrier can not only enhance the loading efficiency but also reduce the initial burst and consequently improve the controllability of drug release. Firstly, 5-fluorouracil (5-Fu)-loaded hydroxypropyl cellulose/chitosan (HPC/CS@5-Fu) and GO/HPC/CS@5-Fu aerogels were successfully fabricated through chemical cross-linking with glutaraldehyde. Then, the obtained aerogels were characterized using scanning electron microscopy (SEM), Fourier transform infrared (FITR), X-ray diffraction (XRD), differential scanning calorimetry (DSC), thermogravimetry (TG), and the effect of HPC and GO content on the drug loading (DL) and encapsulation efficiency (EE) for the two aerogels were investigated, respectively. Finally, the drug release behavior of the GO/HPC/CS@5-Fu aerogels with different GO content was evaluated at two different pH values, and four kinds of kinetic models were used to evaluate the release behavior.

    Abstract: The flavor of coffee can be affected by the preparation parameters. In this investigation, the flavor profiles of three coffee brands under three conditions (bean, powder, and brew) were analyzed by gas chromatography—ion mobility spectrometry (GC-IMS) and the electronic nose (E-nose). The flavor results were further studied using multiple factor analysis (MFA). A total of 117 peaks were identified in all coffee samples by GC-IMS, and the principal component analysis (PCA) showed these coffee samples could be grouped and separated. A total of 37 volatile organic compounds (VOCs) were selected as biomarkers to distinguish coffee samples, including 5 aldehydes, 10 ketones, 8 alcohols, 2 acids, 4 esters, 5 furans, and 3 other compounds. The comparison between E-nose and GC-IMS data using partial least squares regression (PLSR) and MFA showed GC-IMS could present very close sample spaces. Compared with E-nose, GC–IMS could not only be used to classify coffee samples in a very short time but also provide VOC bio-markers to discriminate coffee samples.

    Abstract: Remote sensing nighttime lights (NTLs) offers a unique perspective on human activity, and NTL images are widely used in urbanization monitoring, light pollution, and other human-related research. As one of the payloads of sustainable development science Satellite-1 (SDGSAT-1), the Glimmer Imager (GI) provides a new multi-spectral, high-resolution, global coverage of NTL images. However, during the on-orbit testing of SDGSAT-1, a large number of stripes with bad or corrupted pixels were observed in the L1A GI image, which directly affected the accuracy and availability of data applications. Therefore, we propose a novel destriping algorithm based on anomaly detection and spectral similarity restoration (ADSSR) for the GI image. The ADSSR algorithm mainly consists of three parts: pretreatment, stripe detection, and stripe restoration. In the pretreatment, salt-pepper noise is suppressed by setting a minimum area threshold of the connected components. Then, during stripe detections, the valid pixel number sequence and the total pixel value sequence are analyzed to determine the location of stripes, and the abnormal pixels of each stripe are estimated by a clustering algorithm. Finally, a spectral-similarity-based method is adopted to restore all abnormal pixels of each stripe in the stripe restoration. In this paper, the ADSSR algorithm is compared with three representative destriping algorithms, and the robustness of the ADSSR algorithm is tested on different sizes of GI images. The results show that the ADSSR algorithm performs better than three representative destriping algorithms in terms of visual and quantitative indexes and still maintains outstanding performance and robustness in differently sized GI images.
