    Abstract: The radiance of sky brightness differs principally with wavelength passband. Atmospheric scattering of sunlight causes the radiation in the near-infrared band. The Antarctic is a singular area of the planet, marked by an unparalleled climate and geographical conditions, including the coldest temperatures and driest climate on Earth, which leads it to be the best candidate site for observing in infrared bands. At present, there are still no measurements of night-sky brightness at DOME A. We have developed the Near-Infrared Sky Brightness Monitor (NISBM) in the J, H, and Ks bands for measurements at DOME A. The instruments were installed at DOME A in 2019 and early results of NIR sky brightness from 2019 January–April have been obtained. The variation of sky background brightness with solar elevation and scanning angle is analysed. The zenith sky flux intensity for the early night at DOME A in the J band is in the 600–1100 μJy arcsec−2 range, that in the H band is between 1100 and 2600 μJy arcsec−2, and that in the Ks band is in the range ∼200–900 μJy arcsec−2. This result shows that the sky brightness in J and H bands is close to that of Ali in China and Mauna Kea in the USA. The sky brightness in the Ks band is much better than that in Ali, China and Mauna Kea, USA. This shows that, from our early results, DOME A is a good site for astronomical observation in the Ks band.

    Abstract: Common variable immunodeficiency (CVID) is a ‘late-onset’ primary immunodeficiency characterized by variable manifestations and genetic heterogeneity. A monogenic cause of CVID has been reported in 10% of patients. In this study, we identified two novel pathogenic variants implicated in monogenic CVID by whole exome sequencing (WES) analysis: a heterozygous nuclear factor κB subunit 1 (NFKB1) p.G686fs mutation and a homozygous inducible T-cell co-stimulator (ICOS) p.L96Sfs mutation. The predicted crystal models indicated premature truncation of the two mutated proteins. Both variants were demonstrated as loss-of-function mutations and were associated with overlapped manifestations of respiratory fungal infection and splenomegaly. We further performed a detailed assessment of immunologic phenotypes and impaired lymphocyte functions in patients. Moreover, we discovered an association between monoclonal T-large granular lymphocyte proliferation and ICOS-deficient CVID for the first time. These observations lead to a new perspective on the underlying genetic heterogeneity of CVID.

    Abstract: Heading date (flowering time), which greatly influences regional and seasonal adaptability in rice (Oryza sativa), is regulated by many genes in different photoperiod pathways. Here, we characterized a heading date gene, Early heading date 5 (Ehd5), using a modified bulked segregant analysis method. The ehd5 mutant showed late flowering under both short-day and long-day conditions, as well as reduced yield, compared to the wild type. Ehd5, which encodes a WD40 domain-containing protein, is induced by light and follows a circadian rhythm expression pattern. Transcriptome analysis revealed that Ehd5 acts upstream of the flowering genes Early heading date 1 (Ehd1), RICE FLOWERING LOCUS T 1 (RFT1), and Heading date 3a (Hd3a). Functional analysis showed that Ehd5 directly interacts with Rice outermost cell-specific gene 4 (Roc4) and Grain number, plant height, and heading date 8 (Ghd8), which might affect the formation of Ghd7–Ghd8 complexes, resulting in increased expression of Ehd1, Hd3a, and RFT1. In a nutshell, these results demonstrate that Ehd5 functions as a positive regulator of rice flowering and provide insight into the molecular mechanisms underlying heading date.

    Abstract: Grasslands are globally threatened by climate changes and unsustainable land-use, which often cause transitions among alternative stable states, and even catastrophic transition to desertification. Spatial vegetation patch configurations have been shown to signify such transitions at large spatial scale. Here, we demonstrate how small-scale patch configurations can also indicate state transitions. Methods The whole spatial series of degradation successions were chosen in alpine grasslands characterized as seven typical communities. Patch numbers, and perimeter and cover of each patch were recorded using adjacent quadrats along transects in each type of the communities. Species abundance within each patch was measured. Important Findings Across seven grazing-induced degradation stages in the world’s largest expanse of grassland, from dense ungrazed turf to bare black-soil crust, patch numbers and perimeters first increased as patch cover decreased. Numbers and perimeters then decreased rapidly beyond an intersection point at 68% of initial continuous vegetation cover. Around this point, the vegetation fluctuated back and forth between the sedge-dominated grassland breaking-up phase and the forb-dominated phase, suggesting impending shift of grassland state. This study thus demonstrates how ground-based small-scale vegetation surveys can provide a quantitative, easy-to-use signals for vegetation degradation, with promise for detecting the catastrophic transition to desertification.

    Abstract: Grain yield and grain quality are major determinants in modern breeding controlled by many quantitative traits loci (QTLs) in rice (Oryza sativa). However, the mechanisms underlying grain shape and quality are poorly understood. Here, we characterize a QTL for grain size and grain quality via map-based cloning from wild rice (W1943), GS10 (Grain Size on Chromosome 10), which encodes a protein with 6 tandem armadillo repeats. The null mutant gs10 shows slender and narrow grains with altered cell size, which has a pleiotropic effect on other agronomical traits. Functional analysis reveals that GS10 interacts with TUD1 (Taihu Dwarf1) and is epistatic to OsGSK2 (glycogen synthase kinase 2) through regulating grain shape and lamina joint inclination, indicating it is negatively involved in brassinosteroid (BR) signaling. Pyramiding gs10 and the grain size gene GW5 into cultivar GLA4 substantially improved grain shape and appearance quality. Natural variation analysis revealed that gs10 from the wild rice Oryza rufipogon W1943 is a rare allele across the rice population. Collectively, these findings advance our understanding of the underlying mechanism of grain shape and provide the beneficial allele of gs10 for future rice breeding and genetic improvement.

    Abstract: Ticagrelor has been shown to reduce the risk of pneumonia and improve lung function, but the findings across studies were inconsistent. The objective is to investigate the relative safety of ticagrelor vs. clopidogrel on infection outcomes in patients with cardiovascular diseases. Methods and results We searched MEDLINE, Embase, Cochrane Library, and ClinicalTrials.gov up to 15 October 2019. Randomized controlled trials comparing ticagrelor and clopidogrel that reported infection outcomes were included. The primary outcome was pneumonia. Secondary outcomes were upper respiratory tract infection (URTI), urinary tract infection (UTI), and sepsis. Study quality was assessed using the Cochrane Risk of Bias tool. Study selection, data extraction, and quality assessment were conducted by independent authors. Random-effects model was used for data synthesis. Relative risks (RRs) and 95% confidence intervals (CIs) were pooled with a random-effects model. Out of 5231 citations, 10 trials with altogether 37 514 patients were included. Ticagrelor was associated with a lower risk of pneumonia (RR 0.80, 95% CI 0.67–0.95) compared to clopidogrel. There were no statistically significant differences for URTI (RR 0.71, 95% CI 0.34–1.48), UTI (RR 1.06, 95% CI 0.73–1.64), or sepsis (RR 0.79, 95% CI 0.50–1.26). Conclusion Compared to clopidogrel, ticagrelor reduces the risk of pneumonia, but not URTI, UTI, or sepsis. Our study provides further evidence for recommending ticagrelor to patients with acute coronary syndrome at risk of pneumonia, although the mechanism by which ticagrelor reduces the risk of pneumonia merits further research.

    Abstract: Manganese oxide nanofibers with layered structure and γ-MnOOH structure have been prepared from a layered manganese oxide by using a self-assembling hydrothermal process. This process includes preparation of manganese oxide nanosheets by exfoliating the layered manganese oxide, self-assembling the nanosheets with a cationic surfactant into a nanocomposite, and transforming the nanocomposite into the nanofibers under hydrothermal conditions.

    Abstract: Wild rice (Oryza rufipogon) has a lower panicle seed setting rate (PSSR) and gamete fertility than domesticated rice (Oryza sativa), but the genetic mechanisms of this phenomenon remain unknown. Here, we cloned a null allele of OsMLH1, an ortholog of MutL-homolog 1 to yeast and mammals, from wild rice O. rufipogon W1943 and revealed a 5.4-kb retrotransposon insertion in OsMLH1 is responsible for the low PSSR in wild rice. In contrast to the wild-type, a near isogenic line NIL-mlh1 exhibits defective crossover (CO) formation during meiosis, resulting in reduced pollen viability, partial embryo lethality, and low PSSR. Except for the mutant of mismatch repair gene postmeiotic segregation 1 (Ospms1), all other MutL mutants from O. sativa indica subspecies displayed male and female semi-sterility similar to NIL-mlh1, but less severe than those from O. sativa japonica subspecies. MLH1 and MLH3 did not contribute in an additive fashion to fertility. Two types of MutL heterodimers, MLH1-PMS1 and MLH1-MLH3, were identified in rice, but only the latter functions in promoting meiotic CO formation. Compared to japonica varieties, indica cultivars had greater numbers of CO events per meiosis. Our results suggest that low fertility in wild rice may be caused by different gene defects, and indica and japonica subspecies have substantially different CO rates responsible for the discrepancy between the fertility of mlh1 and mlh3 mutants.

    Abstract: Rad52 is a DNA-binding protein that stimulates the annealing of complementary single-stranded DNA. Only the N terminus of Rad52 is evolutionarily conserved; it contains the core activity of the protein, including its DNA-binding activity. To identify amino acid residues that are important for Rad52 function(s), we systematically replaced 76 of 165 amino acid residues in the N terminus with alanine. These substitutions were examined for their effects on the repair of γ-ray-induced DNA damage and on both interchromosomal and direct repeat heteroallelic recombination. This analysis identified five regions that are required for efficient γ-ray damage repair or mitotic recombination. Two regions, I and II, also contain the classic mutations, rad52-2 and rad52-1, respectively. Interestingly, four of the five regions contain mutations that impair the ability to repair γ-ray-induced DNA damage yet still allow mitotic recombinants to be produced at rates that are similar to or higher than those obtained with wild-type strains. In addition, a new class of separation-of-function mutation that is only partially deficient in the repair of γ-ray damage, but exhibits decreased mitotic recombination similar to rad52 null strains, was identified. These results suggest that Rad52 protein acts differently on lesions that occur spontaneously during the cell cycle than on those induced by γ-irradiation.
