    Abstract: High levels of starch is known to have positive effects on both energy supply and milk yield but increases the risk of rumen acidosis. The use of sugar as a non-structural carbohydrate could circumvent this risk while maintaining the benefits, but its effects and that of the simultaneous use of both sugar and starch are not as well-understood. This study aimed to evaluate the effects of different combinations of sugar and starch concentrations on ruminal fermentation and bacterial community composition in vitro in a 4 ×4 factorial experiment. Sixteen dietary treatments were formulated with 4 levels of sugar (6, 8, 10, and 12% of dietary dry matter), and 4 levels of starch (21, 23, 25, and 27% of dietary dry matter). Samples were taken at 0.5, 1, 3, 6, 12, and 24 h after cultivation to determine the disappearance rate of dry matter, rumen fermentation parameters and bacterial community composition. Butyric acid, gas production, and Treponema abundance were significantly influenced by the sugar level. The pH, acetic acid, and propionic acid levels were significantly influenced by starch levels. However, the interactive effect of sugar and starch was only observed on the rate of dry matter disappearance. Furthermore, different combinations of starch and sugar had different effects on volatile fatty acid production rate, gas production rate, and dry matter disappearance rate. The production rate of rumen fermentation parameters in the high sugar group was higher. Additionally, increasing the sugar content in the diet did not change the main phylum composition in the rumen, but significantly increased the relative abundance of Bacteroidetes and Firmicutes phyla, while the relative abundance of Proteobacteria was reduced. At the genus level, the high glucose group showed significantly higher relative abundance of Treponema ( P & lt; 0.05) and significantly lower relative abundance of Ruminobacter, Ruminococcus, and Streptococcus ( P & lt; 0.05). In conclusion, different combinations of sugar and starch concentrations have inconsistent effects on rumen fermentation characteristics, suggesting that the starch in diets cannot be simply replaced with sugar; the combined effects of sugar and starch should be considered to improve the feed utilization rate.

    Abstract: This study aimed to establish a mathematical survival model database containing cell-specific coefficients from human digestive system cells exposed to electroporation pulses (EPs). Materials and methods A total of 20 types of human digestive system cell lines were selected to investigate the effect of EPs on cell viability. Cell viability was measured after exposure to various pulse settings, and a cell survival model was established using the Peleg–Fermi model. Next, the cell-specific coefficients of each cell line were determined. Results Cell viability tended to decrease when exposed to stronger electric field strength (EFS), longer pulse duration, and more pulse number, but the decreasing tendency varied among different cell lines. When exposed to a lower EFS ( & lt;1,000 V/cm), only a slight decrease in cell viability occurred. All cell lines showed a similar tendency: the extent of electrical injury (EI) increased with the increase in pulse number and duration. However, there existed differences in heat sensitivity among organs. Conclusions This database can be used for the application of electroporation-based treatment (EBT) in the digestive system to predict cell survival and tissue injury distribution during the treatment.

    Abstract: The change in temperature will change the composition of intestinal microorganisms of juvenile Eriocheir sinensis , and the composition of intestinal microorganisms will affect the growth and development of juvenile crabs. In order to explore the relationship between intestinal microorganisms and growth of E. sinensis at different temperatures, the status of growth and intestinal microflora of juvenile E. sinensis reared at different water temperatures (15 °C, 23 °C, and 30 °C) were compared in this study. The results showed that the respective survival rate of juvenile E. sinensis in the three water temperature groups was 100%, 87.5%, and 64.44%. Moreover, the molting rate increased with an increase in water temperature, which was at 0%, 10%, and 71.11% for the three respective temperature groups. The average weight gain rate showed an overall increasing trend with the increase of water temperature. Moreover, the final fatness of the crabs in the 30 °C water temperature group was significantly lower than that in the 15 °C and 23 °C groups ( p & lt; 0.05); there was no significant difference in the liver-to-body ratio among the three groups. The results of the alpha diversity analysis of the 16S rRNA data revealed that there was no significant difference in the intestinal microbial abundance among the three water temperature groups; however, the intestinal microbial diversity in the 23 °C water temperature group was significantly lower than that in the 15 °C and 30 °C groups. At the phylum level, the dominant flora of the three groups was Firmicutes, Proteobacteria, and Bacteroidota. At the genus level, the abundance of Parabacteroides and Aeromonas in the intestine of the crabs in the 30 °C water temperature group was significantly higher than that in the 15 °C and 23 °C groups ( p & lt; 0.05). The function prediction showed that the main functional diversity of intestinal microflora of juvenile E. sinensis in the three water temperature groups was similar and mainly involved in metabolic-related functions, but there were still differences in the effects of water temperature on functional pathways such as metabolism, immunity, and growth among each group, either promoting or inhibiting. In conclusion, different water temperatures can affect the composition and function of intestinal flora of E. sinensis , and 23 °C–30 °C is the optimal water temperature for the growth of juvenile E. sinensis .

    Abstract: To provide basic drift-ice parameters for new-energy structures in ice-infested nearshore waters, an estimation method of the design drift-ice concentration and floe area was established. A total of 700 satellite images in winters from 2010 to 2021 were used to obtain the drift-ice concentration and floe area from 26 sites in the Bohai Sea. The floe area with 90% cumulative probability was selected as the characteristic floe area in each satellite image. Furthermore, the water temperature threshold ranging from −1.6 °C to −0.5 °C was adopted by considering the drift-ice freezing/melting states because of different salinity values in nearshore waters. The upper limit values of the ice concentration and characteristic floe area at the lowest water temperature of −1.6 °C were finally used as the design drift-ice indexes. The maximum design ice concentrations (floe area) in the Liaodong Bay, Bohai Bay, and Laizhou Bay were 96% (0.43 km 2 ), 91% (0.39 km 2 ), and 87% (0.29 km 2 ), respectively. The design ice concentration at the bay bottom was higher than that near the mouth. The design floe area in the Liaodong Bay and Bohai Bay increased with latitudes, but the trend was opposite in the Laizhou Bay. The method developed in the current study has universal applicability, which can be extended to other ice-cover waters and specific structure designs by adjusting the cumulative probability threshold and the water temperature threshold.

    Abstract: Demographic information has been shown to help predict high antibody titers of COVID-19 convalescent plasma (CCP) in CCP donors. However, there is no research on the Chinese population and little evidence on whole-blood donors. Therefore, we aimed to investigate these associations among Chinese blood donors after SARS-CoV-2 infection. Methods In this cross-sectional study, 5,064 qualified blood donors with confirmed or suspected SARS-CoV-2 infection completed a self-reported questionnaire and underwent tests of SARS-CoV-2 Immunoglobulin G (IgG) antibody and ABO blood type. Logistic regression models were used to calculate odds ratios (ORs) for high SARS-CoV-2 IgG titers according to each factor. Results Totally, 1,799 participants (with SARS-CoV-2 IgG titers≥1:160) had high-titer CCPs. Multivariable analysis showed that a 10-year increment in age and earlier donation were associated with higher odds of high-titer CCP, while medical personnel was associated with lower odds. The ORs (95% CIs) of high-titer CCP were 1.17 (1.10–1.23, p & lt; 0.001) and 1.41 (1.25-1.58, p & lt; 0.001) for each 10-year increment in age and earlier donation, respectively. The OR of high-titer CCP was 0.75 (0.60-0.95, p = 0.02) for medical personnel. Female early donors were associated with increased odds of high-titer CCP, but this association was insignificant for later donors. Donating after 8 weeks from the onset was associated with decreased odds of having high-titer CCP compared to donating within 8 weeks from the onset, and the HR was 0.38 (95% CI: 0.22-0.64, p & lt;0.001). There was no significant association between ABO blood type or race and the odds of high-titer CCP. Discussion Older age, earlier donation, female early donors, and non-medical-related occupations are promising predictors of high-titer CCP in Chinese blood donors. Our findings highlight the importance of CCP screening at the early stage of the pandemic.

    Abstract: Cadmium (Cd) is a major heavy metal toxicant found in industrial zones. Humans and animals are exposed to it through their diet, which results in various physiological problems. In the current study, the toxic effects of Cd on the liver were investigated by whole-transcriptome sequencing (RNA-seq) of the livers of Xiangxi heifers fed a diet with excess Cd. We randomly divided six healthy heifers into two groups. The first group received a control diet, whereas the second group received Cd-exceeding diets for 100 days. After 100 days, the livers were collected. A total of 551 differentially expressed mRNAs, 24 differentially expressed miRNAs, and 169 differentially expressed lncRNAs were identified ( p & lt; 0.05, |log2FC| & gt;1). Differentially expressed genes (DEGs) were analyzed by gene ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses. We found that under Cd exposure, DEGs were enriched in the adenosine 5'-monophosphate–activated protein kinase pathway, which is involved in autophagy regulation, and the peroxisome proliferator–activated receptor pathway, which is involved in lipid metabolism. In addition, the apolipoprotein A4 gene, which has anti-inflammatory and antioxidant effects, the anti-apoptotic gene ATPase H + /K + transporting the nongastric alpha2 subunit, and the cholesterol metabolism–associated gene endothelial lipase gene were significantly downregulated. C–X–C motif chemokine ligand 3, cholesterol 7α-hydroxylase, and stearoyl-CoA desaturase, which are involved in the development of fatty liver, were significantly upregulated. These genes revealed the main effects of Cd on the liver of Xiangxi yellow heifers. The current study provides insightful information regarding the DEGs involved in autophagy regulation, apoptosis, lipid metabolism, anti-inflammation, and antioxidant enzyme activity. These may serve as useful biomarkers for predicting and treating Cd-related diseases in the future.

    Abstract: Rhizophagus intraradices , one of the common arbuscular mycorrhizal fungi (AMF) grown in the roots of Flaveria bidentis , facilitates the invasion of this exotic plant species into China. However, it is still unknown whether nutrient transfer through the common mycorrhizal networks (CMN) between this exotic species and the native species enhances the competitive growth of F. bidentis over the native species. To elucidate this question and the related mechanism, an isotopic labeling technique was used to test the transfer of carbon (C) and nitrogen (N) by CMN. Native species like Setaria viridis and Eclipta prostrata were selected to compete with F. bidentis in a polyvinyl chloride (PVC) box. Two competitive groups ( F. bidentis - S. viridis and F. bidentis - E. prostrata ), three treatments (monoculture of F. bidentis , the mixture of F. bidentis and the native plant, and the monoculture of the native plant), and two levels of AMF (presence or absence) were assigned. Results showed that the corrected index of relative competition intensity (CRCI) of F. bidentis in the presence of AMF & lt; 0 suggests that the competition facilitated the growth of F. bidentis with either S. viridis or E. prostrata . The reason was that the inoculation of R. intra radices significantly increased the C and N contents of F. bidentis in the mixtures. However, the effects of R. intra radices on the two native species were different: negative effect on the growth of S. viridis and positive effect on the growth of E. prostrata . The change of N content in S. viridis or E. prostrata was consistent with the variation of the biomass, suggesting that the N uptake explains the effects of R. intraradices on the competitive interactions between F. bidentis and the two native species. Moreover, the transfer of C and N via AMF hyphal links did occur between F. bidentis and the native species. However, the transfer of C and N by the CMN was not positively related to the competitive growth of F. bidentis .

    Abstract: Many microbial pathogens have evolved a range of capabilities to evade host immune defense mechanisms and to survive and multiply in host cells. The presence of host intracellular bacteria makes it difficult for specific antibodies to function. After the intracellular bacteria escape the attack of the innate immune system, such as phagocytes, they survive in cells, and then adaptive immunity comes into play. Cytotoxic T lymphocytes (CTLs) play an important role in eliminating intracellular bacteria. The regulation of key transcription factors could promote CD4 + /CD8 + T cells to acquire cytolytic ability. The TCR-CD3 complex transduces activation signals generated by TCR recognition of antigen and promotes CTLs to generate multiple pathways to kill intracellular bacteria. In this review, the mechanism of CD4/CD8 CTLs differentiation and how CD4/CD8 CTLs kill intracellular bacteria are introduced. In addition, their application and prospects in the treatment of bacterial infections are discussed.
