    Abstract: Varicella–zoster virus (VZV) is the causative agent of varicella and herpes zoster (HZ) and can pose a significant challenge to human health globally. The initial VZV infection—more common in children—causes a self-limiting chicken pox. However, in later life, the latent VZV can become reactivated in these patients, causing HZ and postherpetic neuralgia (PHN), a serious and painful complication. VZV glycoprotein E (gE) has been developed into a licensed subunit vaccine against HZ (Shingrix). However, its efficacy relies on the concomitant delivery of a robust adjuvant (AS01B). Here, we sought to create a new immunogen for vaccine design by displaying the VZV–gE on the baculovirus surface (Bac–gE). Correct localization and display of gE on the engineered baculovirus was verified by flow cytometry and immune electron microscopy. We show that Bac–gE provides excellent antigenicity against VZV and induces not only stronger gE-specific CD4+ and CD8+ T cell responses but also higher levels of VZV–specific neutralizing antibodies as compared with other vaccine strategies in mice. Collectively, we show that the baculovirus display of VZV–gE confers ideal humoral and cellular immune responses required for HZ vaccine development, paving the way for a baculovirus-based vaccine design.

    Abstract: Background: Aneurysmal subarachnoid hemorrhage (aSAH) causes long-term functional dependence and death. Early prediction of functional outcomes in aSAH patients with appropriate intervention strategies could lower the risk of poor prognosis. Therefore, we aimed to develop pre- and post-operative dynamic visualization nomograms to predict the 1-year functional outcomes of aSAH patients undergoing coil embolization. Methods: Data were obtained from 400 aSAH patients undergoing endovascular coiling admitted to the People’s Hospital of Hunan Province in China (2015–2019). The key indicator was the modified Rankin Score (mRS), with 3–6 representing poor functional outcomes. Multivariate logistic regression (MLR)-based visual nomograms were developed to analyze baseline characteristics and post-operative complications. The evaluation of nomogram performance included discrimination (measured by C statistic), calibration (measured by the Hosmer–Lemeshow test and calibration curves), and clinical usefulness (measured by decision curve analysis). Results: Fifty-nine aSAH patients (14.8%) had poor outcomes. Both nomograms showed good discrimination, and the post-operative nomogram demonstrated superior discrimination to the pre-operative nomogram with a C statistic of 0.895 (95% CI: 0.844–0.945) vs. 0.801 (95% CI: 0.733–0.870). Each was well calibrated with a Hosmer–Lemeshow p-value of 0.498 vs. 0.276. Moreover, decision curve analysis showed that both nomograms were clinically useful, and the post-operative nomogram generated more net benefit than the pre-operative nomogram. Web-based online calculators have been developed to greatly improve the efficiency of clinical applications. Conclusions: Pre- and post-operative dynamic nomograms could support pre-operative treatment decisions and post-operative management in aSAH patients, respectively. Moreover, this study indicates that integrating post-operative variables into the nomogram enhanced prediction accuracy for the poor outcome of aSAH patients.

    Abstract: Liriodendron hybrid (L. chinense × L. tulipifera), an essential medium-sized tree generally famous for its timber, is also used as an ornamental and greenery tool in many places around the world. The Liriodendron hybrid (L. hybrid) tree goes through many hurdles to achieve its maximum strength and vigor, such as loss of habitat, vast genetic variation, and low seed setting rate. The establishment of an effective and well-organized somatic embryogenesis (S.E.) system could be used to overcome these obstacles, rather than the old-fashioned seed culture and organogenesis. This study is based on the impact of chitosan oligosaccharide (COS) and its role in the induction of S.E. on the callus of four genotypes of the L. hybrid. The optimal concentration of COS could enhance the momentum and effectiveness in S.E.’s mechanism, which further improves the growth rate of the L. hybrid tree’s plantlets. This study shows that COS has a prominent role in endogenous hormones like indole acetic acid (IAA), zeatin (Z.T.), and gibberellic acid (GA3). Furthermore, COS improves the growth development, growth speed, as well as the development situation of plant germination ability. COS can also regulate branch development and root growth, which could be linked to the antagonistic effect on growth factors to some extent or by affecting auxin synthesis and polar transport.

    Abstract: Achieving sustainable development is an issue of global concern. Accounting for the gross ecosystem product (GEP) value can specifically quantify the value of ecosystems for people, which is conducive to the formulation of sustainable eco-management decisions. Multi-source data, including remote sensing images, geospatial data, and statistical bulletin information, were used to quantify the GEP value of material products, regulating services, and cultural services for Fujian Province, China, during 2000–2020. On this basis, the spatio-temporal characteristics of GEP and the coupling relationship between GEP and GDP were analyzed. The results showed that: (1) the value of GEP in Fujian Province increased by 27.9% from CNY 3589.04 billion in 2000 to CNY 4590.25 billion in 2020. Among the service values, the contribution rate of regulating services to GEP was always the highest during the study period. (2) The spatial distribution pattern of GEP in Fujian Province was higher in the west and lower in the east. Comparing prefecture-level cities, Nanping maintained its GEP at the maximum value level over the past 21 years, while Xiamen and Putian maintained their GEP at the minimum value level. (3) GDP grew faster relative to GEP over the past 21 years, and the difference between GEP and GDP decreased. GEP had a long-term positive effect on GDP, while GDP had a smaller effect on GEP in the short term. The research was not only enriched in relation to GEP accounting, but also the policy recommendations for improving the mechanisms related to the optimization of sustainable development goals have some practical significance.

    Abstract: Pulmonary emphysema is a fatal lung disease caused by the progressive thinning, enlargement and destruction of alveoli that is closely related to inflammation and oxidative stress. Oxymatrine (OMT), as a bioactive constituent of traditional Chinese herbal Sophora flavescens, has great potential to alleviate pulmonary emphysema via its anti-inflammatory and antioxidative activities. Pulmonary administration is the most preferable way for the treatment of lung diseases. To improve the in vivo stability and pulmonary retention of OMT, OMT-loaded liposome with carboxymethyl chitosan (CMCS) modification was developed. The CMCS was modified on the surface of OMT liposomes via electrostatic attraction and covalent conjugation to obtain Lipo/OMT@CMCS and CMCS-Lipo/OMT, respectively. A porcine pancreatic elastase (PPE)-induced emphysema mice model was established to evaluate the alleviation effects of OMT on alveolar expansion and destruction. CMCS-modified liposomal OMT exhibited superior ameliorative effects on emphysema regardless of the preparation methods, and higher sedimentation and longer retention in the lung were observed in the CMCS-Lipo group. The mechanisms of OMT on emphysema were related to the downregulation of inflammatory cytokines and the rebalancing of antioxidant/oxidation via the Nrf2/HO-1 and NF-κB/IκB-α signaling pathways, leading to reduced cell apoptosis. Moreover, the OMT liposomal preparations further enhanced its anti-inflammatory and antioxidative effects. In conclusion, pulmonary administration of OMT is a potential strategy for the treatment of emphysema and the therapeutic effects can be further improved by CMCS-modified liposomes.

    Abstract: Poly (ADP-ribose) polymerase-1 (PARP-1) plays critical roles in many biological processes and is considered as a potential target for anticancer therapy. Although some PARP-1 inhibitors have been reported, their clinical application in cancer therapy is limited by some shortcomings such as weak affinity, low selectivity and adverse side effects. To identify highly potent and selective PARP-1 inhibitors, an integrated protocol that combines pharmacophore mapping, virtual screening and molecular docking was constructed. It was then used as a screening query to identify potent leads with unknown scaffolds from an in-house database. Finally, four retrieved compounds were selected for biological evaluation. Biological testing indicated that the four compounds showed strong inhibitory activities on the PARP-1 (IC50 〈 0.2 μM). MTT assay confirmed that compounds 1–4 inhibited the growth of human lung cancer A549 cells in a dose-dependent manner. The obtained compounds from this study may be potential leads for PARP-1 inhibition in the treatment of cancer.

    Abstract: H2S is a poisonous gas that is widespread in nature and human activities. Its rapid and sensitive detection is essential to prevent it from damaging health. Herein, we report Pd- and Pt-bimetallic-nanoparticle-doped In2O3 hollow microspheres that are synthesized using solvothermal and in situ reduction methods for H2S detection. The structure of as-synthesized 1 at% Pd/Pt-In2O3 comprises porous hollow microspheres assembled from In2O3 nanosheets with Pd and Pt bimetallic nanoparticles loaded on its surface. The response of 1 at% Pd/Pt-In2O3 to 5 ppm H2S is 140 (70 times that of pure In2O3), and the response time is 3 s at a low temperature of 50 °C. In addition, it can detect trace H2S (as low as 50 ppb) and has superior selectivity and an excellent anti-interference ability. These outstanding gas-sensing performances of 1 at% Pd/Pt-In2O3 are attributed to the chemical sensitization of Pt, the electronic sensitization of Pd, and the synergistic effect between them. This work supplements the research of In2O3-based H2S sensors and proves that Pd- and Pt-bimetallic-doped In2O3 can be applied in the detection of H2S.

    Abstract: Herein, gold–nitrogen-doped graphene quantum dot (Au/NGQD) composite modified electrodes were fabricated and applied as “on–off” nanosensors for the photo-electrochemical (PEC) detection of caffeic acid under visible-light irradiation. An effective and simple strategy was established for the preparation of Au/NGQD composites by hydrothermal and calcination methods. Owing to the quantum confinement effect of NGQDs, the local surface plasmon resonance (LSPR) effect of Au nanoparticles (NPs), and the synergistic effect between Au and NGQDs NPs, the Au/NGQDs showed excellent PEC performance, with wide linear concentration ranges (0.11 to 30.25 μM and 30.25 to 280.25 μM), a low detection limit (0.03 μM), excellent sensitivity, and high stability. The present study may provide an advanced strategy for the simple design of Au/NGQD composites to allow their effective application for selective and sensitive sensing of small biological molecules.
