    Notes: Riassunto Cellule portatrici di virus derivate da adenocarcinoma della ghiandola salivare coltivate in vitro sono oncogeniche per l'ospite isogenico per circa sei mesi di coltura. Dopo tale periodo perdono l'oncogenicità e non è più possibile ritrovare particelle virali. Sulla base di questi risultati si conclude che il virus è l'agente causale dell'adenocarcinoma della ghiandola salivare del topo C3H/He.

    Description / Table of Contents: Zusammenfassung Die Onkogenität von Zellen des C3H/He-Speicheldrüsen-Adenocarcinoms der Maus in Gewebekulturen ist eng verknüpft mit der Anwesenheit von Virus-partikeln (Bittner). Sowohl in quantitativer wie in qualitativer Hinsicht ist das Wachstum der implantierten Tumorzellen ähnlich dem in vivo beobachteten, jedoch vollkommen verschieden von dem des ebenfalls durch das Bittner-Virus hervorgerufenen Mammatumors. Nach 180 Tagen verschwinden die Viruspartikel aus den Zellen, wonach sie nicht mehr onkogen sind, wenn sie in isogene Wirtstiere transplantiert werden. Die frühen Phasen der Tumorzellen in der Gewebekultur sind durch epitheliale Beschaffenheit und Bildung von Pseudotubuli charakterisiert. Dieses Verhalten ändert sich auch nicht beim Altern der Kulturen, doch wurde gelegentlich eine pseudo-fibroblastische Umwandlung bei sehr alten Kulturen beobachtet. Dieser Vorgang ist durch eine „eosinophile Dengeneration” gekennzeichnet. In der Evolution des Tumors in vitro scheint eine Beziehung zwischen Verschwinden der Viruspartikel, dem Verlust der Onkogenität und dem Auftreten von eosinophilen Körperchen zu bestehen.
    Notes: Summary The oncogenicity of the cultivated cells of C3H/He mouse salivary gland adenocarcinoma is strictly correlated with the presence of the viral particles (Bittner). From both a quantitative and a qualitative standpoint the growth of the implanted tumor is similar to that observed in vivo and completely different from the mammary gland tumor which is also produced by Bittner virus. After 180 days the viral particles disappear from the cell and thereafter the cells are no longer oncogenic when implanted in isogeneic hosts. The early phases of culture are characterized by the epithelial feature of the tumor with a pseudotubular pattern. This feature is not modified with aging of the cultures, however a pseudo-flbroblastic transformation has occasionally been observed in very old samples. This step of evolution is characterized by “eosinophilic degeneration”. In the evolution of in vitro-tumor a relationship appears to exist between the disappearance of virus particles, the loss of oncogenicity and the presence of eosinophilic bodies.

    Notes: Summary Influenza NS2 protein was expressed inSaccharomyces cerevisiae using a copper-inducible promoter. The protein produced had a molecular weight of 13 kDa, was reactive with anti-NS2 antiserum and was localised to the yeast cell nucleus. Two-hybrid analysis identified a direct protein-protein interaction between NS2 and the M2 protein of the virus, involving the C-terminal 163 residues of M1. A filter-binding assay localised the M1 binding region to the C-terminal 70 amino acids of NS2.

    Notes: Summary A convenient shuttle vector that enables high level secretion of proteins from Kluyveromyces lactis has been developed. The vector, pEPS1, contains a unique cloning site that allows the construction, in a single ligation step, of episomal plasmids capable of directing secretion of foreign gene products from K. lactis. As an example we demonstrate the production of β-lactamase and determine optimal conditions for its secretion into the culture media.

    Notes: Summary Sequences encoding GST-fusion proteins were cloned into the Saccharomyces cerevisiae secretion vector, pYEX-S1, to direct secretion into the culture medium. GST and metallothionein fused to GST were secreted successfully and the fusion proteins purified. With several other GST-fusion proteins however, the proteins were retained inside the cell, indicating limitations to the types of proteins that can be secreted from yeast.

    Notes: Apoptosis seems to play an important role in the decline of CD4+ T-cells in patients infected with HIV-1. Moreover, extensive interest in apoptosis comes from the observation that it correlates both with the progression and the severity of HIV-1 infection. A cross-sectional study was made to evaluate whether such correlation may also extend to the early phases of ex vivo apoptosis, after 20 h of culture. DNA fragmentation, a parameter associated with apoptosis, was evaluated with the terminal deoxynucleotidyl-transferase-mediated dUTP nick end labelling (TUNEL) technique, which preferentially labels apoptosis in comparison to necrosis. The results obtained indicate that a negative correlation exists between the proportion of lymphocytes exhibiting DNA strand breaks and the absolute number of CD4+ T-cells per &μl. DNA fragmentation was significantly higher in patients with AIDS or advanced HIV-1 infection as compared to asymptomatic patients or seronegative individuals. No significant difference was found in relation to antiretroviral therapy. Furthermore, the addition of nicotinamide to the cultures significantly reduced DNA fragmentation of both in vitro HIV-1-infected MT-4 cells and lymphocytes from six HIV-1-seropositive individuals. The results of this study confirm that DNA fragmentation, as an early marker of apoptosis, correlates with the severity of HIV-1 infection and suggest that nicotinamide may be involved in the modulation of HIV-1-related apoptosis. © 1997 John Wiley & Sons, Ltd.
