    Abstract: Background: Podocyte infolding glomerulopathy (PIG) is a peculiar and very rare manifestation in renal pathology. Its underlying pathogenesis mechanism and clinical characteristics remain unclear due to sparse reports. Objective: To further elucidate the clinical profile of PIG by carefully reporting our four cases and a comprehensive review of cases in the literature. Methods: This study retrospectively reviewed four cases of PIG from 2010 to 2022 in our centre. Clinical and pathological profiles were reported. PIG cases in the literature were searched in the MEDLINE database and analysed together with our cases. Results: Four cases of PIG identified from our centre and 40 cases from the current literature were reported. The pooled analysis of these 44 cases indicated 79.5% (35/44) were females, 93.2% (41/44) were East Asians, and 63.6% (28/44) were reported in Japan. The average age was 42.0 ± 12.5 years old. The average amount of proteinuria at the time of renal biopsy was 3.06 ± 3.2 g/day. The most reported comorbidities were connective tissue diseases, mainly systemic lupus erythematosus, and 20.5% (9/44) of the cases did not have any contaminant disease. Most of the cases (81.8%, 36/44) had been treated with immunosuppressants, of which a combination of corticosteroids and one other type of immunosuppressant was most commonly reported. In addition, 45.4% (20/44) and 34.1% (15/44) of the cases had achieved complete response and partial response, respectively, after treatment. Whole exosome sequencing indicated mutations in the INF2 gene. Conclusions: PIG is a rare condition and seen in relatively younger populations, often associated with connective tissue diseases clinically and one or two other glomerulopathies histologically. The outcomes following immunosuppressive treatment are relatively good. Mutations in INF2 might be involved in the development of PIG; however, the implications of these results need to be investigated.

    Abstract: Encouraging advances have been made in the application of microbial mineralization towards fixing and improving desertified sandy soils. However, desert soils in arid areas exhibit high salinity that may limit urease activity and production in microbial strains, thereby affecting the solidification effects of microbial calcium binders in saline soils. In this study, a salt-tolerant microbial strain (A80) that produced urease was identified from saline soils of the Qaidam Basin. The culture conditions of the strain were optimized using single-factor tests and response surface methods to optimize urease yields and activity. The optimal composition of the A80 medium included an inoculation amount of 6.32% (V/V), a yeast extract powder concentration of 15.43 g/L, a glucose concentration of 5.20 g/L, a salinity of 3%, and an incubation temperature of 36 °C. Urease activity increased by 64.80% after using optimized medium. The A80 microbial calcium-cementing agent was also used to solidify saline soils, leading to an increased unconfined compressive strength of the solidified saline soil by 25.70%. Thus, the optimization method resulted in improvements in the cultivation of a salt-tolerant strain.

    Abstract: In this work, random forest (RF), support vector machine, k-nearest neighbor and C4.5 decision tree, were used to establish classification models for predicting whether an unknown molecule is an inhibitor of human topoisomerase I (Top1) protein. All these models have achieved satisfactory results, with total prediction accuracies from 89.70% to 97.12%. Through comparative analysis, it can be found that the RF model has the best forecasting effect. The parameters were further optimized to generate the best-performing RF model. At the same time, features selection was implemented to choose properties most relevant to the inhibition of Top1 from 189 molecular descriptors through a special RF procedure. Subsequently, a ligand-based virtual screening was performed from the Maybridge database by the optimal RF model and 596 hits were picked out. Then, 67 molecules with relative probability scores over 0.7 were selected based on the screening results. Next, the 67 molecules above were docked to Top1 using AutoDock Vina. Finally, six top-ranked molecules with binding energies less than −10.0 kcal/mol were screened out and a common backbone, which is entirely different from that of existing Top1 inhibitors reported in the literature, was found.

    Abstract: A tertiary lymphoid structure (TLS) is a special component in the immune microenvironment that is mainly composed of tumor-infiltrating lymphocytes (TILs), including T cells, B cells, DC cells, and high endothelial venules (HEVs). For cancer patients, evaluation of the immune microenvironment has a predictive effect on tumor biological behavior, treatment methods, and prognosis. As a result, TLSs have begun to attract the attention of researchers as a new potential biomarker. However, the composition and mechanisms of TLSs are still unclear, and clinical detection methods are still being explored. Although some meaningful results have been obtained in clinical trials, there is still a long way to go before such methods can be applied in clinical practice. However, we believe that with the continuous progress of basic research and clinical trials, TLS detection and related treatment can benefit more and more patients. In this review, we generalize the definition and composition of TLSs, summarize clinical trials involving TLSs according to treatment methods, and describe possible methods of inducing TLS formation.

    Abstract: Rocky desertification (RD), a natural and human-induced process of land degradation in karst areas, has become the primary ecological disaster and one of the obstacles to sustainable ecological development in southwest China. Nevertheless, the variation of soil physical and chemical properties, bacterial and fungal communities, and their relationships in RD forests remains limited. Therefore, soil samples were collected from forests under four degrees of RD (NRD, non-RD; LRD, light RD; MRD, moderate RD; and SRD, severe RD) and subjected to high-throughput sequencing of 16S rRNA and ITS1 genes. The results showed a significant reduction in bacterial richness and diversity, while fungal richness and diversity decreased markedly and then showed a balanced trend with the increase in RD degree, indicating that bacteria and fungi did not present the same dynamics in response to the process of RD. The bacterial communities were dominated by Proteobacteria, Actinobacteria, Acidobacteria, and Chloroflexi, while the fungal communities were dominated by Basidiomycota, Ascomycota, and Mortierellomycota. The PCoA and NMDS demonstrated significant differences in microbial communities in study sites, among which the fungal communities in non-RD forest and LRD forest clustered together, suggesting that fungal communities were more stable than bacteria in RD forest. The db-RDA, Mantel test, and random forest model confirmed the important role of soil BD, pH, SOC, AN, and AP in driving microbial diversity and communities. The IndVal analysis suggested that Chloroflexi, Patescibacteria, Atheliales, and Cantharellales with high indicator values were identified as potential bio-indicators for RD forests. This study could not only improve our understanding of bacterial and fungal community dynamics across RD gradients, but also could provide useful information for the further use of microorganisms as indicators to reflect the environmental changes and ecosystem status during forest RD.

    Abstract: Osteoarthritis (OA) is a worldwide chronic disease that can cause severe inflammation to damage the surrounding tissue and cartilage. There are many different factors that can lead to osteoarthritis, but abnormally progressed programmed cell death is one of the most important risk factors that can induce osteoarthritis. Prior studies have demonstrated that programmed cell death, including apoptosis, pyroptosis, necroptosis, ferroptosis, autophagy, and cuproptosis, has a great connection with osteoarthritis. In this paper, we review the role of different types of programmed cell death in the generation and development of OA and how the different signal pathways modulate the different cell death to regulate the development of OA. Additionally, this review provides new insights into the radical treatment of osteoarthritis rather than conservative treatment, such as anti-inflammation drugs or surgical operation.

    Abstract: Aspect-level sentiment analysis is a research focal point for natural language comprehension. An attention mechanism is a very important approach for aspect-level sentiment analysis, but it only fuses sentences from a semantic perspective and ignores grammatical information in the sentences. Graph convolutional networks (GCNs) are a better method for processing syntactic information; however, they still face problems in effectively combining semantic and syntactic information. This paper presents a sentiment-supported graph convolutional network (SSGCN). This SSGCN first obtains the semantic information of the text through aspect-aware attention and self-attention; then, a grammar mask matrix and a GCN are applied to preliminarily combine semantic information with grammatical information. Afterward, the processing of these information features is divided into three steps. To begin with, features related to the semantics and grammatical features of aspect words are extracted. The second step obtains the enhanced features of the semantic and grammatical information through sentiment support words. Finally, it concatenates the two features, thus enhancing the effectiveness of the attention mechanism formed from the combination of semantic and grammatical information. The experimental results show that compared with benchmark models, the SSGCN had an improved accuracy of 6.33–0.5%. In macro F1 evaluation, its improvement range was 11.68–0.5%.
