    Abstract: Knee osteoarthritis is a common chronic degenerative joint disease in middle-aged and elderly people. Intra-articular injection for the treatment of knee osteoarthritis is a regularly utilized nonsurgical treatment in modern medicine. Hyaluronic acid (HA) and platelet-rich plasma (PRP) are two frequently employed intra-articular devices. Hyaluronic acid (HA) is an accepted nonsurgical treatment for symptomatic KOA, and platelet-rich plasma is a popular option in the treatment of KOA in recent years. The purpose of this research is to compare the efficacy and safety of intra-articular injection of platelet-rich plasma (PRP) versus hyaluronic acid (HA) on the pain score scale, knee function, and related inflammatory biomarkers in KOA patients using a clinical randomized controlled trial. Participants are being randomized into either the hyaluronic acid (HA) or into the platelet-rich plasma (PRP) group. All patients receive 4 weeks of treatment (once a week), and well-being support and quadriceps training (3 times a week). The primary outcomes are measured using the Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC) and the visual analog scale (VAS). The secondary outcomes include the activities of daily living score, erythrocyte sedimentation rate, C-reactive protein testing, interleukin-6 levels, and X-ray examination. In order to monitor the occurrence of irregularities and abnormalities, patients are assessed at each visit, and restorative treatment is given if necessary. The results of this clinical trial will verify the efficacy of PRP and HA in the treatment of KOA and provide important evidence for the clinical treatment of KOA. The trial was enlisted at the Chinese Clinical Trial Registry on 26 September 2020 (ChiCTR2000038635).

    Abstract: In this paper, a handheld liquor authenticity detection system is demonstrated based on the laser spectroscopy technique, which consists of a handheld laser spectrometer and a mobile phone display terminal. In this system, the semiconductor laser is integrated into the spectrometer and the laser beam is further angled to the optical axis of the spectrometer to avoid interference of the fluorescence generated by the bottle wall. During the system operation, the laser excites the tested liquor to generate fluorescence and Raman spectroscopy signals, which are digitized and wirelessly transmitted by Wi-Fi to the Android mobile terminal. After the image processing by the mobile phone APP, the tested liquor spectrum curve is obtained. At the same time, based on the standard liquor spectrum curve stored in the database, the Pearson correlation coefficient is calculated and the matching similarity is given. In addition, this paper proposes a calibration method based on pure water Raman intensity to achieve accurate measurement of fluorescence intensity and minimize the influence of fluorescence intensity saturation on the measurement results. In the experiment, we measured the similarity of 12 brands of Chinese liquor by using our self-developed handheld laser spectrometer. Their authenticity of liquor could be given accurately and effectively.

    Abstract: Compared with federated avionic architecture, the integrated modular avionic (IMA) system architecture in the aircraft can provide more sophisticated and powerful avionic functionality, and meanwhile, it becomes structurally dynamic, variably interconnected, and highly complex. The traditional approach such as fault tree analysis (FTA) becomes neither convenient nor sufficient in making safety analysis of the IMA system. In order to overcome the limitations, the approach that FTA combines with generalized stochastic petri net (GSPN) is proposed. First, FTA is used to establish the static model for the top level of the IMA system, while GSPN is used to build a dynamic model for each cell system. Finally, the combination model is generated, which is called the FTGPN model. Moreover, the FTGPN model is made safety analysis with the PIPE2 tool. According to the simulation result, corresponding measures are taken to meet the safety requirements of the IMA system.

    Abstract: Lightweight Clay-EPS Soil (LCES) is a newly developed material which has many merits such as the adjustability of strength and density, simplicity for construction, and economical efficiency. It has been widely applied in practical engineering, e.g., soft ground improvement, the solvent of bridge head jump, earthfill of pipeline, and broadening of highway. Meanwhile, construction castoff and industrial waste can be recycled as a major ingredient in LCES. The dynamic deformation characteristics of LCES and clay were comprehensively studied using laboratory dynamic triaxial tests. It was found that the compressive strain of LCES increased while the growth rate of strain decreased with the increasing number of cycles, which conformed to a hyperbola model. The dynamic secant elastic modulus of LCES decreased with the increase of dynamic strain, which was represented by strain softening. The dynamic modulus of clay decreased sharply, while that of LCES decreased marginally. Moreover, the damping ratio of LCES tended to increase with the increasing dynamic strain. The damping ratio of LCES was lower than that of clay at the same strain level. It was also found that cement content had a negative relationship with the damping ratio of LCES, while the effect of EPS beads content was adverse. The damping ratio of both LCES and clay decreased moderately.

    Abstract: Outer membrane proteins (OMPs) can induce an immune response. Omp18 (HP1125) of H. pylori is a powerful antigen that can induce significant interferon- γ (IFN- γ ) levels. Previous studies have suggested that IFN- γ plays an important role in H. pylori clearance. However, H. pylori has multiple mechanisms to avoid host immune surveillance for persistent colonization. We generated an omp18 mutant ( H. pylori 26695 and H. pylori SS1) strain to examine whether Omp18 interacts with IFN- γ and is involved in H. pylori colonization. qRT-PCR revealed that IFN- γ induced Omp18 expression. qRT-PCR and western blot analysis revealed reduced expressions of virulence factors CagA and NapA in H. pylori 26695 with IFN- γ treatment, but they were induced in the Δ omp18 strain. In C57BL/6 mice infected with H. pylori SS1 and the Δ omp18 strain, the Δ omp18 strain conferred defective colonization and activated a stronger inflammatory response. Signal transducer phosphorylation and transcription 1 (STAT1) activator was downregulated by the wild-type strain but not the Δ omp18 strain in IFN- γ -treated macrophages. Furthermore, Δ omp18 strain survival rates were poor in macrophages compared to the wild-type strain. We concluded that H. pylori Omp18 has an important function influencing IFN- γ -mediated immune response to participate in persistent colonization.

    Abstract: The inflammatory storm in the early stage and immunosuppression in the late stage are responsible for the high mortality rates and multiple organ dysfunction in sepsis. In recent years, studies have found that the body’s cholinergic system can spontaneously and dynamically regulate inflammation and immunity in sepsis according to the needs of the body. Firstly, the vagus nerve senses and regulates local or systemic inflammation by means of the Cholinergic Anti-inflammatory Pathway (CAP) and activation of α7-nicotinic acetylcholine receptors (α7nAChRs); thus, α7nAChRs play important roles for the central nervous system (CNS) to modulate peripheral inflammation; secondly, the activation of muscarinic acetylcholine receptors 1 (M1AChRs) in the forebrain can affect the neurons of the Medullary Visceral Zone (MVZ), the core of CAP, to regulate systemic inflammation and immunity. Based on the critical role of these two cholinergic receptor systems in sepsis, it is necessary to collect and analyze the related findings in recent years to provide ideas for further research studies and clinical applications. By consulting the related literature, we draw some conclusions: MVZ is the primary center for the nervous system to regulate inflammation and immunity. It coordinates not only the sympathetic system and vagus system but also the autonomic nervous system and neuroendocrine system to regulate inflammation and immunity; α7nAChRs are widely expressed in immune cells, neurons, and muscle cells; the activation of α7nAChRs can suppress local and systemic inflammation; the expression of α7nAChRs represents the acute or chronic inflammatory state to a certain extent; M1AChRs are mainly expressed in the advanced centers of the brain and regulate systemic inflammation; neuroinflammation of the MVZ, hypothalamus, and forebrain induced by sepsis not only leads to their dysfunctions but also underlies the regulatory dysfunction on systemic inflammation and immunity. Correcting the neuroinflammation of these regulatory centers and adjusting the function of α7nAChRs and M1AChRs may be two key strategies for the treatment of sepsis in the future.

    Abstract: Endometrial carcinoma (EC) is a commonly diagnosed gynecological malignancy. Interleukin-6 (IL6) plays a critical role in modulating the progression of several types of tumors, including EC. However, the specific mechanism of IL6 in regulating EC progression has not been clearly elucidated. In this study, we performed a series of functional experiments to explore the potential mechanisms involved in IL6 function in the progression of EC. Here, we found that IL6 increased reactive oxygen species (ROS) generation by enhancing the NADPH oxidase (NOX) level and induced mtDNA leakage in EC cells, which further caused the activation of the downstream cGAS-STING signaling and increased production of extracellular vesicle (EV) production from EC cells. Besides, the activation of cGAS-STING signaling enhanced the expression of type I IFN and its downstream molecule PD-L1 through the TBK1-IRF3 pathway. Importantly, a high level mtDNA and PD-L1 were present in EVs derived from IL6-induced EC cells; these vesicles were shown to be able to induce T cell apoptosis. Finally, anti-PD-L1 treatment in mice showed that blockade of PD-L1 significantly reversed tumor immune escape mediated by IL6-induced EVs. Together, we provide evidence that IL6 induced mtDNA leakage to regulate the immune escape of EC cells. Our findings may provide a novel clue for the development of therapeutic targets for EC.
