    Abstract: Acute lung injury is a common complication after cardiopulmonary bypass (CPB). Oxidative damage greatly impacts CPB-induced lung ischemic pathogenesis and may represent a target for treatment. We aimed to investigate whether curcumin upregulates heme oxygenase 1 (HO-1) expression and ameliorates lung injury in a rat CPB model. Methods: A total of 80 male Sprague-Dawley rats were divided into 2 sets of 5 groups (n = 8 per group): sham; control (CPB); vehicle; low-dose curcumin (L-Cur); and high-dose curcumin (H-Cur). Animals were pretreated with a single intraperitoneal injection of vehicle, L-Cur (50 mg/kg), or H-Cur (200 mg/kg) 2 hours prior to CPB. Lung tissue, serum, and bronchoalveolar lavage fluid was harvested 2 or 24 hours postoperatively. In the control group, CPB-induced lung injury was confirmed by histopathologic examination and a significantly increased wet-to-dry lung weight ratio and pulmonary permeability index value was observed ( P 〈 .05 vs sham group). Cardiopulmonary bypass was associated with a marked rise in the level of malondialdehyde and myeloperoxidase and a fall in superoxide dismutase 2 and 24 hours after surgery ( P 〈 .05 vs sham group). Administration of curcumin ameliorated lung damage and reversed the oxidative stress markers in a partially dose-dependent manner ( P 〈 .05 vs vehicle group). Furthermore, HO-1 gene transcription and protein expression were elevated to a greater extent in the lungs after curcumin pretreatment compared with the vehicle pretreatment. Conclusions: Curcumin has the potential to provide protection from CPB-induced lung damage reflected in the expression of oxidative stress markers. The antioxidant effect of curcumin may be partly related to upregulation of HO-1.

    Abstract: Indigowoad root polysaccharide (IRPS) is a natural polysaccharide isolated from the traditional Chinese medicinal herb Radix Isatidis, and has many kinds of biological activities. However, the IRPS antiviral activity, especially the anti-porcine reproductive and respiratory syndrome virus (PRRSV) effect, has not been evaluated. Methods PRRSV was propagated in the MARC-145 cell line, and viral titre was determined by cytopathic effect and expressed as the 50% tissue culture infection dose (TCID 50 ) in the current study. The cell cytotoxic effect of IRPS toward MARC-145 was evaluated by MTT assay firstly, then the inhibitory effects of IRPS on PRRSV replication in vitro were investigated by determining the effect of IRPS upon a single replicative cycle of PRRSV in MARC- 145 cells. The effects of IRPS on viral RNA and protein synthesis in PRRSV-infected cells were investigated using real-time PCR and double-antibody (sandwich) ELISA. Results IRPS was able to effectively suppress the infectivity of the PRRSV in a dose-dependent manner, especially by adding IRPS during the PRRSV infection. IRPS could affect the attachment of PRRSV to MARC-145 cells, and also inhibit the viral RNA and protein synthesis. Conclusions IRPS has an antiviral effect on PRRSV replication in MARC-145 cells and might be useful in medical development for antiviral research. However, the precise mechanism of the host and viral targets of IRPS are unknown, so further studies should be conducted to investigate the precise mechanism of IRPS inhibitory effect on PRRSV infection.

    Abstract: A series of novel (1-arylmethyl-3-aryl-1 H-pyrazol-5-yl)(4-arylpiperazin-1-yl)methanone derivatives were synthesised. Preliminary study of the structure–activity relationship revealed that 4-chlorophenyl, 4- tert-butylphenyl, 4-fluorophe-nyl and 3-methoxyphenyl had a promising effect on the antifungal activity.

    Abstract: Locally advanced rectal adenocarcinoma is typically treated with neoadjuvant chemoradiotherapy and surgery. We assessed the effect of an additional cycle of capecitabine/oxaliplatin chemotherapy before surgery in 57 patients with T3/4, N+/- or T1/2, N+ rectal cancer. Materials and Study Design Radiotherapy (total dose, 50.4 Gy) was combined with three cycles of chemotherapy (two cycles concomitant with radiotherapy), and each cycle consisted of oxaliplatin (130 mg/m 2 on day 1) and capecitabine (825 mg/m 2 , twice per day from day 1 to day 14) for 21 days. In addition to assessing the safety of this treatment, the primary endpoint was pathological complete response (pCR). The secondary endpoint was the change in primary tumor and node stage from pre-treatment to post-surgery. Results Eleven patients (19%) experienced complete tumor regression and 23 patients (40%) experienced tumor regression grade 3. Tumor down-staging occurred in 31 patients (54.4%) and down-staging of nodes occurred in 25 patients (43.9%). There was a significant difference in tumor stage between pre-treatment and post-surgery (P 〈 0.001). Patients with less advanced N stages had significantly better recurrence-free survival but similar metastasis-free survival and overall survival. Tumor regression grade was not associated with overall survival, recurrence-free survival or metastasis-free survival. The most common adverse events were pulmonary infection (n = 6, 10.5%) and intestinal obstruction (n = 6, 10.5%). Conclusions An additional cycle of chemotherapy given after chemoradiotherapy and before surgery provided good efficacy and had a satisfactory safety profile in patients with locally advanced rectal cancer.

    Abstract: In order to fabricate the abrasive wheels with good grain self-sharpening capacity, two types of multicrystalline CBN grains, that is, polycrystalline CBN (PCBN) and binderless CBN (BCBN), were brazed using Cu-Sn-Ti alloy, respectively. Comparative investigation on the brazing interface, compressive strength, and wear properties of the different grains was carried out. Results obtained show that the PCBN grains have more intricate reaction, more complicated resultants, and thicker reaction layer than the BCBN counterparts under the identical brazing conditions. Though the average compressive strength of the PCBN grains is similar to that of BCBN ones, stronger self-sharpening action by virtue of the microfracture behavior takes place with BCBN grains during grinding. As a consequence, compared to the brazed PCBN wheels and the conventional monocrystalline CBN (MCBN) ones, longer service life is obtained for the brazed BCBN wheels.

    Abstract: Autism is a severe neurodevelopmental disorder with a strong genetic basis.The methyl-CpG binding protein 2 gene (MECP2) is a dosage-sensitive gene in brain development and has been implicated as a candidate gene for autism. Duplication of the MECP2 gene has been reported in a few boys with autistic features. To further investigate the association of MECP2 duplication with autism, the authors performed real-time quantitative polymerase chain reaction (PCR) to detect copy number variations of the MECP2 gene in 82 autistic boys. No copy number variation was found in these patients, indicating that duplication of the MECP2 gene is not frequent in autistic patients. The authors consider that duplication of the MECP2 gene has no major effect on the susceptibility to autism. Replication of studies in a large-sized sample and a well-characterized subgroup of autism are warranted to further identify the association of MECP2 gene duplication with autism.

    Abstract: Conventional instrumentation has difficulty to measure displacements inside laboratory-scale physical models. In this paper, an innovative fiber Bragg grating (FBG) sensing bar has been developed to perform high-accuracy and real-time monitoring of internal displacements. According to Euler-Bernoulli beam theory, the strain distribution measured by the quasi-distributed FBG sensors adhered on the bar surface can be used to compute the displacement profile along the sensing bar. The effectiveness of the FBG sensing bar has been verified by calibration tests. In the physical model of the Wudu gravity dam, two FBG sensing bars developed by the authors were installed, together with linear variable displacement transformers (LVDTs) and strain rosettes. The FBG sensing bars successfully captured the variation of internal displacement profiles in the model dam. The monitoring results during the overloading test were presented, based on which the deformation mechanism of the dam-foundation system is explained in details. The displacements measured by the FBG sensing bars are validated by their good agreements with those from LVDTs.

    Abstract: Umbilical cord mesenchymal stem cells (UC-MSCs) have several advantages for clinical therapy: the material is easily obtainable, the donation procedure is painless and there is low risk of viral contamination. UC-MSCs play important roles in tissue regeneration, tissue damage repair, autoimmune disease and graft-versus-host disease. In this study, we investigated the normal mRNA expression profile of UC-MSCs, and analyzed the candidate proteins responsible for the signaling pathway that may affect the differentiation characteristics of UC-MSCs. UC-MSCs were isolated by mincing UC samples into fragments and placing them in growth medium in a six-well plate. The immunophenotype characteristics and multilineage differentiation potential of the UC-MSCs were measured by flow cytometry and immunohistochemical assays. In addition, the pathway-focused gene expression profile of UC-MSCs was compared with those of normal or tumorous cells by realtime quantitative polymerase chain reaction. We successfully isolated and cultured UC-MSCs and analyzed the appropriate surface markers and their capacity for osteogenic, adipogenic and neural differentiation. In total, 168 genes focusing on signal pathways were examined. We found that the expression levels of some genes were much higher or lower than those of control cells, either normal or tumorous. UC-MSCs exhibit a unique mRNA expression profile of pathway-focused genes, especially some stemness genes, which warrants further investigation.

    Abstract: A small amount of literature has been produced on the study of panic spread in a large-scale emergency evacuation, especially that which involves rescue guidance. In a large-scale evacuation action, there are complicated interactions between people and the disaster environment, and it is very difficult to present such interactions in quantitative functions or specific values. In this paper, a qualitatively simulated approach to model and study the panic spread is proposed. First, the internal structure of the evacuation system is described and various internal and external phenomena related to the change of evacuees’ behaviors in the evacuation process are qualitatively interpreted. Based on the qualitative knowledge, a qualitative simulation model of a large-scale evacuation system is established. The calculation results of inverse group matrix verify the rationality and stability of our model. According to the implementation of a series of scenarios with different input, some uncertainty factors that can affect the panic spread in the evacuation process are analyzed, in which the spread of disaster, the rescue guidance, and the normal emotional evacuees are mainly considered. This model reproduces a well-known phenomenon in crowd evacuation, namely “fast is slow”, and confirms that the severity of disaster exponentially positively correlates with the panic spread, and the effectiveness of rescue guidance is influenced by the leading emotion in the crowds as a whole.
