Bennett, Claire Perilleux, Tom Beeckman, and Xavier Draye Lateral root (LR) emergence represents a highly coordinated process in which the plant hormone auxin plays a central role. Reactive oxygen species (ROS) have been proposed to function as important signals during auxin-regulated LR formation; however, their mode of action is poorly understood. Here, we report that Arabidopsis roots exposed to ROS show increased LR numbers due to the activation of LR pre-branch sites and LR primordia (LRP). Strikingly, ROS treatment can also restore LR formation in pCASP1:shy2-2 and aux1 lax3 mutant lines in which auxin-mediated cell wall accommodation and remodeling in cells overlying the sites of LR formation is disrupted. Specifically, ROS are deposited in the apoplast of these cells during LR emergence, following a spatiotemporal pattern that overlaps the combined expression domains of extracellular ROS donors of the RESPIRATORY BURST OXIDASE HOMOLOGS (RBOH). We also show that disrupting (or enhancing) expression of RBOH in LRP and/or overlying root tissues decelerates (or accelerates) the development and emergence of LRs. We conclude that RBOH-mediated ROS production facilitates LR outgrowth by promoting cell wall remodeling of overlying parental tissues.

    Description: This article uses possible-world semantics to model the changes that may occur in an agent's knowledge as she loses information. This builds on previous work in which the agent may forget the truth-value of an atomic proposition, to a more general case where she may forget the truth-value of a propositional formula. The generalization poses some challenges, since in order to forget whether a complex proposition is the case, the agent must also lose information about the propositional atoms that appear in it, and there is no unambiguous way to go about this. We resolve this situation by considering expressions of the form [] , which quantify over all possible (but ‘minimal’) ways of forgetting whether . Propositional atoms are modified non-deterministically, although uniformly, in all possible worlds. We then represent this within action model logic in order to give a sound and complete axiomatization for a logic with knowledge and forgetting. Finally, some variants are discussed, such as when an agent forgets (rather than forgets whether ) and when the modification of atomic facts is done non-uniformly throughout the model.

Fernandez, Fabio Piano, Kristin C. Gunsalus, and Valerie Reinke The complex cellular events that occur in response to fertilization are essential for mediating the oocyte-to-embryo transition. Here, we describe a comprehensive small-molecule screen focused on identifying compounds that affect early embryonic events in Caenorhabditis elegans . We identify a single novel compound that disrupts early embryogenesis with remarkable stage and species specificity. The compound, named C22, primarily impairs eggshell integrity, leading to osmotic sensitivity and embryonic lethality. The C22-induced phenotype is dependent upon the upregulation of the LET-607/CREBH transcription factor and its candidate target genes, which primarily encode factors involved in diverse aspects of protein trafficking. Together, our data suggest that in the presence of C22, one or more key components of the eggshell are inappropriately processed, leading to permeable, inviable embryos. The remarkable specificity and reversibility of this compound will facilitate further investigation into the role and regulation of protein trafficking in the early embryo, as well as serve as a tool for manipulating the life cycle for other studies such as those involving aging.

    Description: In this work, we shall study structural abduction and how ways of searching for solutions to the corresponding abductive problems could be modeled. Specifically, we shall define modal semantic tableaux for normal modal systems and study its applications to structural abduction. This method even makes structural abduction clearer and, as it shall be seen, when a radical change of logic is epistemologically required, the corresponding tableau will have pertinent information to suggest it.

    Description: We present a framework for understanding abduction within modal logic and Kripke semantics; worlds of a Kripke frame will represent possible theories, and a change in theory will be understood as a passage from one world to an adjacent possible world. Further, these steps may agree with the accessibility relation or may ‘backtrack’, accordingly as new information refutes or reinforces our present theory. Our formalism can be used to model not only abduction, but also to talk about the inner structure of theories as well as relations between them, allowing us to interpret many ideas from philosophy of science within the well-understood framework of modal logic.
