    Abstract: Metastatic breast cancer dominates the female cancer-related mortality. Tumour-associated molecules represents a crucial for early disease detection and identification of novel therapeutic targets. Nanomaterial technologies provide promising novel approaches to disease diagnostics and therapeutics. In the present study we extend the investigations of antitumoral properties of Carbon Dots prepared from N -hydroxyphthalimide (CD-NHF) precursor. We evaluate the effect of CD-NHF on tumour cell migration and invasion in vitro and their impact on tumour progression using an in vivo model. Furthermore, we investigate the molecular mechanisms involved in CD-NHF antitumour effects. In vivo mammary tumours were induced in Balb/c female mice by injecting 4T1 cells into the mammary fat pad. Conditional treatment with CD-NHF significantly impair both migration and invasion of metastatic breast cancer cells. The presence of CD-NHF within the 3D cell cultures strongly inhibited the malignant phenotype of MDA-MB-231, 4T1 and MCF-7 cells in 3D culture, resulting in culture colonies lacking invasive projections and reduction of mammospheres formation. Importantly, breast tumour growth and metastasis dissemination was significantly reduced upon CD-NHF treatments in a syngeneic mouse model and is associated with down-regulation of Ki67 and HSP90 expression. CD-NHF nanostructures provide exciting perspective for improving treatment outcome in breast cancer.

    Abstract: In the past decade, the probiotic market has grown rapidly, both for foods and supplements intended to enhance wellness in healthy individuals. Different lactic acid bacteria (LAB), especially Lactobacillus spp., of different origins have already been used to develop commercial probiotic products. Nowadays, LAB new alternative sources, such as non-dairy fermented food products, are being exploited. One such source is Kombucha, a fermented low-alcohol beverage made of tea leaves. In this regard, we tested seven Pediococcus spp. strains isolated from a local industrial Kombucha for their biotechnological potential. Two, out of the seven isolates, identified as Pediococcus pentosaceus (L3) and Pediococcus acidiliactici (L5), were selected as successful candidates for the food industry, due to their probiotic and technological properties. In regard to their resistance in the gastro-intestinal tract, both selected strains were tolerant to a pH of 3.5, presence of 0.3% pepsin, and 0.5% bile salt concentration. On the antagonistic side, the fresh suspension of selected isolates had high inhibitory activity against pathogenic bacteria, such as Salmonella enterica Typhimurium, Listeria monocytogenes, Listeria ivanovii, Bacillus cereus, Proteus hauseri, and methicillin resistant Staphylococcus aureus. In addition, moderate to high inhibitory activity was noticed against foodborne molds (e.g., Penicillium expansum and Penicillium digitatum). These safety issues were supported by their negative hemolytic activity and good antioxidant potential (56–58%). Selected isolates were sensitive to ampicillin, penicillin, erythromycin, and lincomycin, while a broad range of other antibiotics were not effective inhibitors. On the technological side, both strains tolerated 5% NaCl and, during the freeze-drying process, had a good survival rate (86–92%). The selected Pediococcus strains have proven properties to be used for further development of functional products.

    Abstract: A new technique for the assay of carbohydrates is described in which separation and quantification of neutral saccharides, aminosaccharides, glycuronic acids, and disaccharides may be accomplished in less than 50 min of total run time. This method involves optimized anion-exchange liquid chromatography coupled with integrated pulse amperometric detection. Complex carbohydrates from various sources, including dietary supplements, were hydrolyzed in a dilute solution of trifluoroacetic acid, freeze-dried, and reconstituted in water containing 2-deoxygalactose as the internal standard. The solution was filtered and separated on CarboPac PA20 column. The eluted saccharides were detected by oxidation on a gold electrode with quadruple-pulsed integrated amperometry. The calibration plots for the saccharides were linear with an average correlation coefficient of 0.999. Method precisionc regarding peak retention time and resolution used in the peak identifications was verified. With this method, previously difficult-to-separate saccharides, such as galactosamine, glucosamine, and N-acetylglucosamine, were successfully resolved from the neutral saccharides rhamnose, arabinose, and galactose. Mannose was also resolved from xylose, and de-acetylation of aminosaccharides prior to separation was not necessary. This technique provides an accurate and efficient means to assay carbohydrates in dietary supplements, which new federal regulations will soon mandate.

    Abstract: Aloe Vera inner leaf gel has been used as a medicinal remedy for many years. Yet some aloe products do not demonstrate beneficial effects, indicating that poor quality products are reaching the market. Therefore, an efficient and accurate method is needed to evaluate the quality of aloe products. This paper describes a quick, quantitative colorimetric assay that has been developed for the determination of glucomannan in aloe gel and products. With this method, interference by non-aloe polysaccharides or other extraneous components was absent or negligible. Data indicate that the glucomannan can be determined at parts per million (mg/L) in aqueous solutions with an accuracy of 100 ± 5% at a 10 mg/L concentration. The correlation coefficient is 0.999, and linearity is from 0.9 to 72.7 mg/L in the test solution. The method is inexpensive, simple, sensitive, and reproducible. This method was applied to determine the polysaccharide content of commercial aloe products. Both qualitative and quantitative information can be obtained in about 5 min.

    Abstract: Rosa canina L. fruit have been used internally as tea for treatment of viral infections and disorders of the kidneys and urinary tract. Also, it was reported that the R. canina fruit, with its high ascorbic acid, phenolics and flavonoids contents, have antioxidant, antimutagenic and anticarcinogenic effects. Antioxidants neutralize free radicals, which cause oxidative damage to lipids, proteins, and nucleic acids and thus protect the organism. Research studies conducted on this topic have shown that natural antioxidants are involved in protection against many diseases: cancer, cardiovascular diseases and osteoporosis. Different processing and storage methods of the fruit could affect the bioactive nutrients while preserving the antioxidant capacity of them is very important. The aim of this study was to evaluate the effect of storage in a dry or frozen state on the antioxidants content of R. canina fruit. The evaluation involved determination of carotenes, ascorbic acid and total polyphenols using spectrophotometrically methods. Analysis performed on the R. canina fruit showed significant changes in the antioxidants content in fresh, dried and frozen fruit during storage.

    Abstract: Since ancient times, wild plants have widely been traditionally consumed by different communities but today are gaining relevance due to their healthy properties. Vegetables, including wild edible species, constitute an important source of active natural products: micronutrients, especially vitamins and minerals and phytochemical compounds with antioxidant properties important in the prevention of various pathologies including degenerative, cardiovascular and neurological diseases. Some species of wild and cultivated edible plants were comparatively evaluated considering the content in bioactive compounds and the antioxidant capacity. Biochemical analysis of the fresh leaves indicated similar or even higher values of nutritive compounds (sugars, protids) and antioxidants (polyphenols, carotenoids, flavones, chlorophylls, ascorbic acid) in the species from spontaneous flora as dandelion (Taraxacum officinale), lesser celandine (Ficaria verna), wild garlic (Allium ursinum) than in the green lettuce and garden rocket commonly consumed around the world. Therefore, these wild plants could be recommended for consumers not only as new ingredients to improve their diet diversity but also for providing potential health benefits.
