    Abstract: Icaritin (ICT) is the main component in the traditional Chinese herb Epimedium , and it has been shown to have anti-Alzheimer’s disease (AD) effects, but its neuroprotective effects and the pharmacological mechanisms are unclear. In the present study, senescence-accelerated mouse prone 8 (SAMP8) mice were randomly divided into a model group and an ICT-treated group. Learning and memory abilities were detected by the Morris water maze assay, and the expression of amyloid beta protein (A β ) and β -site APP cleavage enzyme 1 (BACE1) was determined by Western blotting and polymerase chain reaction (PCR). Histological changes in CA1 and CA3 were detected by hematoxylin-eosin staining (H & E staining), and the immunohistochemical analysis was used to detect the expression and localization of Bax and Bcl-2. The results showed that compared with the SAMP8 mice, the ICT-treated SAMP8 mice showed improvements in spatial learning and memory retention. In addition, the number of necrotic cells and the morphological changes in CA1 and CA3 areas were significantly alleviated in the group of ICT-treated SAMP8 mice, and the expression of BACE1, A β 1-42 levels, and the Bax/Bcl-2 ratio in the hippocampus was obviously decreased in the ICT-treated group compared with the control group. The results demonstrated that ICT reduced BACE-1 levels, the contents of A β 1-42, and the Bax/Bcl-2 ratio, suggesting that ICT might have potential therapeutic benefits by delaying or modifying the progression of AD.

    Abstract: Detection of substation equipment can promptly and effectively discover equipment overheating defects and prevent equipment failures. Traditional manual diagnosis methods are difficult to deal with the massive infrared images generated by the autonomous inspection of substation robots and drones. At present, most of the infrared image defect recognition is based on traditional machine learning algorithms, with low recognition accuracy and poor generalization capability. Therefore, this paper develops a method for identifying infrared defects of substation equipment based on the improvement of traditional ones. First, based on the Faster RCNN, target detection is performed on 6 types of substation equipment including bushings, insulators, wires, voltage transformers, lightning rods, and circuit breakers to achieve precise positioning of the equipment. Afterwards, different classes are identified based on the sparse representation-based classification (SRC), so the actual label of the input sample can be obtained. Finally, based on the temperature threshold discriminant algorithm, defects are identified in the equipment area. The measured infrared images are used for experiments. The average detection accuracy achieved by the proposed method for the 6 types of equipment reaches 92.34%. The recognition rate of different types of equipment is 98.57%, and the defect recognition accuracy reaches 88.75%. The experimental results show the effectiveness and accuracy of the proposed method.

    Abstract: Background. To observe different roles of direct bilirubin (Dbil) on portopulmonary hypertension (POPH) and idiopathic pulmonary arterial hypertension (IPAH). Methods. Thirty incident patients with POPH and 180 with IPAH (matched by the WHO functional classification in a 1 : 6 ratio) between March 2010 and December 2020 were included. The receiver operating curve and Kaplan–Meier method were applied to estimate the ability to distinguish between the two and survival, respectively. Univariate and forward multiple stepwise regression analyses were performed to access the relationship between pulmonary vascular resistance (PVR) and clinical indices. Results. Compared to IPAH, the POPH group had better hemodynamics including PVR (7.08 ± 3.95 vs. 14.89 ± 7.11, P 〈 0.001 ) and higher total bilirubin (Tbil) and Dbil. Tbil and Dbil had a negative correlation with PVR in the POPH group (r = −0.394, P = 0.031 ; r = −0.364, P = 0.048 , respectively) but positive correlation in the IPAH group (r = 0.218, P = 0.003 ; r = 0.178, P = 0.018 , respectively). Increased neutrophil counts (r = 0.394, P = 0.031 ) and elevated NT-proBNP (r = 0.433, P 〈 0.001 ) would help predict the elevation of PVR in POPH and IPAH groups independent of Dbil, respectively. Dbil could distinguish POPH from IPAH (AUC = 0.799, P = 0.009 ), and the ability was elevated when taking aspartate aminotransferase together (AUC = 0.835, P 〈 0.001 ). The overall survival was better in POPH than in IPAH (7 dead cases of POPH and 96 of IPAH, P = 0.002 ). Survival was better in POPH than in IPAH in the group of Dbil ≥7 μmol/L ( P = 0.001 ) but showed no significant difference between POPH and IPAH in the group of Dbil 〈 7 μmol/L ( P = 0.192 ). Conclusions. The POPH group had a better hemodynamic profile than IPAH. Dbil was associated oppositely with the elevation of PVR in POPH and IPAH. Patients with POPH had better survival than those with IPAH in the total cohort and in the group of Dbil ≥7 μmol/L, but limited dead cases of POPH should be noted.

    Abstract: Drought significantly threatens crop productivity and food security worldwide. However, the severity of drought is predicted to increasingly intensify in the future. To provide an antidrought strategy for farmers and breeders, the response of stomatal behavior of crops to water stress should be well studied. In this study, a lysimeter experiment was conducted to study the relationship between gas exchange parameters and grain yields of winter wheat. Light, moderate, and severe drought levels were imposed at seedling, jointing, heading, and filling stages. The results showed that crop evapotranspiration (ETc, mm) of winter wheat during the entire growing season was limited by drought imposed at any growth stage, and ETc under severe drought treatment was always the lowest. The stomatal limitation value had a significant linear correlation with the stomatal conductance (Gs, μmol mol H2O m–2 s–1) and transpiration rate (Tr, mmol H2O m–2 s–1). Light and moderate drought levels at the seedling stage did not generate irreversible physiological stress on wheat plants, while severe drought at any growth stage caused significant reduction in gas exchange parameters and grain yields. Theoretical threshold values of leaf water use efficiency (WUEl) for light, moderate, and severe drought levels were 2.62, 3.36, and 4.11 μmol mmol–1, respectively. The threshold values are useful to provide theoretical reference for achieving smart irrigation in the North China Plain.

    Abstract: Enhancer RNAs (eRNAs) have emerged as key players in the pathology of several tumors, including uveal melanoma. Here, we aimed to explore the prognostic values of eRNAs in uveal melanoma (UVM) patients. The expressing data and survival data of UVM patients were downloaded from TCGA and GSE22138 datasets. The Kaplan-Meier methods with the log-rank test were applied to screen survival-related eRNAs in UVM. GEPIA was applied to analyze the associations between expressions of eRNA and disease-free survival. KEGG assays were applied to explore the potential signaling pathways of the key eRNA. The prognostic values of eRNAs were further explored by multivariate assays by the R package survival. The eRNAs were validated in pan-cancer. In this study, we identified 89 survival-related eRNAs in UVM based on TCGA datasets. Based on GSE22138 datasets, we found 27 survival-related eRNAs in UVM. Only two eRNAs (LINC00689 and ELFN1-AS1) were overlapped in both two datasets. The results of multivariate analysis revealed that both LINC00689 and ELFN1-AS1 were independent prognostic factors in UVM patients. The pan-cancer validation results further confirmed the prognostic values of LINC00689 and ELFN1-AS1 in eight tumors. Overall, we identified two novel UVM-related eRNAs, LINC00689 and ELFN1-AS1 which may serve as prognostic and diagnostic biomarkers of UVM patients for clinical decision-making.

    Abstract: Recent studies suggest that the epithelial sodium channel (ENaC) is expressed in the endothelial cells. To test whether high salt affects the NO production via regulation of endothelial ENaC, human umbilical vein endothelial cells (HUVECs) were incubated in solutions containing either normal or high sodium (additional 20 mM NaCl). Our data showed that high sodium treatment significantly increased α -, β -, and γ -ENaC expression levels in HUVECs. Using the cell-attached patch-clamp technique, we demonstrated that high sodium treatment significantly increased ENaC open probability ( P O ). Moreover, nitric oxide synthase (eNOS) phosphorylation (Ser 1177) levels and NO production were significantly decreased by high sodium in HUVECs; the effects of high sodium on eNOS phosphorylation and NO production were inhibited by a specific ENaC blocker, amiloride. Our results showed that high sodium decreased AMP-activated kinase (AMPK) phosphorylation in endothelial cells. On the other hand, metformin, an AMPK activator, prevented high sodium-induced upregulation of ENaC expression and P O . Moreover, metformin prevented high salt-induced decrease in NO production and eNOS phosphorylation. These results suggest that high sodium stimulates ENaC activation by negatively modulating AMPK activity, thereby leading to reduction in eNOS activity and NO production in endothelial cells.

    Abstract: Background . Acute exacerbation of IPF (AE-IPF) is associated with high mortality. We studied changes in pathogen involvement during AE-IPF and explored a possible role of infection in AE-IPF. Objectives . Our purpose is to investigate the role of infection in AE-IPF. Methods . Overall, we recruited 170 IPF patients (48 AE-IPF, 122 stable) and 70 controls at Shanghai Pulmonary Hospital. Specific IgM against microbial pathogens and pathogens in sputum were assessed. RNA sequences of pathogens in nasopharyngeal swab of IPF patients were detected by PathChip. A panel of serum parameters reflecting immune function were assessed. Results . Antiviral/bacterial IgM was higher in IPF vs. controls and in AE-IPF vs. stable IPF. Thirty-eight different bacterial strains were detected in IPF patient sputum. Bacteria-positive results were found in 9/48 (18.8%) of AE-IPF and in 26/122 (21.3%) stable IPF. Fifty-seven different viruses were detected in nasopharyngeal swabs of IPF patients. Virus-positive nasopharyngeal swabs were found in 18/30 (60%) of tested AE-IPF and in 13/30 (43.3%) of stable IPF. AE-IPF showed increased inflammatory cytokines (IL-6, IFN- γ , MIG, IL-17, and IL-9) vs. stable IPF and controls. Mortality of AE-IPF in one year (39.5%) was higher compared to stable IPF (28.7%) .Conclusions . IPF patients had different colonization with pathogens in sputum and nasopharyngeal swabs; they also displayed abnormally activated immune response, which was exacerbated during AE-IPF.

    Abstract: Background . To date, studies have shown that polymorphisms in an autophagy-related gene, IRGM , are linked with different diseases, especially autoimmune diseases. The present study aimed to examine the roles of IRGM polymorphisms in autoimmune thyroid diseases (AITD). Methods . Three polymorphisms in IRGM gene (rs10065172, rs4958847, and rs13361189) were genotyped in 1569 participants (488 with Graves’ disease, 292 with Hashimoto’s thyroiditis, and 789 healthy controls) using PCR-based ligase detection reaction method. Gene-disease associations were evaluated for the three SNPs. Results . T allele of rs10065172, A allele of rs4958847, and C allele of rs13361189 were all higher in Graves’ disease patients than controls, and the ORs were OR = 1.207 ( P = 0.022 ), OR = 1.207 ( P = 0.027 ), and OR = 1.200 ( P = 0.027 ), respectively. After adjusting for sex and age, rs10065172 and rs13361189 were still associated with GD under both the allele model and dominant model, and the adjusted ORs for rs10065172 were 1.20 ( P = 0.0 33 ) and 1.33 ( P = 0.02 4 ), while the adjusted ORs for rs13361189 were 1.19 ( P = 0.0 42 ) and 1.33 ( P = 0.02 6 ), respectively. No significant difference was found between Hashimoto’s thyroiditis patients and controls. Haplotype analysis found that CTA frequency was distinguishingly higher in Graves’ disease patients (OR = 1.195, P = 0.030 ). The frequency of TCG haplotype was distinguishingly lower in AITD and Graves’ disease patients (OR = 0.861, P = 0.044 ; OR = 0.816, P = 0.017 ). Conclusions . Our study reveals IRGM as a susceptibility gene of AITD and Graves’ disease for the first time.

    Abstract: Background. CircFBXW7 has been determined to be involved in various cancers; however, its role in nonsmall cell lung cancer (NSCLC) remains unclear. This study examined the function and potential mechanism of circFBXW7 in NSCLC. Methods. The structure of circFBXW7 was verified via RT-PCR and Sanger sequencing. The expression of circFBXW7 in NSCLC was determined by qRT-PCR. The effect of circFBXW7 overexpression on the proliferation, migration, and invasion of NSCLC cells was examined by CCK-8 and Transwell assays. Furthermore, a circFBXW7-miRNA network was established to explore their interaction. Predicted miRNA was determined by qRT-PCR. Moreover, the miRNA mimics were synthesized, wherein its effect on proliferation, migration, and invasion of NSCLC cells overexpressed circFBXW7 was assessed. Results. The circularity of circFBXW7 was verified. The expression of circFBXW7 was found to be downregulated in NSCLC cells compared with that in normal human lung epithelial BEAS-2B cells. Overexpression of circFBXW7 reduced cell proliferation, migration, and invasion. Furthermore, according to the circFBXW7-miRNA network prediction and qRT-PCR validation, miR-492 was identified to be the target of circFBXW7. The inhibitory effect of circFBXW7 overexpression on cell proliferation, migration, and invasion was reversed by miR-492 mimics. Conclusion. CircFBXW7 is downregulated in NSCLC. CircFBXW7 inhibits NSCLC cells proliferation, migration, and invasion by regulating miR-492.
