    Abstract: Graves’ ophthalmopathy (GO), which is characterized by orbital tissue inflammation, expansion, and fibrosis, is the ocular manifestation in 25% to 50% of patients with Graves’ disease. As the pathology of GO is driven by autoimmune inflammation, many proinflammatory cytokines/chemokines, including TNF-α, IL-1β, IL-6, and CCL20, are crucial in the pathogenesis of GO to activate the orbital fibroblasts. Cysteine-rich protein 61 (CYR61), which is known to regulate cell proliferation, adhesion, and migration, plays a proinflammatory role in the pathogenesis of many inflammatory diseases, such as rheumatoid arthritis. CYR61 was considered a potential biomarker of GO in recent studies. Statins, which are cholesterol-lowering drugs, were found to reduce the risk of GO, probably through their anti-inflammatory and immunomodulatory effects. In this study, we established a link between CYR61 and statins in the pathogenesis and potential treatment for GO. Firstly, our data showed the overexpression of CYR61 in the orbital tissue ( n = 4 ) and serum specimens ( n = 6 ) obtained from the patients with inactive GO. CYR61 could induce the production of IL-6 and CCL20 in cultured GO orbital fibroblasts. The expression of CYR61 in cultured GO orbital fibroblasts was upregulated via TNF-α stimulation. Secondly, we pretreated cultured GO orbital fibroblasts using simvastatin, a statin, followed by TNF-α stimulation. The data revealed that simvastatin could inhibit TNF-α-induced CYR61 expression by modulating the activity of transcription factor FoxO3a. Our results provided insights into some cellular mechanisms that may explain the possible protective effects of simvastatin against the development of GO.

    Abstract: Hydrogen sulfide (H 2 S) is an important mediator participating in both physiological and pathological systems and related to the inflammatory process. Acupuncture has a therapeutic effect on inflammatory pain. However, whether H 2 S generated in the central nervous system (CNS) is a mediator of electroacupuncture (EA) treatment for inflammatory pain is unknown. We injected complete Freund’s adjuvant (CFA) to induce inflammatory pain and applied EA treatment as an interventional strategy for pain relief. The results presented here show that S-adenosyl-l-methionine (SAM), an allosteric activator of cystathionine- β -synthetase (CBS), may reverse the therapeutic effect of EA. CBS-induced H 2 S generation might get involved in the mechanism of EA-induced analgesia in the hippocampus on chronic inflammatory pain.

    Abstract: Background. CircFBXW7 has been determined to be involved in various cancers; however, its role in nonsmall cell lung cancer (NSCLC) remains unclear. This study examined the function and potential mechanism of circFBXW7 in NSCLC. Methods. The structure of circFBXW7 was verified via RT-PCR and Sanger sequencing. The expression of circFBXW7 in NSCLC was determined by qRT-PCR. The effect of circFBXW7 overexpression on the proliferation, migration, and invasion of NSCLC cells was examined by CCK-8 and Transwell assays. Furthermore, a circFBXW7-miRNA network was established to explore their interaction. Predicted miRNA was determined by qRT-PCR. Moreover, the miRNA mimics were synthesized, wherein its effect on proliferation, migration, and invasion of NSCLC cells overexpressed circFBXW7 was assessed. Results. The circularity of circFBXW7 was verified. The expression of circFBXW7 was found to be downregulated in NSCLC cells compared with that in normal human lung epithelial BEAS-2B cells. Overexpression of circFBXW7 reduced cell proliferation, migration, and invasion. Furthermore, according to the circFBXW7-miRNA network prediction and qRT-PCR validation, miR-492 was identified to be the target of circFBXW7. The inhibitory effect of circFBXW7 overexpression on cell proliferation, migration, and invasion was reversed by miR-492 mimics. Conclusion. CircFBXW7 is downregulated in NSCLC. CircFBXW7 inhibits NSCLC cells proliferation, migration, and invasion by regulating miR-492.

    Abstract: Objective. Neonatal hyperbilirubinemia is caused by the excessive production of bilirubin and decreased excretion ability in the neonatal period. It leads to a concentration of blood bilirubin that exceeds a certain threshold. Yinzhihuang oral liquid (YZH) is a traditional Chinese medicine mixture used in the treatment of neonatal hyperbilirubinemia in China. This article systematically explores the pharmacological mechanisms by which YZH acts in the treatment of neonatal hyperbilirubinemia through network pharmacology at the molecular level. Methods. We adopted the method of network pharmacology, which includes active component prescreening, target gene prediction, gene enrichment analysis, and network analysis. Results. According to the network pharmacological analysis, 8 genes (STAT3, AKT1, MAPK14, JUN, TP53, MAPK3, ESR1, and RELA) may be targets of YZH in the treatment of neonatal hyperbilirubinemia. In addition, Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses showed that YZH may regulate antioxidation, modulate lipid metabolism, and have anti-infective properties. Conclusion. In this study, the pharmacological action and molecular mechanisms of YZH were predicted as a whole. It was found that YZH is a promising drug for treating oxidative stress due to bilirubin, as it reduces immunosuppression and helps to eliminate virus infection.

    Abstract: This paper comparatively studies the electromagnetic force and mechanical response of the end winding before and after 3 kinds of typical electromechanical faults in turbo-generator. The analytical expression of electromagnetic force of end winding is derived under the composite fault of static eccentricity and rotor interturn short circuit. Meanwhile, the three-dimensional transient finite element simulation is carried on, and the frequency composition and amplitude variation characteristics of the radial, axial, and tangential electromagnetic force are analyzed for the end windings under static eccentricity, rotor interturn short circuit, and composite fault. Therefore, it provides a reference for the vibration wear detection and electromagnetic force control of the end winding. Moreover, the maximum stress and deformation of different positions on the end involute are obtained. And the three-directional vibration acceleration characteristics of the end winding are further analyzed. Finally, the distribution law of winding fatigue failure and vibration wear is acquired, which lays a foundation for the reverse suppression of end winding fatigue failure and insulation wear.

    Abstract: In order to study the vibration wearing regularity and the strength failure point of stator end windings before and after static rotor eccentricity, the three-dimensional electromagnetic forces and the subsequent mechanical responses are studied in this paper. The electromagnetic force, stress, and deformation on the end winding of the QFSN-600-2YHG turbo-generator are calculated by the finite element method (FEM) through an electromagnetic-structure coupling. The radial vibration characteristics of the winding are verified by experiments. It shows that the vibration wearing in the same layer is more serious than that between two neighboring layers. For different layers, the interphase coils endure a larger wearing risk than the innerphase coils. Inside the same phase, the last coil along the rotating direction has the highest risk of insulation wearing. The occurrence of static rotor eccentricity will significantly increase the electromagnetic forces and the vibration amplitudes on some coils. The end-phase coil which is close to the minimum air-gap point is the most dangerous one due to the lasting overstresses and the intensified deformations.

    Abstract: This paper comparatively studies on the end winding electromagnetic force and mechanical response for generators with different numbers of poles. The analytical expression of the end winding electromagnetic force is derived under the rotor winding interturn short circuit (RISC) considering the pole number. Meanwhile, the three-dimensional transient finite element simulation is carried on two generators with one-pair poles and three-pair poles. Then, the frequency composition and amplitude variation characteristics of the radial, axial, and tangential electromagnetic forces are analyzed. Further, the maximum stress and deformation on the end winding are calculated and the similarity and difference of the coil failure law are obtained for two kinds of generators. It is found that RISC will bring odd harmonics to electromagnetic force for one-pair pole generators but it will bring odd and fraction harmonics for multipair pole generators. Moreover, the max mechanical response under RISC will decrease for one-pair pole generators but it will increase for multipair pole generators.

    Abstract: Background. An injurious fall is one of the main indicators of care quality in healthcare facilities. Despite several fall screen tools being widely used to evaluate a patient’s fall risk, they are frequently unable to reveal the severity level of patient falls. The purpose of this study is to build a practical system useful to predict the severity level of in-hospital falls. This practice is done in order to better allocate limited healthcare resources and to improve overall patient safety. Methods. Four hundred and forty-six patients who experienced fall events at a large Taiwanese hospital were referenced. Eight predictors were used to ascertain the severity of patient falls solely based on the above study population. Multinomial logistic regression, Naïve Bayes, random forest, support vector machine, eXtreme gradient boosting, deep learning, and ensemble learning were adopted to establish predictive models. Accuracy, F1 score, precision, and recall were utilized to assess the models’ performance. Results. Compared to other learners, random forest exhibited satisfying predictive performance in terms of all metrics (accuracy: 0.844, F1 score: 0.850, precision: 0.839, and recall: 0.875 for the test dataset), and it was adopted as the base learner for a severity-level predictive system which is web-based. Furthermore, age, ability of independent activity, patient sources, use of assistive devices, and fall history within the past 12 months were deemed the top five important risk factors for evaluating fall severity. Conclusions. The application of machine learning techniques for predicting the severity level of patient falls may result in some benefits to monitor fall severity and to better allocate limited healthcare resources.
