    Abstract: Blastocystis spp. is a ubiquitous protozoon in the intestinal tract of human and many animals. Microscopic examination is the main method of clinical diagnosis for Blastocystis spp. , which is prone to false negative. A simple and rapid diagnosis of Blastocystis spp. infection is an important step to prevent and control blastocystosis. Here, a recombinase polymerase amplification-lateral flow dipstick (RPA-LFD) assay was developed for rapid visual detection of Blastocystis spp. DNA amplification could be performed within 18 min at 37°C. The minimum DNA detection limit was 1 pg/μL, and there was no cross-reactivity with 12 other non-target pathogens, which was consistent with the sensitivity of conventional PCR (cPCR). Furthermore, 56 fecal samples from the Third Affiliated Hospital of Xinxiang Medical University were tested using RPA and cPCR methods respectively, and the results were completely consistent. The results show that RPA-LFD method has high accuracy and visual results, which provides a new choice for the differential diagnosis and rapid field detection of Blastocystis spp.

    Abstract: Global climate extremes are increasingly frequent and intense, especially in Africa, which is most vulnerable to climate change (de Sherbinin 2013  Clim. Change   123 23–37). However, the vulnerability of the landscapes composed of diverse ecosystems to climate extremes is far from being clearly understood. This study constructed a set of index systems based on the ‘exposure-sensitivity-adaptive capacity’ framework to assess landscape vulnerability driven by abnormal drought and precipitation in sub-Saharan Africa. In addition, correlation analysis was used to discover factors affecting landscape vulnerability. The results showed that a high level of landscape vulnerability was determined by high exposure and high sensitivity, as adaptive capacity exhibited little difference. The drought and wet events occurred in 80.9% and 51.3% of the climate change-dominated areas during 2001–2020, respectively. In areas where drought anomalies occur, about 8% of the landscapes, primarily formed by sparse vegetation and grasslands, were susceptible to drought. Moreover, in areas with abnormal precipitation, high vulnerability occurred only in about 0.6% of landscapes mostly covered by grasslands and shrubs. In addition, the intensity of landscape vulnerability driven by drought was higher than that driven by precipitation anomalies in the areas that experienced both dry and wet anomalies. Furthermore, the greater the deviation of landscape richness, diversity, and evenness from the normal climate state, the stronger the landscape vulnerability. The results add new evidence for landscape instabilities—an obvious contrast driven by drought and wetness—from the perspective of landscape vulnerability. The methodology of assessing landscape vulnerability established in this study can provide a new way to guide the regulation of landscape composition in response to frequent climate extremes on a macro level.

    Abstract: In order to reveal the evolution of sediment provenance and paleoenvironment of the central North Jiangsu Plain (NJP) since marine isotope stage 2 (MIS 2), we analyzed lithology, accelerator mass spectrometry (AMS) 14 C dating, macrofossils and foraminifera, grain size, and geochemical compositions of sediments from two sediment cores (Core M and Core Y) recovered in the northern side of the Yangtze River. Our results show that fluvial environments prevailed in the central NJP during MIS 2, followed by alternations of coastal-marsh environments and shallow-bay environments in north central NJP and coastal-marsh environments in south central NJP during MIS 1. Provenance analysis suggests a major change in sediment provenance from the Yangtze River to the Yellow River at early MIS 1 in north central NJP; these Yellow River sediments might be transported from previously deposited sediments by strong tidal currents and intense waves to this area or be a reflection of a southward shift of the Yellow River during this period. However, the Yangtze River-derived sediments dominated the south central NJP since MIS 2. The depositional succession and sediment supplies since MIS 2 were dominantly controlled by sea-level fluctuations, regional geomorphic patterns, shift of the rivers, etc.

    Abstract: Systemic lupus erythematosus (SLE) is a prototypic autoimmune disease mainly mediated by IgG autoantibody. While follicular helper T (Tfh) cells are crucial for supporting IgG autoantibody generation in human SLE, underlying mechanisms for Tfh cell mal-differentiation remain unclear. Methods In total, 129 SLE patients and 37 healthy donors were recruited for this study. Circulating leptin was determined by ELISA from patients with SLE and healthy individuals. CD4 T cells isolated from SLE patients and healthy donors were activated with anti-CD3/CD28 beads under cytokine-unbiased conditions in the presence or absence of recombinant leptin protein, followed by detection for Tfh cell differentiation by quantifying intracellular transcription factor Bcl-6 and cytokine IL-21. AMPK activation was assessed by analyzing phosphor-AMPK using phosflow cytometry and immunoblots. Leptin receptor expression was determined using flow cytometry and its overexpression was achieved by transfection with an expression vector. Humanized SLE chimeras were induced by injecting patients’ immune cells into immune-deficient NSG mice and used for translational studies. Results Circulating leptin was elevated in patients with SLE, inversely associated with disease activity. In healthy individuals, leptin efficiently inhibited Tfh cell differentiation through inducing AMPK activation. Meanwhile, leptin receptor deficiency was a feature of CD4 T cells in SLE patients, impairing the inhibitory effect of leptin on the differentiation of Tfh cells. As a result, we observed the coexistence of high circulating leptin and increased Tfh cell frequencies in SLE patients. Accordingly, overexpression of leptin receptor in SLE CD4 T cells abrogated Tfh cell mal-differentiation and IgG anti-dsDNA generation in humanized lupus chimeras. Conclusion Leptin receptor deficiency blocks the inhibitory effect of leptin on SLE Tfh cell differentiation, serving as a promising therapeutic target for lupus management.

    Abstract: When talking about Sino-African relationships recently, few concern about China- Africa relationships in the context of the changing world. This study generally reviewed the background of the changing world and the evolution of China-Africa relations. For a long time, the Africa has been regarded as the hopeless continent. In the new century, the African countries experienced a transition from hopeless continent to rising Africa with more potentials. The state and private Chinese enterprises went to African countries not only for the crude oil and gas, the mining, and the forestry business, but also as important partner in infrastructure construction, joint enterprises as well as international trade partners. Most western views of China in Africa contained with negative attitude in the globalization era. The globalization and changing world gave the opportunity for China to improve the relations with African countries, especially in the economic relations. From 2009 to present, China became the top international trade partner in the whole Africa. There are also arguments with China in Africa in the future. The world is a complex socio-ecological system and corresponding with rapid changes, we provide the perspectives on the China-Africa relations in the context of changing world.

    Abstract: As an important part of the terrestrial ecosystem, vegetation dynamics are subject to impacts from both climate change and human activities. Clarifying the driving mechanisms of vegetation variation is of great significance for regional ecological protection and achieving sustainable development goals. Here, net primary productivity (NPP) was used to investigate the spatiotemporal variability of vegetation dynamics from 2000 to 2020 in East Africa, and its correlations with climate factors. Furthermore, we utilized partial derivatives analysis and set up different scenarios to distinguish the relative contributions of climatic and human factors to NPP changes. The results revealed that NPP exhibited a significant increase with 4.16 g C/m 2 /a from 2000 to 2020 in East Africa, and an upward trend was detected across 71.06% of the study area. The average contributions of precipitation, temperature, and solar radiation to the NPP inter-annual variations in East Africa were 2.02, −1.09, and 0.31 gC⋅m –2 ⋅a –1 , respectively. Precipitation made the greatest positive contribution among all of the climatic factors, while temperature made strong negative contributions. The contributions of climate change and human activities to NPP changes were 1.24 and 2.34 gC⋅m –2 ⋅a –1 , respectively. Moreover, the contribution rate of human activities to NPP increase was larger than that of climate change, while the role of climate change in NPP decrease was larger than that of human activities. The findings of the study can provide new evidence for a deeper understanding of ecosystem stability and carbon cycling in East Africa, as well as a reference for decision-making and scientific support for ecological environmental protection.

    Abstract: As an apicomplexan pathogen, Toxoplasma gondii still remains a major threat to public health and requires special attention. In fact, positive attempts to identify more effective antigens to provide protection are important to control toxoplasmosis. Latest scientific advances in T. gondii study hint at the probability of the T. gondii bradyzoite-formation deficient 1 (TgBFD1) as an ideal vaccine candidate, since this molecule plays a critical role in regulating the chronic infection of T. gondii . Thus, BALB/c mouse models of acute and chronic T. gondii infections were used to evaluate the TgBFD1 protection efficacy in this study. Before conducting animal trials, antigen analysis of TgBFD1 was performed using DNAstar software and Western blots. The preliminary results suggested that TgBFD1 should be a potent immunogen. Then, this conclusion is confirmed by ELISA assays. After immunization with rTgBFD1, high levels of specific IgG, IgG1, IgG2a, and cytokines (Interferon γ and interleukin 10) were observed, indicating that TgBFD1 could induce strong protective antibody responses. While TgBFD1-specific IgG antibodies were measurable in vaccinated mice, no protection was observed in the acute T. gondii infection (RH strain) assay. However, a noticeable decrease in brain cysts counts of immunized mice compared with negative controls in the latent T. gondii infection (PRU strain) assay was observed. Taken together, these results indicated that rTgBFD1 had the remarkable ability to elicit both humoral and cellular immune responses and could provide partial protective immunity against chronic T. gondii infection.
