    Abstract: With the fast development of the cycle-pulse lasers, the control of the carrier-envelope phase offset (CEO) frequency has been an important research subject in such fields as femtosecond optical frequency metrology, strong field physics and high harmonic generation. In this paper, a different measurement method for CEO is demonstrated and subsequently a better beat frequency signal with the signal-to-noise ratio greater than 40 dB is obtained. Based on this optimized beat frequency signal, the CEO of the Ti:sapphire laser is stabilized to the standard signal of 10 MHz of the microwave reference source TV-Rb clock by using the electronic phase-locked loop techniques and the piezo-transducers which are mounted on the mirror inside the laser cavity to change the frequency and phase. The repetition rate of the Ti:sapphire laser is locked to the same TV-Rb clock simultaneously. The experimental results showed that the stability of the repetition rate reached the same level as the TV-Rb clock and that of the CEO frequency was increased by three orders.

    Abstract: Few studies have investigated the associations of childhood growth trajectories with the prenatal metabolic risks of mothers and their interaction with children’s genetic susceptibility. Objective To investigate the effects of gestational metabolic syndrome (GMS) risks and children’s polygenic risk scores (PRSs), and their interaction effect on the BMI trajectory and obesity risk of offspring from birth to 6 years of age. Methods A total of 2,603 mother-child pairs were recruited from the Ma’anshan birth cohort (Anhui Province of China) study. Data on maternal prepregnancy obesity, gestational weight gain (GWG), gestational diabetes mellitus (GDM), and hypertensive disorders of pregnancy (HDP) were used to evaluate maternal GMS risk. In addition, 1,482 cord blood samples were used to genotype 11 candidate single-nucleotide polymorphisms (SNPs) to calculate children’s PRSs. The latent class growth model using the longitudinal BMI-for-age z scores (BMIz) was applied to validly capture the BMIz growth trajectory. Results Maternal GMS status was associated with higher BMIz scores and with an increased risk of overweight/obesity. Positive relationships were revealed between PRS and the risk of overweight/obesity among girls. Additionally, maternal GMS significantly interacted with the child’s PRS on BMIz scores and the risk of overweight/obesity among girls. Hierarchical BMI trajectory graphs by different exposure groups showed consistent findings, and both boys’ and girls’ BMIz trajectories were divided into three groups. Among girls, the higher the GMS risk or PRS they had, the higher the probability of being in the high BMIz trajectory group. Conclusions Maternal GMS status increased BMIz scores and the risk of obesity in both boys and girls and elevated the child’s BMI trajectory from birth to 6 years of age among girls. PRSs were significantly associated with children’s BMI trajectory and the risk of obesity and modified the associations between maternal GMS status and obesity biomarkers only among girls. Thus, regarding childhood obesity, steps should be taken to decrease maternal metabolic risks before and during pregnancy, and sex discrepancies should be noted to identify high-risk populations after birth to hierarchically manage them.

    Abstract: While chimeric antigen receptor (CAR)-T cell therapy is becoming widely used in hematological malignancies with remarkable remission rate, their high recurrence remains an obstacle to overcome. The role of consolidative transplantation following CAR-T cell-mediated remission remains controversial. We conducted a retrospective study to explore whether bridging to unrelated cord blood transplantation (UCBT) could improve the prognosis of patients entering remission after CAR-T therapy with different characteristics through subgroup analyses. Methods We reviewed 53 patients with relapsed/refractory (R/R) B-cell acute lymphoblastic leukemia (B-ALL) successfully infused with CD19 CAR-T cells and achieved complete remission (CR). In this study, 25 patients received consolidative UCBT (UCBT group) and 28 patients did not accept any intervention until relapse (non-UCBT group). Subgroup analysis on prognosis was then performed according to gender, age, number of previous relapses, tumor burden, presence of poor prognostic markers, and structure of CAR. Results Compared with the non-UCBT group, patients who underwent consolidative UCBT had better median event-free survival (EFS; 12.3 months vs. 6.2 months; P = 0.035) and relapse-free survival (RFS; 22.3 months vs. 7.2 months; P = 0.046), while no significant difference was found in overall survival (OS; 30.8 months vs. 15.3 months; P = 0.118). Subsequent multivariate analysis revealed that bridging to UCBT was a protective factor for RFS (P = 0.048) but had no significant effect on EFS (P = 0.205) or OS (P = 0.541). In the subgroup analysis, UCBT has an added benefit in patients with specific characteristics. Patients who experienced ≥2 relapses or with sustained non-remission (NR) showed better RFS (P = 0.025) after UCBT. Better EFS was seen in patients with poor prognostic markers (P = 0.027). In the subgroup with pre-infusion minimal residual disease (MRD) ≥5% or with extramedullary disease (EMD), UCBT significantly prolonged EFS (P = 0.009), RFS (P = 0.017), and OS (P = 0.026). Patients with occurrence of acute graft-versus-host disease (aGVHD) appeared to have a longer duration of remission (P = 0.007). Conclusion Consolidative UCBT can, to some extent, improve clinical outcomes of patients with R/R B-ALL entering remission following CD19 CAR-T therapy, especially in patients with more recurrences before treatment, patients with poor prognostic markers, and patients with a higher tumor burden. The occurrence of aGVHD after UCBT was associated with better RFS.

    Abstract: We firstly investigate based on 2022 European Leukemia Net (ELN) risk stratification, whether standard-dose cytarabine based multiagent sequential chemotherapy (SDMSC) is more beneficial than high-dose cytarabine (HDAC) monotherapy in consolidation for the survival of adult acute myeloid leukemia (AML) patients. Methods One hundred and eighty-three AML patients with complete remission (CR) were evaluated. Results and discussion The 3-year relapse rate was 33.4% in the HDAC group and 50.5% in the SDMSC group ( p =0.066). The 3-year overall survival (OS) and event-free survival (EFS) rates in the HDAC group (69.2%, 60.7%) were significantly higher than that in the SDMSC group (50.8%, 42.1%) ( p =0.025, 0.019). For patients in the intermediate risk group, the 3-year OS and EFS rates in the HDAC group (72.5%, 56.7%) were higher than that in the SDMSC group (49.1%, 38.0%) ( p =0.028, 0.093). This study indicates that for young adult AML patients, HDAC consolidation achieves a higher long-term survival than SDMSC, especially for patients in the intermediate-risk group according to the 2022 ELN risk stratification.

    Abstract: Introduction: Hemicellulose is an important component in lignocellulose materials, which is second only to cellulose, accounting for 15%–35% of the dry weight of plants. In the current situation of energy shortage, making full use of lignocellulose materials to produce fuel ethanol has become an important way to solve the energy problem. Xylanase plays a crucial role in the utilization of hemicellulose. It is a necessary means to reduce the cost of hemicellulose utilization by improving the activity of xylanase. Moreover, most naturally xylanases are mesophilic enzymes, which limits their industrial application. Methods: In this study, Myceliophthora thermophila was used to produce xylanases and a thermostable mutant M 2103 was obtained by atmospheric room temperature plasma (ARTP) mutagenesis. The research work started with exploring the effects of ARTP mutagenesis on the antioxidase system [superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), polyphenol oxidase (PPO), and antioxidant capacity (AOC)] of M. thermophile , and found that superoxide dismutase activity increased by 221.13%, and polyphenol oxidase activity increased by 486.04% as compared with the original strain when the implantation time was 300 s. So as to determine the conditions for subsequent mutagenesis. Results and Discussion: For the mutant M 2103, the reaction temperature for xylanase production remained stable in the range of 70°C–85°C. Its optimum temperature was 75°C, which was 15°C higher than that of the original strain. And its xylanase activity increased by 21.71% as compared with the original strain. M 2103 displayed a significantly higher relative xylanase activity than the original strain in the acidic (pH 4.0–7.0) range, and the xylanase activity was relatively stable in the pH range of 6.0–8.5. These results provide an alternative biocatalyst for the production of xylooligosaccharide, and a potential usage of ARTP in the mutagenesis of thermostable mutant.

    Abstract: The gamma-ray spectrum analysis is an important method for quantitative analysis of radionuclide. Although widely used, the weak peak identification and overlapping peaks resolution are still difficult for gamma-ray spectrum analysis. To solve the problem, a new method based on compressed sensing is proposed for improving gamma-ray spectrum analysis in this paper. The proposed method models physical modulation of gamma spectrometer as a linear equation, and formulates the gamma-ray spectrum analysis as a corresponding inverse problem. The true gamma spectrum is obtained by solving the inverse problem by applying sparsity constraint under the framework of compressed sensing. The feasibility of the proposed method is demonstrated by both numerical simulation and Monte Carlo simulation experiments. Results demonstrate that the proposed method can simultaneously resolve overlapped peaks and reduce the fluctuations of gamma-ray spectrum, effectively improving the accuracy of gamma-ray spectrum analysis.

    Abstract: The automatic ball balancer is equipment used to balance the rotor system online and control its forced vibration. Although the rotor system is a nonlinear system actually, especially the occurrence of the nonlinear elastic restoring force in the support of rolling elements bearings, many researches still focus on the linear rotor systems at present. The Jeffcott rotor acted by unsymmetrical nonlinear restoring force is studied in this paper. Through the resolving theoretically and simulating numerically, the principal resonance respond of the rotor system controlled by the balancer with two balls and its stability are studied, while the vibration characteristics and movement laws of the rotor and balls in every rotary speed region are analyzed. The results showed that the balancer can suppress the principal vibration of the nonlinear rotor system in high speed region very well.

    Abstract: Against defects of lacking consideration of air absorption characteristics to the high frequency signal in Schroeder reverberation model, and the more complex structure in Moorer reverberation model, use equiripple Chebyshev approximation criterion to design 4 FIR comb filter with low-pass characteristics and make the FIR filters in parallel, to get a pure parallel reverberation model. This reverberation model considers the absorption characteristics of air to the high-frequency signal, and has a more simple structure.
