    Abstract: Chaeturichthys stigmatias Richardson, 1844, a fish species of the family Gobiidae, is an offshore warm-temperate fish species and a dominant component of estuarine ecosystems. In this study, restriction site-associated DNA sequencing was adopted to analyze the traits of candidate microsatellite markers for C. stigmatias , and 30 polymorphic loci were developed. A total of 5631 microsatellites with primer fragments were detected, among which trinucleotide repeats (57.56% of the total simple sequence repeats) were the most abundant, followed by di- (23.30%), tetra- (11.79%), penta- (4.14%), and hexa- (3.21%) nucleotide repeats type. The numbers of alleles per locus ranged from 6 to 14 with the mean value of 10.4. The mean value of observed heterozygosity and the expected heterozygosity were 0.349 and 0.870, respectively. The microsatellite locus with the lowest polymorphic information content (PIC) was 0.749, which indicated that all sites were highly polymorphic (PIC & gt; 0.50). This is the first microsatellite development and characterization of this species to be reported.

    Abstract: The resistance of methicillin-resistant Staphylococcus aureus (MRSA) has augmented due to the abuse of antibiotics, bringing about difficulties in the treatment of infection especially with the formation of biofilm. Thus, it is essential to develop antimicrobials. Here we synthesized a novel small-molecule compound, which we termed SYG-180-2-2 (C 21 H 16 N 2 OSe), that had antibiofilm activity. The aim of this study was to demonstrate the antibiofilm effect of SYG-180-2-2 against clinical MRSA isolates at a subinhibitory concentration (4 μg/ml). In this study, it was showed that significant suppression in biofilm formation occurred with SYG-180-2-2 treatment, the inhibition ranged between 65.0 and 85.2%. Subsequently, confocal laser scanning microscopy and a bacterial biofilm metabolism activity assay further demonstrated that SYG-180-2-2 could suppress biofilm. Additionally, SYG-180-2-2 reduced bacterial adhesion and polysaccharide intercellular adhesin (PIA) production. It was found that the expression of icaA and other biofilm-related genes were downregulated as evaluated by RT-qPCR. At the same time, icaR and codY were upregulated when biofilms were treated with SYG-180-2-2. Based on the above results, we speculate that SYG-180-2-2 inhibits the formation of biofilm by affecting cell adhesion and the expression of genes related to PIA production. Above all, SYG-180-2-2 had no toxic effects on human normal alveolar epithelial cells BEAS-2B. Collectively, the small-molecule compound SYG-180-2-2 is a safe and effective antibacterial agent for inhibiting MRSA biofilm.

    Abstract: The significant increase in resistance of methicillin-resistant Staphylococcus aureus (MRSA) to fusidic acid (FA) is a worrying public concern. However, the data on the prevalence of FA-resistant MRSA isolates in China is still limited. This study aims to investigate the prevalence of FA resistance and resistance determinants among MRSA isolates from six tertiary hospitals in different regions of China between 2016 and 2020. The antimicrobial susceptibility of MRSA isolates was performed by disk diffusion test and broth microdilution method. Whole-genome sequencing was conducted to evaluate the determinants of FA resistance and molecular characterization of FA-resistant MRSA isolates. In this study, a total of 74 (74/457, 16.2%) isolates were identified to be FA-resistant among 457 non-duplicate MRSA isolates. The prevalence of 74 FA-resistant isolates was as follows: Hubei (28/70, 40%), Shanghai (18/84, 21.4%), Jiangxi (7/58, 12.1%), Inner Mongolia Autonomous Region (6/38, 15.8%), Guangdong (12/112, 10.7%), and Sichuan (3/95, 3.2%). The mutations in fusA were present in 79.7% (59/74) of FA-resistant MRSA isolates, with 54 (54/74, 73%) having L461K mutation and conferring high-level resistance [Minimum Inhibitory Concentration (MIC) & gt;128 μg/ml]. Acquired gene, fusB , with low-level resistance (MIC & lt;16 μg/ml) was found in 20.3% (15/74) FA-resistant MRSA isolates. ST5-MRSA-II-t2460 was the most prevalence clone with high-level resistance, accounting for 51.4% (38/74), which was distributed in Hubei (24/28, 85.7%), Inner Mongolia Autonomous Region (4/6, 66.7%), Shanghai (7/18, 38.9%), and Guangdong (3/12, 25%). ST630-t4549 MRSA isolates with low-level resistance were the most common in Jiangxi (3/7, 42.9%) and Sichuan (2/3, 66.7%). In brief, the prevalence of FA resistance among MRSA isolates in China was relatively high with geographic differences. High-level FA resistance was associated mostly with fusA mutations, especially the L461K mutation, whereas fusB usually conferred the low-level resistance to FA. The spread of ST5-MRSA-II-t2460 clone with high-level resistance to FA contributed greatly to the increase of FA-resistant MRSA isolates in most regions, especially in Hubei.

    Abstract: Duck plague virus (DPV) can cause high morbidity and mortality in many waterfowl species within the order Anseriformes. The DPV genome contains 78 open reading frames (ORFs), among which the LORF2, LORF3, LORF4, LORF5, and SORF3 genes are unique genes of avian herpesvirus. In this study, to investigate the role of this unique LORF5 gene in DPV proliferation, we generated a recombinant virus that lacks the LORF5 gene by a two-step red recombination system, which cloned the DPV Chinese virulent strain (DPV CHv) genome into a bacterial artificial chromosome (DPV CHv-BAC); the proliferation law of LORF5-deleted mutant virus on DEF cells and the effect of LORF5 gene on the life cycle stages of DPV compared with the parent strain were tested. Our data revealed that the LORF5 gene contributes to the cell-to-cell transmission of DPV but is not relevant to virus invasion, replication, assembly, and release formation. Taken together, this study sheds light on the role of the avian herpesvirus-specific gene LORF5 in the DPV proliferation life cycle. These findings lay the foundation for in-depth functional studies of the LORF5 gene in DPV or other avian herpesviruses.

    Abstract: Developing evidence-based planning interventions for promoting physical activity (PA) is considered an effective way to address urban public health issues. However, previous studies exploring how the built environment affects PA over-relied on small-sample survey data, lacked human-centered measurements of the built environment, and overlooked spatially-varying relationships. To fill these gaps, we use cycling and running activity trajectories derived from the Strava crowdsourcing data to comprehensively measure PA in the central city area of Chengdu, China. Meanwhile, we introduce a set of human-scale, eye-level built environment factors such as green, sky, and road view indexes by extracting streetscape characteristics from the Baidu street-view map using the fully Convolutional Neural Network (CNN). Based on these data, we utilize the geographically weighted regression (GWR) model to scrutinize the spatially heterogeneous impact of the built environment on PA. The results are summarized as follows: First, model comparisons show that GWR models outperform global models in terms of the goodness-of-fit, and most built environment factors have spatially varying impacts on cycling and running activities. Second, the green view index restrains cycling activities in general. In contrast, it has a wide-ranging and positive impact on running activities while hampers them in the PA-unfriendly old town. Third, the sky view index stimulates cycling activities in most areas. However, it has a mixed influence on running activities. Fourth, the road view index widely promotes cycling and running activities but hinders them in some areas of the old town dominated by automobiles and under construction. Finally, according to these empirical findings, we propose several recommendations for PA-informed planning initiatives.

    Abstract: The construction of new urban areas has become one of the important ways for urban regeneration throughout the process of polycentric urban development. New urban area construction following transit-oriented development (TOD) conception advocates development around the public transportation stations. To gain more benefits around the stations, it is necessary to conduct an ex-ante evaluation of TOD projects in the new urban area. The Node-place (NP) model is a commonly used method for TOD evaluation and classification, which essentially designs an analytical framework for assessing station areas in both transport (node) and land use (place) aspects. The objective of our study is twofold. First, based on the original NP model, we propose the node-place-system support (NPS) model by introducing a novel evaluation dimension—system support—which quantitatively describes the relationship between local stations and the overall urban system. Second, taking advantage of multi-sourced data and Geographical Information System (GIS) techniques, we employ the proposed NPS model to evaluate and classify the metro stations in the Tianfu New Area of Chengdu, China. The results show that most stations present a balanced relationship between transport and land use performances. However, for a fraction of these balanced stations, we observe a mismatch between the system support and NP performances. Accordingly, we identify the system-mismatched stations and provide targeted improvement strategies for urban design.

    Abstract: In this research, we described a very rare case of secondary lupus nephritis associated with B-cell lymphoma. An 84-year-old man was hospitalized at our institute for lower extremity edema persisting for over 2 months. He was diagnosed with systemic lupus erythematosus based on clinical and laboratory criteria, which showed impaired renal function and nephrotic syndrome with predominant hematuria. Renal biopsy showed IV+V lupus nephritis with highly infiltrated lymphoid cells in the kidney. Secondary lupus nephritis was suspected based on the possible pathogenesis of glomerular injury due to mantle cell lymphoma. Low-dose dexamethasone, rituximab, and lenalidomide were immediately started on the patient, and his renal function was improved after the first cycle of chemotherapy.

    Abstract: The lack of physical activity has become a rigorous challenge for many countries, and the relationship between physical activity and the built environment has become a hot research topic in recent decades. This study uses the Strava Heatmap (novel crowdsourced data) to extract the distribution of cycling and running tracks in central Chengdu in December 2021 (during the COVID-19 pandemic) and develops spatial regression models for numerous 500 × 500 m grids ( N = 2,788) to assess the impacts of the built environment on the cycling and running intensity indices. The findings are summarized as follows. First, land-use mix has insignificant effects on the physical activity of residents, which largely contrasts with the evidence gathered from previous studies. Second, road density, water area, green space area, number of stadiums, and number of enterprises significantly facilitate cycling and running. Third, river line length and the light index have positive associations with running but not with cycling. Fourth, housing price is positively correlated with cycling and running. Fifth, schools seem to discourage these two types of physical activities during the COVID-19 pandemic. This study provides practical implications (e.g., green space planning and public space management) for urban planners, practitioners, and policymakers.

    Abstract: Transit-oriented development (TOD) has been regarded as an effective way to improve urban vibrancy and facilitate affordable, equitable, and livable communities in metro station areas (MSAs). Previous studies placed great attention on the interplay between the MSA-level built environment and overall human activities while neglecting the heterogeneity among different age groups. To address this gap, we leverage the mobile phone signaling data to quantify the spatio-temporal distribution of the MSA-level human activities among different age groups as measured by the vibrancy index (VI). Furthermore, we investigate the impact of the MSA-level built environment on the VI and its intergenerational differences by employing multiple linear regressions based on multi-sourced data. To this end, Chengdu—a TOD-thriving megacity in China—is chosen as a case study. The results indicate that: (1) Residential and bus stop density are positively associated with the VI. And the magnitudes of the correlation coefficients are similar among different age groups. (2) Distance to CBD is negatively associated with the VI of teenagers (12–18 years), middle-aged adults (40–59 years), and older adults (above 60 years) but unrelated to the VI of young adults (19–39 years). (3) Employment density is positively associated with the VI of young and middle-aged adults but insignificantly associated with the VI of teenagers and older adults. (4) The correlations between the floor area ratio and the VI are positive for all age groups. As age increases, the significance of such correlations becomes more pronounced. (5) Streetscape greenery shows a more significant positive correlation with the VI of teenagers and older adults as compared to those of young and middle-aged adults. (6) Significant negative correlations exist between housing price and the VI of different age groups. The findings can inform the development and design of vibrant TOD communities.
