    Abstract: Background: Necroptosis is closely related to the tumorigenesis and development of cancer. An increasing number of studies have demonstrated that targeting necroptosis could be a novel treatment strategy for cancer. However, the predictive potential of necroptosis-related long noncoding RNAs (lncRNAs) in lung adenocarcinoma (LUAD) still needs to be clarified. This study aimed to construct a prognostic signature based on necroptosis-related lncRNAs to predict the prognosis of LUAD. Methods: We downloaded RNA sequencing data from The Cancer Genome Atlas database. Co-expression network analysis, univariate Cox regression, and least absolute shrinkage and selection operator were adopted to identify necroptosis-related prognostic lncRNAs. We constructed the predictive signature by multivariate Cox regression. Kaplan–Meier analysis, time-dependent receiver operating characteristics, nomogram, and calibration curves were used to validate and evaluate the signature. Subsequently, we used gene set enrichment analysis (GSEA) and single-sample gene set enrichment analysis (ssGSEA) to explore the relationship between the predictive signature and tumor immune microenvironment of risk groups. Finally, the correlation between the predictive signature and immune checkpoint expression of LUAD patients was also analyzed. Results: We constructed a signature composed of 7 necroptosis-related lncRNAs (AC026355.2, AC099850.3, AF131215.5, UST-AS2, ARHGAP26-AS1, FAM83A-AS1, and AC010999.2). The signature could serve as an independent predictor for LUAD patients. Compared with clinicopathological variables, the necroptosis-related lncRNA signature has a higher diagnostic efficiency, with the area under the receiver operating characteristic curve being 0.723. Meanwhile, when patients were stratified according to different clinicopathological variables, the overall survival of patients in the high-risk group was shorter than that of those in the low-risk group. GSEA showed that tumor- and immune-related pathways were mainly enriched in the low-risk group. ssGSEA further confirmed that the predictive signature was significantly related to the immune status of LUAD patients. The immune checkpoint analysis displayed that low-risk patients had a higher immune checkpoint expression, such as CTLA-4, HAVCR2, PD-1, and TIGIT. This suggested that immunological function is more active in the low-risk group LUAD patients who might benefit from checkpoint blockade immunotherapies. Conclusion: The predictive signature can independently predict the prognosis of LUAD, helps elucidate the mechanism of necroptosis-related lncRNAs in LUAD, and provides immunotherapy guidance for patients with LUAD.

    Abstract: BBX (B-box) proteins play a vital role in light-induced anthocyanin biosynthesis. PpBBX18 was an indispensable regulator for the induction of anthocyanin biosynthesis in the peel of red pear fruit ( Pyrus pyrifolia Nakai.). However, the upstream regulation of BBX genes has not been well characterized. In this study, PpZAT5, a cysteine2/histidine2-type transcription factor, was discovered as the upstream negative regulator of PpBBX18. The results showed that PpZAT5 functions as a transcriptional repressor and directly binds to the CAAT motif of PpBBX18 and inhibits its expression. PpZAT5 expression was inhibited by light, which is converse to the expression pattern of anthocyanin-related structural genes. In addition, less anthocyanin accumulated in the PpZAT5- overexpressing pear calli than in the wild-type pear calli; on the contrary, more anthocyanin accumulated in PpZAT5-RNAi pear calli. Moreover, the crucial genes involved in light-induced anthocyanin biosynthesis were markedly down-regulated in the transcriptome of PpZAT5 overexpression pear calli compared to wild-type. In conclusion, our study indicates that PpBBX18 is negatively regulated by a C2H2-type transcriptional repressor, PpZAT5, which reduces anthocyanin content in pear. The present results demonstrate an upstream molecular mechanism of PpBBX18 and provide insights into light-induced anthocyanin biosynthesis.

    Abstract: E3 ubiquitin ligases, an important part of ubiquitin proteasome system, catalyze the covalent binding of ubiquitin to target substrates, which plays a role in protein ubiquitination and regulates different biological process. DNA damage response (DDR) is induced in response to DNA damage to maintain genome integrity and stability, and this process has crucial significance to a series of cell activities such as differentiation, apoptosis, cell cycle. The NEDD4 family, belonging to HECT E3 ubiquitin ligases, is reported as regulators that participate in the DDR process by recognizing different substrates. In this review, we summarize recent researches on NEDD4 family members in the DDR and discuss the roles of NEDD4 family members in the cascade reactions induced by DNA damage. This review may contribute to the further study of pathophysiology for certain diseases and pharmacology for targeted drugs.

    Abstract: Previous studies had suggested that electroacupuncture (EA) can promote white adipose tissue (WAT) browning to counter obesity. But the mechanism was still not very clear. Aim In this study, we aim to study the effect of EA on promoting inguinal WAT (iWAT) browning and its possible mechanism. Method Three-week-old rats were randomly divided into a normal diet (ND) group and a high-fat diet (HFD) group. After 10 weeks, the HFD rats were grouped into HFD + EA group and HFD control group. Rats in the EA group were electro-acupunctured for 4 weeks on Tianshu (ST25) acupoint under gas anesthesia with isoflurane, while the rats in HFD group were under gas anesthesia only. Body weight and cumulative food intake were monitored, and H & amp;E staining was performed to assess adipocyte area. The effect of EA on WAT was assessed by qPCR, immunoblotting, immunoprecipitation and Co-immunoprecipitation. Mitochondria were isolated from IWAT to observe the expression of mitochondrial transcription factor A (TFAM). Results The body weight, WAT/body weight ratio and cumulative food consumption obviously decreased (P & lt; 0.05) in the EA group. The expressions of brown adipose tissue (BAT) markers were increased in the iWAT of EA rats. Nevertheless, the mRNA expressions of WAT genes were suppressed by 4-week EA treatment. Moreover, EA increased the protein expressions of SIRT-1, PPAR γ , PGC-1 α , UCP1 and PRDM16 which trigger the molecular conversion of iWAT browning. The decrease of PPAR γ acetylation was also found in EA group, indicating EA could advance WAT-browning through SIRT-1 dependent PPAR γ deacetylation pathway. Besides, we found that EA could activate AMPK to further regulate PGC-1 α -TFAM-UCP1 pathway to induce mitochondrial biogenesis. Conclusion In conclusion, EA can remodel WAT to BAT through inducing SIRT-1 dependent PPAR γ deacetylation, and regulating PGC-1 α -TFAM-UCP1 pathway to induce mitochondrial biogenesis. This may be one of the mechanisms by which EA affects weight loss.

    Abstract: As an important ecosystem type in the coastal zone, mangroves have important ecological functions, such as maintaining coastal biodiversity, preventing wind and consolidating the coast, promoting silt and building land. It is of great significance to understand the protected status of mangroves in the context of climate change and rapid urbanization. Based on the mangrove classification data from remote sensing interpretation, through vacancy analysis, the in-situ protection status of mangroves in China is analyzed. The results show that the total area of mangroves distributed in China is 264 km2 (excluding the statistical data of Hong Kong, Macao and Taiwan), of which 61.4% are protected in natural reserves. In terms of the main provinces where mangroves are distributed, the mangrove area distributed in Hainan Province is small but the protection proportion is high, while the mangrove area distributed in Guangxi and Guangdong Province is large but the proportion of protected areas is relatively low. Among the three mangrove types, Rhizophora apiculate-Xylocarpus granatum and Rhizophora stylosa-Bruguiera gymnorrhiza had high proportions ( 〉 90%) covered by reserves, but relatively small areas. In contrast, Kandelia candel-Aegiceras corniculatum-Avicennia marina had relatively low reserve coverage (52.6%), but a large area. The study puts forward the key areas of mangrove distribution outside the nature reserve, and suggests that they should be protected by delimiting ecological protection red lines.

    Abstract: Atherosclerosis (AS) is the main cause of cardiovascular disease (CVD) and is characterized by endothelial damage, lipid deposition, and chronic inflammation. Gut microbiota plays an important role in the occurrence and development of AS by regulating host metabolism and immunity. As human mitochondria evolved from primordial bacteria have homologous characteristics, they are attacked by microbial pathogens as target organelles, thus contributing to energy metabolism disorders, oxidative stress, and apoptosis. Therefore, mitochondria may be a key mediator of intestinal microbiota disorders and AS aggravation. Microbial metabolites, such as short-chain fatty acids, trimethylamine, hydrogen sulfide, and bile acids, also affect mitochondrial function, including mtDNA mutation, oxidative stress, and mitophagy, promoting low-grade inflammation. This further damages cellular homeostasis and the balance of innate immunity, aggravating AS. Herbal medicines and their monomers can effectively ameliorate the intestinal flora and their metabolites, improve mitochondrial function, and inhibit atherosclerotic plaques. This review focuses on the interaction between gut microbiota and mitochondria in AS and explores a therapeutic strategy for restoring mitochondrial function and intestinal microbiota disorders using herbal medicines, aiming to provide new insights for the prevention and treatment of AS.

    Abstract: Tendon injury is a common disease of the musculoskeletal system, accounting for roughly 30%–40% of sports system disorder injuries. In recent years, its incidence is increasing. Many studies have shown that low-level laser therapy (LLLT) has a significant effect on tendon repair by firstly activating cytochrome C oxidase and thus carrying out the photon absorption process, secondly acting in all the three phases of tendon repair, and finally improving tendon recovery. The repair mechanisms of LLLT are different in the three phases of tendon repair. In the inflammatory phase, LLLT mainly activates a large number of VEGF and promotes angiogenesis under hypoxia. During the proliferation phase, LLLT increases the amount of collagen type III by promoting the proliferation of fibroblasts. Throughout the remodeling phase, LLLT mainly activates M2 macrophages and downregulates inflammatory factors, thus reducing inflammatory responses. However, it should also be noted that in the final phase of tendon repair, the use of LLLT causes excessive upregulation of some growth factors, which will lead to tendon fibrosis. In summary, we need to further investigate the functions and mechanisms of LLLT in the treatment of tendon injury and to clarify the nature of LLLT for the treatment of diverse tendon injury diseases.

    Abstract: Valley photonic crystals (PCs) play a crucial role in controlling light flow and realizing robust nanophotonic devices. In this study, rotated gradient valley PCs are proposed to realize topological rainbow trapping. A topological rainbow is observed despite the presence of pillars of different shapes, which indicates the remarkable universality of the design. Then, the loss is introduced to explore the topological rainbow trapping of the non-Hermitian valley PC. For the step-angle structure, the same or different losses can be applied, which does not affect the formed topological rainbow trapping. For a single-angle structure, the applied progressive loss can also achieve rainbow trapping. The rainbow is robust and topologically protected in both Hermitian and non-Hermitian cases, which is confirmed by the introduction of perturbations and defects. The proposed method in the current study presents an intriguing step for light control and potential applications in optical buffering and frequency routing.

    Abstract: Social interaction is a dynamic and variable process. However, most hyperscanning studies implicitly assume that inter-brain synchrony (IBS) is constant and rarely investigate the temporal variability of the multi-brain networks. In this study, we used sliding windows and k-mean clustering to obtain a set of representative inter-brain network states during different group communication tasks. By calculating the network parameters and temporal occurrence of the inter-brain states, we found that dense efficient interbrain states and sparse inefficient interbrain states appeared alternately and periodically, and the occurrence of efficient interbrain states was positively correlated with collaborative behaviors and group performance. Moreover, compared to common communication, the occurrence of efficient interbrain states and state transitions were significantly higher during creative communication, indicating a more active and intertwined neural network. These findings may indicate that there is a close correspondence between inter-brain network states and social behaviors, contributing to the flourishing literature on group communication.
