    Abstract: γδ T cells are essential for anti-leukemia function in immunotherapy, however, γδ T cells have different functional subsets, including regulatory cell subsets expressing the Foxp3. Whether they are correlated with immune-checkpoint mediated T cell immune dysfunction remains unknown in patients with acute myeloid leukemia (AML). Methods In this study, we used RNA-seq data from 167 patients in TCGA dataset to analyze the correlation between PD-1 and FOXP3 genes and these two genes’ association with the prognosis of AML patients. The expression proportion of Foxp3 + /PD-1 + cells in γδ T cells and two subgroups Vδ1 and Vδ2 T cells were performed by flow cytometry. The expression level of FOXP3 and PD-1 genes in γδ T cells were sorted from peripheral blood by MACS magnetic cell sorting technique were analyzed by quantitative real-time PCR. Results We found that PD-1 gene was positively correlated with FOXP3 gene and highly co-expressed PD-1 and FOXP3 genes were associated with poor overall survival (OS) from TCGA database. Then, we detected a skewed distribution of γδ T cells with increased Vδ1 and decreased Vδ2 T cell subsets in AML. Moreover, significantly higher percentages of PD-1 + γδ, Foxp3 + γδ, and PD-1 + Foxp3 + γδ T cells were detected in de novo AML patients compared with healthy individuals. More importantly, AML patients containing higher PD-1 + Foxp3 + γδ T cells had lower OS, which might be a potential therapeutic target for leukemia immunotherapy. Conclusion A significant increase in the PD-1 + Foxp3 + γδ T cell subset in AML was associated with poor clinical outcome, which provides predictive value for the study of AML patients.

    Abstract: Chronic hepatitis B (CHB) patients with severe liver fibrosis would be more likely to progress to a poorer prognosis. Treatment is considered once the liver fibrosis reaches significant liver fibrosis (≥S2). Leukocyte cell-derived chemotaxin-2 (LECT2) has been shown to contribute to liver fibrosis progression. No research has focused on the role of LECT2 in liver fibrosis in CHB patients. This study enrolled 227 CHB patients and divided them into the training group ( n = 147) and validation group ( n = 80), respectively. The expression of LECT2 in serum, protein and mRNA of the human liver tissues was detected to analyze the possible associations between LECT2 and liver fibrosis. A receiver operating characteristic curve (ROC) was used to estimate the efficacy of LECT2 for predicting liver fibrosis. The data showed that there was a positive relationship between LECT2 and the progression of liver fibrosis. In the training group, LECT2 was demonstrated to have better effectiveness than APRI and FIB-4. The AUC was 0.861, 0.698, and 0.734 for significant liver fibrosis, and 0.855, 0.769, and 0.752 for advanced liver fibrosis. Besides, the efficacy of LECT2 in different statuses of patients with CHB was examined and the effectiveness of LECT2 had also been confirmed in the validation group. All the results confirmed that LECT2 could act as a perfect predictor and thus offers a novel and direct biomarker to estimate liver fibrosis more accurately.

    Abstract: The needs and capacities to achieve the 17 Sustainable Development Goals (SDGs) differ across regions and nations, but little research has been done to investigate their similarities and differences. Here, we proposed using SDG bundles (i.e. groups of regions with similar performances on all individual SDGs) to classify regions when assessing SDG progress and applied the method at the provincial level in China from 2000 to 2015. Five SDG bundles with distinct characteristics were identified. The dominant bundles changed from ‘poor performance for all SDGs’ in 2000 to ‘high scores for environmental and some social SDGs and intermediate scores for others’ and ‘low scores for environmental SDGs but high scores for others’ in 2015, indicating the overall improvement of China’s sustainable development level. However, no bundle had relatively high scores in all SDGs, implying that China has much work left to do. Changes in the SDG bundles across space and time were related to regional socioeconomic development, climate, and geographic conditions. This study sheds light on identifying regions’ strengths and weaknesses in achieving all SDGs, which can inform targeted sustainability actions for regions within certain SDG bundles and promote collaborations among regions with different bundles.

    Abstract: The aim of this paper was to improve the automation of track inspection, data utilization rate, informationization of maintenance dispatch. We designed a track inspection vehicle based on multi-sensor information fusion, then gave the method of coordinate transformation and data fusion. And then researched the service of data displaying, data management and data analysis, proposed the system architecture and network topology, realized the system deployment on the cloud. And then demonstrated stability by comparing the results of two machine tests, and proved accuracy by comparing the results of machine tests with manual tests. The result shows that it had a high accuracy and stability. At last, designed the reserve of cloud platform function extension, discussed the method of data analysis. Research shows that, the track inspection vehicle can measure the track geometric parameter. The application of cloud platform technology can reduce the cost of operation, simplify the function extension. The research result has a positive impact on solving the current problem of track inspection, improving passenger comfort and ensuring safe operation.

    Abstract: This paper analyzes the cause of the overshot of the signal measured by D-dot sensor, and points out that the second-order RLC equivalent circuit model can explain the overshot Taking the asymptotic conical antenna as an example, the inductance L of the second-order model is obtained by using CST simulation and Pspice parameter scanning method. The first-order model and the second-order model are used to calculate the electric field waveform. Under the given conditions, the first-order model is used to obtain the electric field waveform, the waveform overshot is serious, the maximum overshoot reaches 27%, while the second-order model is used to improve the ringing phenomenon, And the maximum overshoot is reduced to 5%.The results show that the second-order model is more accurate than the first-order model and can improve the precision of electric field measurement using D-dot sensor.

    Abstract: Background: Cuproptosis, as a recently discovered type of programmed cell death, occupies a very important role in hepatocellular carcinoma (HCC) and provides new methods for immunotherapy; however, the functions of cuproptosis in HCC are still unclear. Methods: We first analyzed the transcriptome data and clinical information of 526 HCC patients using multiple algorithms in R language and extensively described the copy number variation, prognostic and immune infiltration characteristics of cuproptosis related genes (CRGs). Then, the hub CRG related genes associated with prognosis through LASSO and Cox regression analyses and constructed a prognostic prediction model including multiple molecular markers and clinicopathological parameters through training cohorts, then this model was verified by test cohorts. On the basis of the model, the clinicopathological indicators, immune infiltration and tumor microenvironment characteristics of HCC patients were further explored via bioinformation analysis. Then, We further explored the key gene biological function by single-cell analysis, cell viability and transwell experiments. Meantime, we also explored the molecular docking of the hub genes. Results: We have screened 5 hub genes associated with HCC prognosis and constructed a prognosis prediction scoring model. And the model results showed that patients in the high-risk group had poor prognosis and the expression levels of multiple immune markers, including PD-L1, CD276 and CTLA4, were higher than those patients in the low-risk group. We found a significant correlation between risk score and M0 macrophages and memory CD4 + T cells. And the single-cell analysis and molecular experiments showed that BEX1 were higher expressed in HCC tissues and deletion inhibited the proliferation, invasion and migration and EMT pathway of HCC cells. Finally, it was observed that BEX1 could bind to sorafenib to form a stable conformation. Conclusion: The study not only revealed the multiomics characteristics of CRGs in HCC but also constructed a new high-accuracy prognostic prediction model. Meanwhile, BEX1 were also identified as hub genes that can mediate the cuproptosis of hepatocytes as potential therapeutic targets for HCC.
