    Abstract: Farnesyltransferase inhibitors (FTIs) are focus for the treatment of several diseases, particularly in the field of cancer therapy. Their potential, however, goes even further, as a number of studies have evaluated FTIs for the treatment of infectious diseases such as malaria, African sleeping sickness, leishmaniosis, and hepatitis D virus infection. Little is known about protein prenylation mechanisms in human pathogens. However, disruption of IspA , a gene encoding the geranyltranstransferase of Staphylococcus aureus ( S. aureus ) leads to reprogramming of cellular behavior as well as impaired growth and decreased resistance to cell wall-targeting antibiotics. We used an agar well diffusion assay and a time kill assay and determined the minimum inhibitory concentrations of the FTIs lonafarnib and tipifarnib. Additionally, we conducted cell viability assays. We aimed to characterize the effect of these FTIs on S. aureus , methicillin-resistant Staphylococcus aureus (MRSA), Staphylococcus epidermidis ( S. epidermidis ), Escherichia coli ( E. coli ), Enterococcus faecium ( E. faecium ), Klebsiella pneumoniae ( K. pneumoniae ), Pseudomonas aeruginosa ( P. aeruginosa ), and Streptococcus pneumoniae ( S. pneumoniae ). Both the FTIs lonafarnib and tipifarnib were capable of inhibiting the growth of the Gram-positive bacteria S. aureus , MRSA, S. epidermidis , and S. pneumoniae , whereas no effect was observed on Gram-negative bacteria. The analysis of the impact of lonafarnib and tipifarnib on common human pathogens might lead to novel insights into their defense mechanisms and therefore provide new therapeutic targets for antibiotic-resistant bacterial infections.

    Abstract: The aim of the present study was to adapt and validate the Basic Psychological Need Satisfaction and Frustration Scale within self-determination theory (SDT) within the work domain. Confirmatory factor analyses of three Norwegian samples and one English sample as well as multi-group analyses to examine measurement invariance were performed. The results showed that the adapted work-related scale with its six-factor structure fitted the data well in all four samples, and partial measurement invariance was obtained across samples and languages. Furthermore, internal consistencies for the subscales were acceptable and the subscales predicted work-related correlates as expected, demonstrating the criterion validity of the scale. The current study contributes to a unifying measurement for future research on one of the central underpinnings of SDT within the work domain.

    Abstract: Background: A better understanding of the subjective experience of living with Parkinson’s disease (PD) and the factors that influence this experience can be used to improve wellbeing of people with PD (PwP). Objective: To gain more insight in the subjective experience of PD from the PwP’s perspective, and the factors that contribute to this experience. Methods: In this qualitative review, we performed a systematic search of qualitative studies discussing the subjective experience of PD and extracted reported themes (first order themes). Using a meta-ethnographic approach, we categorized the first order themes into second order themes, and created a third order construct: a holistic model of the subjective experience of living with PD. Results: We included 20 studies with a total sample of 279 PwP. Data-extraction yielded 227 first order themes, which were categorized into the second order themes: 1) Awareness, 2) Disruption, 3) Adjustment, 4) The external environment, and 5) The changing self. With these themes, we developed the “model of dialectic change” which conceptualizes life with PD as a transformative journey, wherein PwP employ strategies to stabilize their changeable relationship with their external environment, while simultaneously redefining their self-concept. Conclusion: Our findings indicate that not only the symptoms of PD, but also the manner in which these cause disruptions in the PwP’s interaction with their personal environment and self-concept, determine the subjective experience of PD and quality of life. Some PwP experience problems with adjusting, resulting in psychological distress. This calls for a holistic, multidisciplinary and participatory approach of PD.

    Abstract: Biosensors become increasingly relevant for medical diagnostics, pharmaceutical industry, and environmental technology, for example, to test new drugs easily and reliably or to detect cell growth in changing environmental conditions. Novel materials like graphene are promising candidates to produce biosensors on an industrial scale by means of printing processes. To reach this aim, methods for the reliable and automated production of electrode structures and their coating are required. We present an impedance biosensor in the format of a microtiter plate, fabricated by highly efficient roll-to-roll printing of graphene-based microstructures on large-area polymer foils. Proof-of-principle experiments show the evidence of the suitability of the printed graphene biosensors for impedance-based monitoring of viral cytopathogenicity and its inhibition in the presence of antiviral drugs. The developed system is a promising approach toward cost-efficient impedimetric biosensors for high-throughput screening in vaccine research and antiviral drug development.

    Abstract: To efficiently monitor the COVID-19 pandemic for surveillance purposes, reliable serological rapid diagnostic tests (RDTs) are desirable for settings where well-established high-throughput bench-top solutions are not available. Here, we have evaluated such an RDT. Methods We have assessed the Xiamen AmonMed Biotechnology COVID-19 IgM/IgG test kit (Colloidal gold) and the EUROIMMUN benchtop assay with serum samples from patients with polymerase chain reaction (PCR)-confirmed COVID-19 disease. Samples from patients with Epstein-Barr-virus (EBV) infection and blood donors were used for specificity testing. Results For the colloid gold rapid test and the EUROIMMUN assay, the study indicated overall sensitivity of 15.2% and 67.4%, respectively, while specificity of 99.0% and 97.9% with the blood donor sera, as well as 100% and 96.8% with the EBV-patients, were observed, respectively. An association of the time period between positive PCR results and serum acquisition with serological test positivity could be observed for the immunologlobulin G subclass of the EUROIMMUN assay only. Conclusions In spite of acceptable specificity of the assessed RDT, the detected poor sensitivity leaves room for improvement. The test results remain difficult to interpret and therefore the RDT can currently not be recommended for routine diagnostic or surveillance use.

    Abstract: In Norway music-streaming services have become mainstream in everyday music listening. This paper examines how 12 heavy streaming users make sense of their experiences with Spotify and WiMP Music (now Tidal). The analysis relies on a mixed-method qualitative study, combining music-diary self-reports, online observation of streaming accounts, Facebook and last.fm scrobble-logs, and in-depth interviews. By drawing on existing metaphors of Internet experiences we demonstrate that music-streaming services can make sense as tools, places, and ways of being. Music streaming as lifeworld mediation is discussed as a fourth framework for understanding online music experiences, particularly those arising from mobile and ubiquitous characteristics of contemporary Internet technology.

    Abstract: No standard protocol based on a multidisciplinary framework currently exists for the evaluation, follow-up and interpretation of Cochlear Implant (CI) outcomes in clinical routine. Therefore, this study aimed to develop and clinically implement a multidimensional Cochlear Implant (CI) outcome assessment protocol for adult CI users based on the International Classification of Functioning, Disability and Health (ICF). Methods An international multidisciplinary core group of CI experts selected the most relevant ICF categories and codes for adult CI users out of the existing ICF core set for hearing loss. A well-known, commonly used instrument or method was identified and assigned to each ICF category along with quantified ICF qualifiers. These qualifiers facilitate the interpretation of CI outcomes by categorizing the outcome on a rating scale between 0 and 4, based on the severity of participants' impairment/restriction/limitation/barrier. The ICF-based CI outcome assessment protocol was applied in clinical routine in a prospective longitudinal multicenter study assessing a consecutive sample of 72 adult CI candidates before and 6 months after implantation. For each relevant ICF category, the participants' improvement, stability or deterioration was specified in a descriptive approach using ICF qualifiers. Results Participants either showed improved or stable ICF qualifiers after cochlear implantation across all ICF domains (e.g., Body Functions and Structures, Activities, and Participation and Environmental factors). The ICF qualifiers for environmental sound tolerance ( n = 30 [45%]) and vestibular functions ( n = 21 [33%]) deteriorated most frequently after implantation. ICF qualifiers for falling ( n = 53 [83%]) and dizziness ( n = 40 [62%]) remained stable in most participants. Auditory perception-related and communication-related categories generally improved in most participants after cochlear implantation, with 46 to 73% ( n = 34 to n = 48) of CI users perceiving an improvement postoperatively. Conclusion This study is the first to propose and apply a ready-to-use CI outcome assessment protocol based on the ICF model, which provides a common language and comprehensive assessment protocol for the description and measurement of CI outcomes worldwide. Its straightforward outcome description and rating enables CI outcome interpretation by non-experts, enhancing multidisciplinary communication and knowledge on individual healthcare needs in CI users.
