    Abstract: People living with HIV (PLWH), despite suppression of viral replication with antiretroviral therapy (ART), have high morbidity and mortality due to immune activation and chronic inflammation. Discovering new biomarkers of immune activation status under ART will be pertinent to improve PLWH quality of life when the majority will be treated. We stipulate that plasma large and small extracellular vesicle (EVs) and their microRNA content could be easily measured biomarkers to monitor immune activation in PLWH. Venous blood samples from n = 128 ART-treated PLWH with suppressed viral load (≤ 20 copies/mL) and n = 60 HIV-uninfected participants were collected at five testing or treatment centers of PLWH in Burkina Faso. Large and small plasma EVs were purified, counted, and the mature miRNAs miR-29a, miR-146a, and miR-155 were quantified by RT-qPCR. Diagnostic performances of large and small EVs miRNAs level were evaluated by receiver operating characteristic (ROC) curve analysis and principal component analysis (PCA). Among the EVs microRNA measured, only large EVs miR-155 copies distinguished PLWH with immune activation, with AUC of 0.75 for CD4/CD8 & lt; 1 (95% CI: 0.58–0.91, P = 0.0212), and 0.77 for CD8 T cells ≥ 500/µL (95% CI: 0.63–0.92, P = 0.0096). In addition, PCA results suggest that large EVs miR-155 copies may be a biomarker of immune activation. Since miR-155 may influence immune cell function, its enrichment in large EV subpopulations could be a functional biomarker of immune activation in PLWH on ART. This measure could help to monitor and diagnose the immune activation with more accuracy.

    Abstract: Aim/Background: To reduce maternal mortality (MM), access to family planning and reproductive health care services are key strategies. Access to and use by marginalized populations pose a substantial challenge. The objective of this study was to describe the reproductive history of female sex workers (FSWs) before and during sex work. Materials and methods: FSWs aged ≥18 years were recruited in Benin, and Mali, to answer a questionnaire during a face-to-face interview. Descriptive analyses were carried out and comparisons made between countries (Pearson chi-square) and between the periods before (BSW) and during sex work (DSW) practice within women in each country (McNemar chi-square). In addition, multiple imputations were used to estimate and compare the incidence rate of pregnancy BSW and DSW calculated with a generalized linear model for count data with a Poisson distribution and a log link. Results: Mean age was 26 years in Mali (n=322) and 35 years in Benin (n=330). More women had at least one pregnancy BSW compared to DSW in both Mali (62.1%-vs-33.5%) and Benin (91.2%-vs-32.7%). The pregnancies occurring DSW had lower livebirth rates (57.9%-vs-74.7% Mali, 17.6%-vs-60.6% Benin) and ended more often with a therapeutic abortion, DSW compared to BSW, especially in Benin (65.2%-vs-25.6%). The level of complications associated with therapeutic abortion was high, both DSW (23%) and BSW (20%). The incidence rate of pregnancy was about twice lower DSW compared to BSW [incidence rate ratio (IRR): 0.49; 95% confidence interval (95%CI): 0.37-0.66-Mali and IRR: 0.45; 95% CI: 0.32-0.63-Benin]. Conclusion: In both Benin and Mali, FSWs had more pregnancies during the period preceding s ex work practice, about twice more than during sex work when considering the differences in incidence rates. Single mothers need to be supported to avoid sex work as a financial solution. On the other hand, the reproductive needs of FSWs need to be acknowledged to reduce maternal mortality within this marginalized population. It is of paramount importance that interventions remain focused on reproductive health and prevent unwanted pregnancies, answer contraceptive needs and provide safe therapeutic abortions.

    Abstract: Internal disorder is a major problem in fruit production and is responsible for considerable economical losses. Symptoms are not externally visible, making it difficult to assess the problem. In recent years, 3D fluorescence spectroscopy has been used to reveal features of interest in agronomical field, such as plant stress and plant infection. Such technique could provide useful information regarding changes that occur at the tissue level, in order to distinguish spectral differences between healthy and disordered fruits. This paper introduces the use of the new three-way feature extraction N-CovSel method, compared to the commonly used N-PLS-DA method. These approaches were used upon front-face fluorescence spectra of 27 fruit pulp and skin samples, by analysing excitation wavelengths ranging from 250 to 650 nm, and emission wavelengths varying from 290 to 800 nm. N-CovSel method was applied to identify the most relevant features on: 1) excitation-emission wavelength couples, 2) excitation wavelengths whatever the emission wavelengths and 3) emission wavelengths whatever the excitation wavelengths. Discriminant analysis of the selected features were performed across classes. The constructed models provided key features to differentiate healthy fruits from disordered ones. These results highlighted the capability of N-CovSel method to extract the most fitted features for enhanced fruit classification using front-face fluorescence spectroscopy. They revealed characteristic fluorophores involved in the structural modifications generated by the physiological disorder studied. This paper provides preliminary results concerning the suitability of N-CovSel method for the desired application. Further investigations could be performed on intact fresh fruits in a non-destructive way, allowing an earlier and faster detection of the internal disorder for in-field or industrial applications.

    Abstract: Background: Extracellular vesicles (EVs) are intercellular messengers with epigenetic potential since they can shuttle microRNA (miRNA). EVs and miRNA play a role in human immunodeficiency virus (HIV) infection immunopathogenesis. Chronic immune activation and systemic inflammation during HIV infection despite effective antiretroviral therapy (ART) are associated with non-acquired immunodeficiency syndrome (AIDS) comorbidities in people living with HIV (PLWH). Analysis of plasma EVs and their miRNA content may be useful as immune activation or inflammatory biomarkers in PLWH receiving ART. In this study, we hypothesized that the number, size, and miRNA of large and small EVs could reflect immune activation associated with an elevated CD8 T-cell count or a low CD4/CD8 ratio in PLWH.Methods: Plasma EVs subtype purified from PLWH and uninfected controls were sized using dynamic light scattering and quantified using flow cytometry and acetylcholine esterase (AChE) activity. Expression of mature miRNAs miR-92, miR-155, miR-223 was measured by quantitative reverse-transcriptase polymerase chain reaction in EVs and leucocytes.Results: HIV infection induces increased production of small EVs in plasma. EV subtypes were differentially enriched in miR-92, miR-155, and miR-223. Positive correlations between CD8 T-cell count and large EVs abundance and small EVs AChE activity were observed. CD4/CD8 ratio was negatively correlated with small EV AChE activity, and miRNA-155 level per small EV was negatively correlated with CD8 T-cell count.Conclusions: These findings suggest that quantifying large or small EVs and profiling miRNA content per EV might provide new functional biomarkers of immune activation and inflammation.
