    Kurzfassung: Nitrapyrin, a nitrification inhibitor, produces liver tumors in B6C3F1 mice. In a 2-year oncogenicity study, increased incidence of mice with hepatocellular tumors was observed following exposure to 125 (females only) or 250 mg/kg/day (males and females) nitrapyrin in the diet. Previous data was generated in male mice to support a mode-of-action (MoA) characterized by constitutive androstane receptor (CAR) nuclear receptor (NR) activation, increased hepatocellular proliferation, and subsequent hepatocellular foci and tumor formation. Uncertainty as to the relevance of this MoA for females remained given the increased sensitivity to tumor formation in female mice. A targeted MoA study was conducted to evaluate CAR activation and hepatic responses in female mice treated with the female carcinogenic dose of nitrapyrin for 4 days. Nitrapyrin induced a treatment-related increase in hepatocellular hypertrophy and hepatocellular proliferation. Nitrapyrin also induced a dose-related increase in the Cyp2b10 /CAR-associated transcript and liver weights. Nitrapyrin-induced liver weights and Cyp2b10 gene expression for both males and females were compared to data generated from three other established CAR activators; methyl isobutyl ketone, phenobarbital, and sulfoxaflor. The response observed in female mice following exposure to nitrapyrin was within range of the degree of change observed in mice following exposure to tumorigenic doses of other CAR activators. Consistent with the liver MoA in male mice, these data support a CAR-mediated mode of action for nitrapyrin-induced liver tumors in female mice, with the understanding that a focused approach minimizing animal use can bridge male and female datasets when sex-specific carcinogenic differences are observed.

    Kurzfassung: SkyPortal is an open-source software package designed to discover interesting transients efficiently, manage follow-up, perform characterization, and visualize the results. By enabling fast access to archival and catalog data, crossmatching heterogeneous data streams, and the triggering and monitoring of on-demand observations for further characterization, a SkyPortal -based platform has been operating at scale for 〉 2 yr for the Zwicky Transient Facility Phase II community, with hundreds of users, containing tens of millions of time-domain sources, interacting with dozens of telescopes, and enabling community reporting. While SkyPortal emphasizes rich user experiences across common front-end workflows, recognizing that scientific inquiry is increasingly performed programmatically, SkyPortal also surfaces an extensive and well-documented application programming interface system. From back-end and front-end software to data science analysis tools and visualization frameworks, the SkyPortal design emphasizes the reuse and leveraging of best-in-class approaches, with a strong extensibility ethos. For instance, SkyPortal now leverages ChatGPT large language models to generate and surface source-level human-readable summaries automatically. With the imminent restart of the next generation of gravitational-wave detectors, SkyPortal now also includes dedicated multimessenger features addressing the requirements of rapid multimessenger follow-up: multitelescope management, team/group organizing interfaces, and crossmatching of multimessenger data streams with time-domain optical surveys, with interfaces sufficiently intuitive for newcomers to the field. This paper focuses on the detailed implementations, capabilities, and early science results that establish SkyPortal as a community software package ready to take on the data science challenges and opportunities presented by this next chapter in the multimessenger era.

    Kurzfassung: We report multiwavelength observations and characterization of the ultraluminous transient AT 2021lwx (ZTF20abrbeie; aka “Barbie”) identified in the alert stream of the Zwicky Transient Facility (ZTF) using a Recommender Engine For Intelligent Transient Tracking filter on the ANTARES alert broker. From a spectroscopically measured redshift of 0.995, we estimate a peak-observed pseudo-bolometric luminosity of log( L max / [ erg s − 1 ] ) = 45.7 from slowly fading ztf- g and ztf- r light curves spanning over 1000 observer-frame days. The host galaxy is not detected in archival Pan-STARRS observations ( g 〉 23.3 mag), implying a lower limit to the outburst amplitude of more than 5 mag relative to the quiescent host galaxy. Optical spectra exhibit strong emission lines with narrow cores from the H Balmer series and ultraviolet semi-forbidden lines of Si iii ] λ 1892, C iii ] λ 1909, and  C ii ] λ 2325. Typical nebular lines in Active Galactic Nucleus (AGN) spectra from ions such as [O ii ] and [O iii ] are not detected. These spectral features, along with the smooth light curve that is unlike most AGN flaring activity and the luminosity that exceeds any observed or theorized supernova, lead us to conclude that AT 2021lwx is most likely an extreme tidal disruption event (TDE). Modeling of ZTF photometry with MOSFiT suggests that the TDE was between a ≈14 M ⊙ star and a supermassive black hole of mass M BH ∼ 10 8 M ⊙ . Continued monitoring of the still-evolving light curve along with deep imaging of the field once AT 2021lwx has faded can test this hypothesis and potentially detect the host galaxy.
