    Kurzfassung: The pathogenesis of ulcerative colitis (UC) is complicated with impaired intestinal epithelial barrier and imbalanced gut microbiota. Both selenium and probiotics have shown effects in regulating intestinal flora and ameliorating UC. The objective of this study is to investigate the alleviating effects of Selenium-enriched Bifidobacterium longum DD98 (Se- B. longum DD98) on dextran sulfate sodium (DSS)-induced colitis in mice and explore the underlying mechanism. After treatment of B. longum DD98, Se- B. longum DD98, and sulfasalazine for 3 weeks, the disease severity of UC mice was decreased, with colon lengthened and pathological phenotype improved. The expression of pro-inflammatory cytokines and oxidative stress parameters were also decreased. Thus, Se- B. longum DD98 showed a stronger effect on relieving the aforementioned symptoms caused by DSS-induced colitis. Exploration of the potential mechanism demonstrated that Se- B. longum DD98 showed higher activities to suppress the inflammatory response by inhibiting the activation of the toll-like receptor 4 (TLR4), compared to B. longum DD98 and sulfasalazine. Se- B. longum DD98 also significantly improved the intestinal barrier integrity by increasing the expression of tight junction proteins including ZO-1 and occludin. 16S rDNA sequencing analyses showed that Se- B. longum DD98 improved the diversity of the intestinal flora and promoted the abundance of health-benefiting taxa including Lachnospiraceae , Lactobacillaceae , and Prevotellaceae in family level. In conclusion, compared to B. longum DD98 and sulfasalazine, Se- B. longum DD98 showed stronger therapeutic effects on DSS-induced colitis in mice and might be a promising candidate for the treatment of UC.

    Kurzfassung: Relapsed or refractory diffuse large B-cell lymphoma (R/R DLBCL) has poor clinical outcomes when treated with conventional salvage chemotherapy. Monotherapy using zanubrutinib, a selective Bruton’s tyrosine kinase (BTK) inhibitor, has achieved modest antitumor effect in R/R DLBCL. Here we aimed to evaluate the efficacy and safety of zanubrutinib plus salvage chemotherapy in R/R DLBCL patients. Methods We retrospectively reviewed R/R DLBCL patients who were administered with zanubrutinib plus salvage chemotherapy in our center between January, 2019 and December, 2021. Targeted panel sequencing of 11 lymphoma-related genes was performed on 8 patients with poor responses to zanubrutinib-based chemotherapy. Results 27 R/R DLBCL patients were enrolled. Median age at this study was 59 years (range, 15-72). The best overall response rate (ORR) was 74.1% and complete remission rate was 33.3%. With a median follow-up of 11 months (range, 1-17), the median progression-free survival (PFS) was 8.1 months, and the overall survival (OS) was not achieved. The most common grade-3/4 adverse events were neutropenia (70.4%), thrombocytopenia (66.7%), and febrile neutropenia (33.3%). In multivariate analysis, early treatment and overall response after chemotherapy were independent favorable prognostic factors for PFS. Overall response after chemotherapy was an independent favorable factor for OS. Among the 8 patients with poor response to zanubrutinib-based treatment, the majority of patients had NOTCH2 mutations (n=8, 100%) and TP53 mutations (n=7, 87.5%). However, these patients achieved an ORR of 75% at 3 months after CD19-CAR-T cell therapy (including 4 cases of complete remission and 2 cases of partial remission). With a median follow-up of 9 months from CAR-T cell infusion (range, 1-16 months), the median PFS was 14.5 months, and the median OS was not reached. Conclusion With high efficacy and manageable tolerability, zanubrutinib plus salvage chemotherapy may be a potential treatment option for R/R DLBCL. CAR-T cell therapy may be a priority strategy for these poor responders to BTKi-based treatment.

    Kurzfassung: Exposure to disasters in early life may induce lifetime health risk, but investigation on earthquake exposure and DM in later life is still limited. The aim of the current study is to evaluate the association between exposure to the Tangshan Earthquake in early life and diabetes mellitus (DM) incidence in adulthood, and explore the modification of lifestyles on DM development. Methods Participants who were free of DM at baseline from the Kailuan Study were included in this study. All participants were divided into fetal-exposed, infant-exposed, early childhood-exposed and nonexposed group. The effect of earthquake exposure on DM and modification of lifestyles were examined by multivariable-adjusted Cox proportional hazard model. Results The exposed group had a higher risk of DM than nonexposed group, especially in infant-exposed and early childhood-exposed group, with hazard ratio (HR) of 1.62 [95% confidence intervals (CI), 1.21–2.17] and 1.46 (95% CI, 1.06–1.99), respectively. After stratifying by lifestyles, a significant modification was observed in alcohol consumption. Conclusion Exposing to earthquake in early life could increase DM incidence in later life, and alcohol consumption might modify the effect of earthquake exposure on DM development. More attention should be paid on the preventions of DM among adults who exposed to earthquake in their early life.

    Kurzfassung: The climate in hot-moist regions like Guangdong is characterized as short in summer and long in winter, which imposes high insulation requirements on the roof of houses. In this case, this paper proposes a roof greening overhead structure, which is developed by making a bracket under the existing planting box after reinforcing the bottom thereof, thus there will be an overhead layer with a height of 300mm between the planting box and the roof. The study used three office rooms with identical conditions on the top floor of the office building as the test rooms. The roofs of the three rooms were set as the roof with the overhead planting box, the roof with the existing planting box and the reference roof, respectively. The temperatures of the structural layers in the three test rooms were measured under the non-air-conditioning closed conditions in summer. The hourly power consumptions of the air-conditioners in the three test rooms were measured under the air-conditioning closed conditions in summer. The results indicates the natural ventilation formed by the overhead structural layer can improve the overall insulation effect of the roof. The energy saving rates of the air conditioners in the room with the overhead planting box on the roof reached 28.2%.

    Kurzfassung: In this article, plasma-assisted NH 3 synthesis directly from N 2 and H 2 over packing materials with different dielectric constants (BaTiO 3 , TiO 2 and SiO 2 ) and thermal conductivities (BeO, AlN and Al 2 O 3 ) at room temperature and atmospheric pressure is reported. The higher dielectric constant and thermal conductivity of packing material are found to be the key parameters in enhancing the NH 3 synthesis performance. The NH 3 concentration of 1344 ppm is achieved in the presence of BaTiO 3 , which is 106% higher than that of SiO 2 , at the specific input energy (SIE) of 5.4 kJ·l −1 . The presence of materials with higher dielectric constant, i.e. BaTiO 3 and TiO 2 in this work, would contribute to the increase of electron energy and energy injected to plasma, which is conductive to the generation of chemically active species by electron-impact reactions. Therefore, the employment of packing materials with higher dielectric constant has proved to be beneficial for NH 3 synthesis. Compared to that of Al 2 O 3 , the presence of BeO and AlN yields 31.0% and 16.9% improvement in NH 3 concentration, respectively, at the SIE of 5.4 kJ·l −1 . The results of IR imaging show that the addition of BeO decreases the surface temperature of the packed region by 20.5% to 70.3°C and results in an extension of entropy increment compared to that of Al 2 O 3 , at the SIE of 5.4 kJ·l −1 . The results indicate that the presence of materials with higher thermal conductivity is beneficial for NH 3 synthesis, which has been confirmed by the lower surface temperature and higher entropy increment of the packed region. In addition, when SIE is higher than the optimal value, further increasing SIE would lead to the decrease of energy efficiency, which would be related to the exacerbation in reverse reaction of NH 3 formation reactions.

    Kurzfassung: Abiotic stress during growth readily reduces cotton crop yield. The different survival tactics of plants include the activation of numerous stress response genes, such as BREVIS RADIX ( BRX ). Methods In this study, the BRX gene family of upland cotton was identified and analyzed by bioinformatics method, three salt-tolerant and cold-resistant GhBRX genes were screened. The expression of GhBRX.1 , GhBRX.2 and GhBRXL4.3 in upland cotton was silenced by virus-induced gene silencing (VIGS) technique. The physiological and biochemical indexes of plants and the expression of related stress-response genes were detected before and after gene silencing. The effects of GhBRX.1 , GhBRX.2 and GhBRXL4.3 on salt and cold resistance of upland cotton were further verified. Results and discussion We discovered 12, 6, and 6 BRX genes in Gossypium hirsutum , Gossypium raimondii and Gossypium arboreum , respectively. Chromosomal localization indicated that the retention and loss of GhBRX genes on homologous chromosomes did not have a clear preference for the subgenomes. Collinearity analysis suggested that segmental duplications were the main force for BRX gene amplification. The upland cotton genes GhBRX.1 , GhBRX.2 and GhBRXL4.3 are highly expressed in roots, and GhBRXL4.3 is also strongly expressed in the pistil. Transcriptome data and qRT‒PCR validation showed that abiotic stress strongly induced GhBRX.1 , GhBRX.2 and GhBRXL4.3 . Under salt stress and low-temperature stress conditions, the activities of superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) and the content of soluble sugar and chlorophyll decreased in GhBRX.1- , GhBRX.2- and GhBRXL4.3 -silenced cotton plants compared with those in the control (TRV: 00). Moreover, GhBRX.1 -, GhBRX.2 - and GhBRXL4.3 -silenced cotton plants exhibited greater malondialdehyde (MDA) levels than did the control plants. Moreover, the expression of stress marker genes ( GhSOS1 , GhSOS2 , GhNHX1 , GhCIPK6 , GhBIN2 , GhSnRK2.6 , GhHDT4D , GhCBF1 and GhPP2C ) decreased significantly in the three target genes of silenced plants following exposure to stress. These results imply that the GhBRX.1 , GhBRX.2 and GhBRXL4.3 genes may be regulators of salt stress and low-temperature stress responses in upland cotton.

    Kurzfassung: Introduction: Some herbal ingredients can reshape the composition of the gut microbiome as well as its metabolites. At the same time, the gut microbiota can also affect drug metabolism. A large number of studies have reported that saponins are biotransformed under the action of intestinal microorganisms to improve drug efficacy and bioavailability. Capilliposide A is a triterpenoid saponin, which is derived from Lysimachia capillipes Hemsl. CPS-A has anti-inflammatory pharmacological activity, but the substance basis in vivo is unknown at present, so studies on the interaction between intestinal microorganisms and CPS-A may clarify the pharmacodynamic substance basis of CPS-A. Methods: This study established a colitis mouse model, collected sterile feces from normal mice and colitis mice, and incubated CPS-A with two different intestinal flora in vitro . Based on LC-MS, the metabolic process of CPS-A mediated by intestinal microbes and the intervention effect of CPS-A on intestinal microbiome derived metabolites were studied. Results: The results of experiments indicate that intestinal microorganisms can mediate the biotransformation of CPS-A and metabolize it into corresponding deglycosylation products, thereby promoting its drug effect. Not only that, CPS-A can also promote metabolites such as Deoxycholic acid, Histamine, 3-Hydroxytridecanoic acid, and Indole-3-acetic acid in the intestinal microbiota of mice with colitis. This may result in anti-colitis effects. CPS-A mainly involved in metabolic pathways such as azathioprine and mercaptopurine, which may also have beneficial or adverse effects. Discussion: This study on the interaction between CPS-A and microbiota provides a new idea for the study of traditional Chinese medicine with poor oral bioavailability. The regulatory effect of CPS-A on the metabolites of intestinal flora in colitis mice was also found. It laid a foundation for exploring the mechanism of action of saponins on colitis mice.

    Kurzfassung: Continuous cropping of soybean leads to soil environment deterioration and soil-borne disease exacerbation, which in turn limits the sustainability of agricultural production. Chitin amendments are considered promising methods for alleviating soybean continuous cropping obstacles; however, the underlying mechanisms of soil sickness reduction remain unclear. In this study, soil amendments with pure and crude chitin at different addition dosages were employed to treat diseased soil induced by continuous cropping of soybean for five years. Chitin amendments, especially crude chitin, remarkably increased soil pH, available phosphorus (AP), potassium (AK) and nitrate nitrogen ( NO 3 − -N) contents, and improved soybean plant growth and soil microbial activities (FDA). Additionally, chitin application significantly enriched the relative abundances of the potential biocontrol bacteria Sphingomonas , Streptomyces , and Bacillus and the fungi Mortierella , Purpureocillium , and Metarhizium while depleted those of the potential plant pathogens Fusarium , Cylindrocarpon and Paraphoma . Moreover, chitin amendments induced looser pathogenic subnetwork structures and less pathogenic cooperation with other connected microbial taxa in the rhizosphere soils. The structural equation model (SEM) revealed that pure and crude chitin amendments promoted soybean plant growth by indirectly regulating soil pH-mediated soil microbial activities and potentially beneficial microbes, respectively. Therefore, the reduction strategies for continuous cropping obstacles by adding pure and crude chitin were distinct; pure chitin amendments showed general disease suppression, while crude chitin exhibited specific disease suppression. Overall, chitin amendments could suppress potential plant pathogens and improve soil health, thereby promoting soybean growth, which provides new prospects for cultivation practices to control soybean continuous cropping obstacles.
