    Abstract: Groundnut leaf miner (Aproaerema modicella. D.) is an important leaf feeding insect in groundnut that significantly reduces the pod yield up to 50 per cent. The growth and development of this insect pest is closely associated with the abiotic weather parameters prevailing in a particular location. Due to paucity of information available on the population dynamics of groundnut leaf miner with abiotic weather factors, a study was undertaken in Erode district of Tamil Nadu during Summer, 2023. Groundnut leaf miner damages were recorded through the fixed plots survey and the adult moth activities were monitored through pheromone traps at weekly intervals. From this study it was found that, the leaf miner incidences were observed from 30 days after sowing. The highest mean leaf miner mean damage (17.84%), pheromone adult catches (58.38 nos./5 traps), coccinellid beetle population (4.70 nos./10 plants) and spider population (5.17 nos./10 plants) were observed during 9th SMW. Whereas, the rove beetle population was maximum (4.62 nos./10 plants) during 10th SMW. The correlation study revealed that the maximum temperature had positive correlation and rainfall had negative correlation with the populations of groundnut leaf miner and its natural enemies. The multiple linear regression analysis indicated that the weather parameters together had 66.00 to 95.00 per cent influence on the groundnut leaf miner and its natural enemy population. The present study clearly indicated that, the population of groundnut leaf miner and its damage levels were closely related to the abiotic weather parameters prevailed in the study area during entire crop duration. Better understanding of the weather factors resulted in better leaf miner management and more net profit for the groundnut growers.

    Abstract: Pearl millet is an ideal fodder crop having highly shiny, lush, palatable, nutrient- dense, excellent silage and biomass production potential and bestowed with various essential nutrients required for the livestock. The study material consists of 54 fodder pearl millet hybrids obtained through the hybridisation between 15 selected superior parents from ICRISAT gene-pool as well as gene pool of Department of Forage Crops, TNAU, Coimbatore. For the better understanding of the relationship between green fodder yield per plant, its contributing traits and various quality traits, genetic parameters such as genotypic and phenotypic correlation as well as path coefficients were analysed. Correlation studies revealed that out of all the seventeen biometric traits studied, all the traits are shown positive correlation with green fodder yield per plant whereas fifteen traits are found to have significant positive correlation with green fodder yield per plant. Ash content (rg =0.855), dry fodder yield per plant (rg =0.850), leaf-stem ratio (rg =0.710) have shown very strong significant positive correlation and plant height (rg =0.641), days to green fodder harvest  (rg =0.630), stem girth (rg =0.616) and days to fifty per cent flowering (rg =0.606) have recorded with moderately strong significant positive correlation with green fodder yield per plant. Number of tillers per plant (0.859), internode length (0.574), ash content (0.421) and dry fodder yield per plant (0.372) revealed highly positive direct effects with green fodder yield per plant. Hence, selection for traits exhibiting positive association and direct effects for green fodder yield would help the breeder to enhance it further.

    Abstract: Banana is parthenocarpic and vegetatively propagated crop due to which in vitro mutagenesis approaches have substantial scope to create variations for crop improvement. Bhimkol cultivar is a wild seeded type and it set seeds naturally. The immature seeds can be used for the in vitro mutation to create variability. Prerequisite in an in vitro mutation study, is to determine the appropriate dosages to produce larger recovery of mutants with diminutive population loss. It is necessary to fix the lethal dose (LD50) for optimizing the radiation doses for immature seeds of banana. In the present investigation, in order to fix the LD50 value immature seeds of banana were subjected to five different doses of gamma irradiation ranged from 5-25 Gy with control (untreated). The present finding was observed that as gamma radiation dose increased, delayed germination and reduction in the germination percentage of immature embryos. And also, seedling height and root length reduced significantly. Based on the probit analysis curve, the germination percentage showed that, LD50 dose of gamma radiation was 15 - 20 Gy for higher germination percentage of immature embryos in banana cv. Bhimkol.

    Abstract: Rabies disease can be preventable through vaccination is the only way for effective control and the vaccine is administered as post exposure prophylaxis method (PEP) along with rabies immunoglobulin. For the preparation of the vaccine, the high titre rabies virus has to be propagated in suitable host system like Vero cells. In this study, the virus was propagated in tissue culture flask in two different tissue culture media namely MEM and RPMI-1640, keeping Vero cell line as the host system. This preliminary study is to find the media that yields a better viral titre as high, the influence of harvesting intervals on viral titre for the reason that the vaccine yield is directly proportional to the viral titre during the virus propagation stage and leads to cost effective vaccine. The Vero cell was revived from passage 154 and the part of cells were subjected for adaptation in RPMI-1640 media with gradual media replacement. The passaging of RPMI 1640 media cells were continued until it reaches the equal cell count of MEM (p-158). The confluent monolayer of Vero cells was maintained in the same passage level with appropriate media. The cell count of MEM media, RPMI-1640 were 10.28 x 106 and 10.35 x 106 respectively in 25cm2 tissue culture flasks. The Vero cells were sub-cultured in 175 cm2 tissue culture flasks infected with 0.2 MOI (Multiplicity of Infection) of virus and the viral titration was estimated through FAT test (Fluorescent Antibody Test). The highest viral titre obtained from RPMI-1640 media batch (10-6.125/ml) and MEM media batch (10-6.25 /ml). The two days interval viral harvests shows  (Batch 1 & 3) the viral titre log ranged between 10-4.375 to 10-5.875 and three days interval harvests (Batch 2 & 4)  the viral titre log ranged between 10-3.750 to 10-6.250 per mL.

    Abstract: Bhimkol (Musa balbisiana, BB) is a wild diploid seeded cultivar, belonging to the family Musaceae. It is tolerance to pest and diseases, since it has high demand for commercialization for its high nutritive values. Therefore, the present study was undertaken to standardize the protocol for in vitro plant regeneration via shoot tips for further breeding programmes (in vitro mutagenesis). The efforts were made to improve the shoot proliferation efficacy through MS medium enhanced with different combinations of cytokinins (BAP and TDZ) and auxins (NAA) to achieve higher number of multiple shoots with shorter period. Among the different combinations of BAP, TDZ and NAA, T5 (MS + TDZ 0.1 mgL-1 + NAA 0.2 mgL-1) was recorded early response (6.60 days), percentage of explant response to initiation (93.33%), days taken for multiple shoot induction (46.40 days) and the highest number of shoots per explant (8.93). The days taken for rooting, number of roots per shoot and highest root length was noted in the treatment half strength MS + 1.0 mgL-1 IBA + 1.0 g L-1 activated charcoal.
